Eine wirksame Krebs therapeutische Methode reduziert und beseitigt Tumoren mit minimaler systemischer Toxizität. Daher ist die selektive Tumor targeting der Schlüssel zum erfolgreiche therapeutischen Methoden zu erforschen. Nanopartikel bieten eine vielversprechende Möglichkeit für die Krebstherapie und molekulare Baugruppen mit unterschiedlichen Funktionsgruppen Wirksamkeit von Medikamenten zu verbessern und reduzieren damit verbundenen Nebenwirkungen1,2. Darüber hinaus nutzen Nanopartikel Systeme vor allem passive und aktive targeting um Ziel Tumoren3zu erreichen.
Passive targeting nutzt die angeborenen Eigenschaften von Nanopartikeln und verbesserte Durchlässigkeit und Aufbewahrung (EPR) Effekte, Tumorzellen zu erreichen. Kationische Liposomen sind erfolgreich benutzt worden, Tumore in klinische Anwendungen4,5,6verschiedene Krebsmedikamente anzubieten. Trotz der potenziellen wirksam Krebs therapeutische Wirkung eine geringe Wirkstoffkonzentration in der tumorregion und der Unfähigkeit, Tumorzellen von normalen Zellen zu unterscheiden sind zwei wesentliche Einschränkungen des passiv-targeting Nanopartikel7.
Aktive targeting-Strategien nutzen Sie Antigen-Antikörper, Ligand-Rezeptor und andere molekulare Erkennung Interaktionen gezielt Medikamente an Tumoren8liefern. Plazentar Chondroitinsulfat Glykosaminoglykan (PlCSA) drückt sich im großen und ganzen auf die meisten Krebszellen und Plazenta Trophoblasten. Darüber hinaus können die Malaria Protein VAR2CSA speziell an PlCSA9,10binden. Daher kann VAR2CSA ein Werkzeug für das targeting von menschlichen Krebszellen sein. Wenn VAR2CSA mit Nanopartikeln konjugiert wird, kann das Full-Length Protein jedoch das Eindringen von Nanopartikeln in Tumorzellen einschränken. Kürzlich entdeckten wir eine PlCSA Bindung Peptid (PlCSA-BP), abgeleitet von der Malaria Protein VAR2CSA. PlCSA-BP-konjugiert Lipid-Polymer Nanopartikel rasch auf Choriocarcinoma Zellen und deutlich erhöhte Doxorubicin (DOX) Anti-Krebs-Aktivität in Vivo11gebunden; Diese Partikel auch speziell auf Plazenta Trophoblasten gebunden und als ein Instrument für die gezielte Abgabe von Medikamenten auf die Plazenta12dienen könnte.
Lipid-Polymer Nanopartikel bestehen aus einer Lipid-Monolayer-Shell und einer hydrophoben Polymeren Kern und repräsentieren einen neuen Träger für Drug-Delivery. Diese Nanopartikel verbinden die Vorteile von Liposomen und Polymeren darin, z. B. steuerbare nanopartikelgröße hohe Biokompatibilität, nachhaltiger Wirkstofffreisetzung, hohe Medikament laden Effizienz (LE) und ausgezeichnete Stabilität13. In dieser Arbeit haben wir eine einstufigen Beschallung Methode Lipid-Polymer Nanopartikel synthetisieren. Diese Methode ist schnell, bequem und Scale-up geeignet und ist am meisten benutzt Lipid-Polymer Nanopartikel Vorbereiten von unserer Gruppe11,14 u. a.15,16,17,18 .
1-Ethyl-3-(3-dimethylaminopropyl) Carbodiimide Hydrochlorid (EDC) ist eine beliebte Carbodiimide als ein Vernetzungsmittel für Biomoleküle mit Aminen und Carboxylates19Konjugation verwendet. Neben EDC ist N-Hydroxysulfosuccinimide (NHS) das am weitesten verbreitete Konjugation Reagenz in Oberfläche und Nanopartikel Konjugation Reaktionen20,21. NHS kann reduzieren die Anzahl der Nebenreaktionen und verbessern die Stabilität und Ausbeute an Ester Zwischenprodukte22,23.
Hier beschreiben wir ein Protokoll für die Synthese von PlCSA gezielt Lipid-Polymer Nanopartikel. Erstens wird die einstufigen Beschallung Synthese von DOX-geladene Lipid-Polymer-Nanopartikeln (DNPs) beschrieben. Dann wird eine EDC/NHS Bioconjugate Technik zur Erzeugung von PlCSA-BP-konjugiert Lipid-Polymer Nanopartikel eingeführt. Diese Bioconjugate Technik kann auch verwendet werden, andere Antikörper und Peptide mit Nanopartikeln zu konjugieren. Schließlich beschreiben wir die physikalisch-chemischen Eigenschaften und in-vitro- Assay verwendet, um die PlCSA ausgerichtete Lipid-Polymer Nanopartikel charakterisieren. Wir glauben, dass diese PlCSA gezielt Lipid-Polymer Nanopartikel ein effektives System für die gezielte Gabe von Medikamenten, um die meisten menschlichen Krebserkrankungen und die gezielte Bereitstellung von Nutzlasten der Plazenta zu plazentare Störungen zu behandeln sein könnte.

synthesis and characterization of placental chondroitin sulfate a (plcsa)-targeting lipid-polymer nanoparticles

Eine wirksame Krebs therapeutische Methode reduziert und beseitigt Tumoren mit minimaler systemischer Toxizität. Aktiv targeting Nanopartikel bieten einen vielversprechenden Ansatz in der Krebstherapie. Plazentar Chondroitinsulfat Glykosaminoglykan (PlCSA) drückt sich auf einer Vielzahl von Krebszellen und Plazenta Trophoblasten und Malaria Protein, das VAR2CSA speziell an PlCSA binden können. Eine gemeldete plazentar Chondroitinsulfat A Bindung Peptid (PlCSA-BP), abgeleitet von Malaria Protein VAR2CSA, kann auch speziell an PlCSA auf Krebszellen und Plazenta Trophoblasten binden. Infolgedessen konnte PlCSA-BP-konjugiert Nanopartikel als Instrument zur zielgerichteten Wirkstoffabgabe menschlichen Krebserkrankungen und Plazenta Trophoblasten eingesetzt werden. In diesem Protokoll beschreiben wir eine Methode zu synthetisieren, PlCSA-BP-konjugiert Lipid-Polymer Nanopartikel geladen mit Doxorubicin (PlCSA-DNPs); die Methode besteht aus einer einzigen Beschallung Schritt und Bioconjugate Techniken. Darüber hinaus werden verschiedene Methoden zur Charakterisierung von PlCSA-DNPs, einschließlich der Bestimmung ihrer physikalisch-chemischen Eigenschaften und zelluläre Aufnahme durch plazentare Choriocarcinoma (JEG3) Zellen beschrieben.

Diese Methode kann helfen, wichtige Fragen im Bereich der Medikamentenabgabe zu beantworten, wie die gezielte Abgabe von Therapeutika auf menschliche Krebsarten und die Plazenta Cystoblast. Der Hauptvorteil dieser Technik ist, dass es eine einfache und reproduzierbare Methode ist, Peptide zu synthetisieren, die in anderen Partikeln konjugiert sind. Zunächst drei Millimeter ethanolvon vier Prozent zu einem sterilen 10-Milliliter-Zentrifugenrohr hinzufügen.Fügen Sie dann 90 Mikrogramm Soja-Lecithin-Lagerlösung, 210 Mikrogramm DSPE-PEG-Carbonsäure-Lagerlösung und 750 Mikrogramm DOX-Lagerlösung hinzu. Verwenden Sie eine Ein-Milliliter-Spritze, um zwei Milligramm PLGA-Lagerlösung zu pipette. Das Zentrifugenrohr in ein Eisbad auf einen Ultraschallprozessor legen und zwei Minuten beschallen.Fügen Sie dann die PLGA-Lagerlösung tropfenweise in das Zentrifugenrohr ein. Dann fügen Sie 0,24 Milligramm NHS zur Reaktion hinzu. Dann lassen Sie die Reaktion für bis zu einer Stunde bei Raumtemperatur im Dunkeln auftreten.Danach die Reaktionskomponenten gründlich mischen und die Reaktion auf einen Shaker legen. Um die Aminreaktion zu starten, verwenden Sie PBS, um den Puffer-pH-Wert von 7,2 auf 7,5 zu erhöhen. Dann fügen Sie 0,5 Milligramm pICSA Targeting Peptid auf die Reaktionslösung.Mischen Sie die Lösung gut und legen Sie sie auf einen Shaker. Dann lassen Sie die Reaktion bei vier Grad Celsius über Nacht im Dunkeln ablaufen. Füllen Sie am nächsten Tag Dialysebeutel mit der Konjugatlösung.Reinigen Sie die pICSA DNPs mit PBS-Puffer bei Raumtemperatur für 24 Stunden im Dunkeln. Nach der Kultivierung von Zellen gemäß dem Textprotokoll entfernen Sie die Medien und fügen Sie einen Milliliter frischer, kalter Medien mit fünf Prozent FBS und pICSA DNPs oder DNPs hinzu. Dann inkubieren Sie die Zelle und Nano-Partikel-Mischung für eine Stunde bei vier Grad Celsius.Danach entfernen Sie die Medien und waschen Sie die Zellen dreimal mit PBS. Dann fügen Sie einen Milliliter frische Medien hinzu und brüten die Zellen für 30 Minuten bei 37 Grad Celsius. Danach entfernen Sie die Medien und waschen Sie die Zellen dreimal mit PBS.Dann fügen Sie zwei Milliliter Kälte, vier Prozent PFA und inkubieren sie die Mischung bei Raumtemperatur für 10 Minuten. Entfernen Sie die PFA und waschen Sie die Zellen mit zwei Milliliter PBS. Fügen Sie dann einen Milliliter PBS mit DAPI für die Kernfärbung hinzu.Lassen Sie die Mischung bei Raumtemperatur für 10 Minuten brüten. Fügen Sie schließlich das Montagemedium hinzu und stellen Sie die Fluoreszenz mit einem Fluoreszenzmikroskop auf. Mit diesem Protokoll wurden DNPs erfolgreich vorbereitet und mit pICSA BP konjugiert. Nach der Konjugation erhöhte sich der Durchmesser der primären DNPs auf etwa 109 Nanometer.Die TEM-Bilder der DNPs und der pICSA DNPs zeigten, dass die Teilchen gut dispergiert waren und im Allgemeinen sphärische Morphologie zeigten. Das Zeta-Potenzial der Nanopartikel betrug negative 20,1 und 29,9 Millivolt, was darauf hindeutet, dass die Nanopartikel sehr stabil waren. Der JEG3-Zellaufnahmetest zeigte die pICSA DNPs an, die innerhalb von 30 Minuten an die JEG3-Zellen gebunden sind.Daher wurde der pICSA BP effizient an die Oberfläche konjugiert. Beim Versuch dieses Verfahrens ist es wichtig, die Mischung ein bis zwei Minuten lang zu beschallen, bevor Sie PLGA hinzufügen. Nach diesem Verfahren können unsere Methoden wie HPLC-Assay durchgeführt werden, um die Konjugationseffizienz genauer zu bestimmen.Nach ihrer Entwicklung ebnete diese Technik den Weg für Forscher auf dem Gebiet der Reproduktionsbiologie, um die Möglichkeit einer Plazenta-Targeting-Behandlung bei Schwangerschaftskomplikationen zu erforschen.
