Ce protocole fournit une méthode efficace et reproductible pour la synthèse de nanoparticules de polymère lipidique plCSA-BP-conjugués. Une méthode thérapeutique efficace cancer réduit et élimine les tumeurs avec une toxicité systémique minimale. Par conséquent, ciblage tumoral sélective est la clé à l’exploration de méthodes thérapeutiques efficaces. NANOPARTICULES offrent une opportunité prometteuse pour le traitement du cancer et des assemblages moléculaires avec différents groupes fonctionnels améliorera l’efficacité du médicament et réduire des effets secondaires associés1,2. En outre, des systèmes de nanoparticules utilisent principalement passives et actives visant à atteindre la cible des tumeurs3.
Ciblage passive exploite les caractéristiques innées des nanoparticules et perméabilité améliorée et des effets de rétention (EPR) pour atteindre les cellules tumorales. Les liposomes cationiques ont été utilisés avec succès pour libérer des médicaments anticancéreux diverses tumeurs dans des applications cliniques4,5,6. Malgré l’effet thérapeutique potentiel de cancer efficace, une concentration faible de drogue dans la région de la tumeur et une incapacité à distinguer les cellules tumorales des tissus normaux sont les deux principales limites de nanoparticules de ciblage passif7.
Les stratégies de ciblage actifs Profitez d’antigène-anticorps, ligand-récepteur et autres interactions de reconnaissance moléculaire à libérer spécifiquement des médicaments tumeurs8. Le sulfate de chondroïtine placentaire glycosaminoglycan un (plCSA) est largement exprimé sur la plupart des cellules cancéreuses et des trophoblastes placentaires. Récemment, nous avons découvert un peptide de la liaison plCSA (plCSA-BP), dérivé de la protéine malarique VAR2CSA. plCSA-BP-conjugué lipide-polymère nanoparticules rapidement collé sur choriocarcinome cellules et une augmentation significative de doxorubicine (DOX) activité anticancéreuse in vivo11; ces particules aussi spécifiquement lié à trophoblastes placentaires et pourraient servir d’outil pour l’administration ciblée des médicaments au placenta12.
NANOPARTICULES lipidiques-polymère sont constitués d’une enveloppe de monocouche de lipides et un noyau de polymère hydrophobe et représentent un nouveau transporteur pour la livraison de drogue. Ces nanoparticules combinent les avantages des liposomes et nanocarriers polymères, tels que les nanoparticules contrôlables taille haute biocompatibilité, libération du médicament soutenue, drogue haute chargement (LE) et l’efficacité excellente stabilité13. Dans ce travail, nous avons utilisé une méthode seule étape sonication à synthétiser des nanoparticules lipidiques-polymère. Cette méthode est rapide, pratique et adapté pour l’intensification et a été largement utilisée pour préparer des nanoparticules lipidiques-polymère par notre groupe de11,14 et d’autres15,16,17,18 .
1-Ethyl-3-(3-dimethylaminopropyl) chlorhydrate de carbodiimide (EDC) est un populaire carbodiimide, utilisé comme un agent de réticulation pour la conjugaison des biomolécules contenant des amines et carboxylates19. En plus de l’EDC, N-hydroxysulfosuccinimide (NHS) est le plus commun réactif de conjugaison en surface et nanoparticules conjugaison réactions20,21. NHS peut réduire le nombre de réactions secondaires et améliorer la stabilité et le rendement d’ester INTERMEDIAIRES22,23.
Nous décrivons ici un protocole pour la synthèse de nanoparticules lipidiques-polymère plCSA ciblées. Tout d’abord, la synthèse de la seule étape sonication de nanoparticules lipidiques-polymère DOX-chargé (DNPs) est décrite. Ensuite, une technique de bioconjugate EDC/NHS pour générer des nanoparticules de polymère lipidique plCSA-BP-conjugué est introduite. Cette technique de bioconjugate permet aussi de conjuguer les autres anticorps et les peptides aux nanoparticules. Enfin, nous décrivons le dosage physico-chimiques, propriétés et in vitro , utilisé pour caractériser des nanoparticules lipidiques-polymère plCSA ciblées. Nous croyons que ces nanoparticules lipidiques-polymère plCSA ciblées pourraient constituer un système efficace pour l’administration ciblée des médicaments à des cancers plus humains et de l’administration ciblée de charges utiles pour le placenta pour traiter les troubles placentaires.

Une méthode thérapeutique efficace cancer réduit et élimine les tumeurs avec une toxicité systémique minimale. NANOPARTICULES activement ciblage offrent une approche prometteuse pour le traitement du cancer. Le sulfate de chondroïtine placentaire glycosaminoglycan un (plCSA) s’exprime sur une large gamme de cellules cancéreuses trophoblastes placentaires et protéine malarique VAR2CSA peut se lier spécifiquement à plCSA. Un peptide de liaison déclarés placentaire chondroïtine sulfate A (plCSA-BP), dérivé de protéine malarique VAR2CSA, pouvez également spécifiquement lier à plCSA sur les cellules cancéreuses et les trophoblastes placentaires. C’est pourquoi, plCSA-BP-conjugués de nanoparticules pourraient servir un outil pour la livraison de médicaments ciblés aux cancers humains et trophoblastes placentaires. Dans le présent protocole, on décrit une méthode pour faire la synthèse des nanoparticules de plCSA-BP-conjugué lipide-polymère chargés avec la doxorubicine (plCSA-DNPs) ; la méthode consiste à une sonication seule étape et bioconjugate techniques. En outre, plusieurs méthodes pour caractériser plCSA-DNPs, y compris la détermination de leurs propriétés physico-chimiques et la capture cellulaire de cellules de choriocarcinome placentaire (JEG3), sont décrites.

Nous présentons ici un protocole pour la synthèse de peptide de liaison placentaire chondroïtine sulfate A (plCSA-BP)-conjugués de lipide-polymère nanoparticules via seule étape sonication et bioconjugate techniques. Ces particules constituent un nouvel outil pour l’administration ciblée d’agents thérapeutiques tumeurs plus humaines et les trophoblastes placentaires pour traiter des cancers et des troubles placentaires.

Cette méthode peut aider à répondre aux questions clés dans le domaine de l’administration de médicaments, telles que la livraison ciblée de thérapeutiques aux cancers humains et le cystoblaste placentaire. Le principal avantage de cette technique est qu’il s’agit d’une méthode simple et reproductible pour synthétiser les peptides conjugués dans d’autres particules. Pour commencer, ajouter trois millimètres d’éthanol à quatre pour cent à un tube stérile centrifugeuse de 10 millilitres.Ajoutez ensuite 90 microgrammes de solution de stock de lécithine de soja, 210 microgrammes de solution de stock d’acide d’acide DSPE-PEG-carboxylic et 750 microgrammes de solution dox stock. Utilisez une seringue d’un millilitre pour pipette deux milligrammes de solution de stock PLGA. Placer le tube de centrifugeuse dans un bain de glace sur un processeur ultrasonique et sonicate pendant deux minutes.Ensuite, ajoutez la solution de stock PLGA drop-wise dans le tube de centrifugeuse. Pour purifier les PND, utilisez un filtre centrifugeuse pour laver la solution dans un tampon MES molaire de 0,1 molaire. Ensuite, centrifugez la solution à 1000 G pendant trois minutes à quatre degrés Celsius.Pour commencer l’activation de l’ester, ajoutez 0,4 milligramme d’EDC à un millilitre de PND. Ensuite, ajoutez 0,24 milligramme de NHS à la réaction. Ensuite, laissez la réaction se produire jusqu’à une heure à température ambiante dans l’obscurité.Après cela, mélanger les composants de réaction à fond et placer la réaction sur un shaker. Pour commencer la réaction à l’amine, utilisez PBS pour augmenter le pH tampon de 7,2 à 7,5. Ensuite, ajoutez 0,5 milligramme de peptide de ciblage pICSA à la solution de réaction.Bien mélanger la solution et la placer sur un shaker. Ensuite, laissez la réaction se poursuivre à quatre degrés Celsius pendant la nuit dans l’obscurité. Le lendemain, remplissez les sacs de dialyse avec la solution conjuguée.Purifiez les PNSP pICSA avec tampon PBS à température ambiante pendant 24 heures dans l’obscurité. Après avoir culture des cellules selon le protocole de texte, retirez les médias et ajoutez un millilitre de médias frais et froids avec cinq pour cent de PBS et pICSA DNPs ou DNPs. Ensuite, incuber le mélange de cellules et de nanoparticules pendant une heure à quatre degrés Celsius.Après cela, retirez le média et lavez les cellules trois fois avec PBS. Ensuite, ajouter un millilitre de médias frais et incuber les cellules pendant 30 minutes à 37 degrés Celsius. Après cela, retirez le média et lavez les cellules trois fois avec PBS.Ensuite, ajouter deux millilitres de froid, quatre pour cent de PFA et incuber le mélange à température ambiante pendant 10 minutes. Retirez le PFA et lavez les cellules avec deux millilitres de PBS. Ensuite, ajoutez un millilitre de PBS avec DAPI pour la coloration des noyaux.Laisser incuber le mélange à température ambiante pendant 10 minutes. Enfin, ajoutez le milieu de montage et l’image de la fluorescence avec un microscope à fluorescence. En utilisant ce protocole, les PND ont été préparés et conjugués avec succès à pICSA BP. Après conjugaison, le diamètre des PND primaires est passé à environ 109 nanomètres.Les images TEM des PNN et des PNN PICSA ont montré que les particules étaient bien dispersées et ont généralement démontré une morphologie sphérique. Le potentiel zeta des nanoparticules était négatif de 20,1 et 29,9 millivolts, ce qui indique que les nanoparticules étaient très stables. L’analyse d’absorption cellulaire jeg3 a indiqué que les PCISA se sont attachés aux cellules JEG3 dans les 30 minutes.Par conséquent, le BP pICSA a été efficacement conjugué à la surface. Tout en essayant cette procédure, il est important de se rappeler de sonifier le mélange pendant une à deux minutes avant d’ajouter PLGA. Suite à cette procédure, nos méthodes comme l’analyse HPLC peuvent être effectuées afin de déterminer l’efficacité de conjugaison avec plus de précision.Après son développement, cette technique a ouvert la voie à des chercheurs dans le domaine de la biologie de la reproduction pour explorer la possibilité d’un traitement ciblé par le placenta dans les complications de la grossesse.
