Salmonella SPP /Shigella spp. gehören zu den häufigsten Bakterien für die Übertragung von akuter Gastroenteritis7,8,9,10,11. Daher haben viele Landkreise veröffentlicht, Gesetze oder Verordnungen für regelmäßige Salmonella SPP /Shigella spp. screening unter Menschen, die Nahrung und Wasser umgehen würden. Zum Beispiel haben die chinesische Regierung erließ Gesetze für obligatorische Salmonella SPP /Shigella spp. screening jährlich.
Der Gold-Standard-Methode für Salmonella SPP /Shigella spp. Erkennung ist Bakterienkultur. Durch Bakterienkultur und aufeinander folgenden biochemische Identifizierung und serologischen Charakterisierung identifizieren wir die Spezies von Bakterien, die das Krankheitsmanagement Ausbruch und antimikrobielle profiling zur Unterstützung der Behandlung von Patienten erleichtert 12. es könnte auch helfen, die Quelle der Infektion während der Salmonella SPP verfolgen /Shigella spp. Ausbruch13. Darüber hinaus lebensfähig aber nicht kultivierbarer (VBNC) Salmonella SPP /Shigella spp.möglicherweise gibt es in einigen Stuhlproben14. Blanco Et Al. zeigten, dass molekulare Tests sehr empfindlich, nicht nur für tragfähige Salmonellen in Kulturen, sondern auch mit teilweise Genomen und tot oder nicht lebensfähig Bakterien aus vergangenen Infektionen oder Kontamination26ist. Daher sollten neuer Technologien entwickelt werden.
Wir hier eine neuartige Methode, verbindet die NAAT Methode und Kultivierung basiert. Wie in Abbildung 1dargestellt, diese neue Methode gilt Real-Time PCR erste screening und positive Proben werden für die Bakterienkultur und Identifikation gesendet.

Diese Methode bietet eine Plattform mit hohem Durchsatz für das Screening von Salmonellen und Shigella. Der Hauptvorteil dieser Technik ist Wiederholung, spezifisch, empfindlich und hoher Durchsatz. Diese Technik kombiniert NAAT schnelle Methode und Kultur, in der Echtzeit-PCR-Screening wurde zuerst angewendet, und dann positive wurde für Bakterienkultur und Identifizierung gesendet.Obwohl sich diese Methode auf Salmonellen- und Shigella-Screening konzentriert, kann sie auch auf andere Krankheitserreger wie Vibrios, Staphylococcus, E.Coli und et cetera angewendet werden. Die Ergebnisse zeigten, dass die Kolonien aus Probe zwei Salmonella paratyphi B waren. Mit Echtzeit-PCR gab es eine erfolgreiche Verstärkung im FAM-Kanal, was darauf hindeutet, dass die Probe für Shigella-Arten positiv war. Weitere Echtzeit-PCR zeigten, dass Probe 10 für Shigella positiv war und für die geführte Kultur ausgewählt wurde.In der geführten Kultur der Shigella-Arten wurden die rosaroten und farblosen Kolonien getrennt plattiert und biochemisch identifiziert. Die Ergebnisse zeigten, dass Probe 10 Shigella sonnei Phase-II-Bakterien enthielt. Beim Versuch dieses Verfahrens ist es wichtig, sich die Einschränkung der Methode aufgrund von Sequenzvariationen zu merken.Nach diesem Verfahren können andere Methoden, wie die direkte Kultur, ausgeführt werden, um falsche negative Echtzeit-PCR-Ergebnisse zu vermeiden. Diese Technik ebnet den Weg für Forscher auf dem Gebiet der Salmonellen- und Shigella-Erkennung, da das neue Protokoll die positive Rate um das Zweifache erhöhen und die Arbeitsbelastung und TAT deutlich verringern könnte. Vergessen Sie nicht, dass die Arbeit mit Salmonellen und Shigella extrem gefährlich sein kann, und die persönliche Schutzausrüstung, PSA, sollte immer während der Durchführung dieses Verfahrens genommen werden.
