Denne protokollen gir en effektiv og reproduserbar metode for å syntetisere plCSA-BP-konjugerte lipid-polymer nanopartikler. Metoden enkeltsteg sonication å forberede lipid-polymer nanopartikler er rask, reproduserbare og forskjellig fra typiske nanoprecipitation metoder som innebærer varme, vortexing eller fordampning. En effektiv kreft terapeutisk metode reduserer og eliminerer svulster med minimal systemisk toksisitet. Derfor er selektiv svulst målretting nøkkelen til å utforske vellykket terapeutiske metoder. Nanopartikler tilbyr en lovende mulighet for kreft terapi, og molekylær samlinger med ulike funksjonsgruppene vil forbedre narkotika effekt og redusere tilhørende bivirkninger1,2. Videre benytter hydrogenion systemer hovedsakelig passive og aktive målretting for å nå målet svulster3.
Passiv målretting utnytter medfødte egenskaper av nanopartikler og økt permeabilitet og oppbevaring (EPR) effekter å nå kreftceller. Kationisk liposomer har blitt brukt til å levere ulike anticancer narkotika til svulster i klinisk bruk4,5,6. Til tross for den potensielle effektive kreft terapeutiske effekten, en lav narkotika konsentrasjon i regionen svulst og en manglende evne til å skille kreftceller fra normalt vev er to store begrensninger av passiv målretting nanopartikler7.
Aktive målretting strategier utnytte antigen-antistoff, ligand-reseptor og andre molekylære anerkjennelse interaksjoner spesielt levere narkotika til svulster8. Glycosaminoglycan placental chondroitin sulfate en (plCSA) er generelt uttrykt på de fleste kreftcellene og placental trophoblasts. Videre malarial protein VAR2CSA kan spesielt binde plCSA9,10. Derfor kan VAR2CSA være et verktøy for målretting menneskelige celler. Men når VAR2CSA er konjugert til nanopartikler, kan full lengde protein begrense gjennomtrengning av nanopartikler i kreftceller. Nylig oppdaget vi en plCSA binding peptid (plCSA-BP), avledet fra malarial protein VAR2CSA. plCSA-BP-konjugerte lipid-polymer nanopartikler raskt limt til choriocarcinoma celler og betydelig økt doksorubicin (DOX) anticancer aktivitet i vivo11; disse partiklene også limt til placental trophoblasts og kan tjene som et verktøy for målrettet levering av narkotika morkaken12.
Lipid-polymer nanopartikler består av en lipid monolayer skall og en hydrofobe polymere kjerne og representerer en ny operatør for narkotika-leveranser. Disse nanopartikler kombinere fordelene med liposomer og polymere nanocarriers, som kontrollerbar hydrogenion størrelse, høy biocompatibility, vedvarende narkotika utgivelsen, høy narkotika lasting effektivitet (LE) og utmerket stabilitet13. I dette arbeidet brukte vi en enkelt-trinns sonication metode for å syntetisere lipid-polymer nanopartikler. Denne metoden er rask, praktisk og egnet for oppskalering og har vært mye brukt til å forberede lipid-polymer nanopartikler av vår gruppe11,14 15,16,17,18 .
1-Ethyl-3-(3-dimethylaminopropyl) carbodiimide hydroklorid (EDC) er en populær carbodiimide brukes som crosslinking agent for bøye biomolecules som inneholder aminer og carboxylates19. EDC er N-hydroxysulfosuccinimide (NHS) den vanligste Bøyning reagensen i overflaten og hydrogenion Bøyning reaksjoner20,21. NHS kan redusere antall siden reaksjoner og forbedre stabilitet og ytelse på ester mellomprodukter22,23.
Her beskriver vi en protokoll for å syntetisere plCSA målrettede lipid-polymer nanopartikler. Først er enkeltsteg sonication syntese av DOX lastet lipid-polymer nanopartikler (DNPs) beskrevet. Deretter er en EDC/NHS bioconjugate teknikk for å generere plCSA-BP-konjugerte lipid-polymer nanopartikler innført. Denne bioconjugate teknikken kan også brukes til å bøy andre antistoffer og peptider å nanopartikler. Til slutt, vi beskrive mekanisk-egenskaper og i vitro analysen benyttes for å karakterisere plCSA målrettede lipid-polymer nanopartikler. Vi tror at disse plCSA målrettede lipid-polymer nanopartikler kan utgjøre et effektivt system for levering av målrettede narkotika mest menneskelige kreft og målrettet levering av payloads i morkaken å behandle placental lidelser.

synthesis and characterization of placental chondroitin sulfate a (plcsa)-targeting lipid-polymer nanoparticles

En effektiv kreft terapeutisk metode reduserer og eliminerer svulster med minimal systemisk toksisitet. Aktivt målretting nanopartikler tilbyr en lovende tilnærming til kreft terapi. Glycosaminoglycan placental chondroitin sulfate en (plCSA) er uttrykt i et bredt spekter av kreftceller og placental trophoblasts og malarial protein VAR2CSA kan spesifikt bindes til plCSA. Et rapportert placental chondroitin sulfate A bindende peptid (plCSA-BP), avledet fra malarial protein VAR2CSA, kan også binde å plCSA kreftceller og placental trophoblasts. PlCSA-BP-konjugerte nanopartikler kan derfor brukes som et verktøy for målrettet stoffet levering til kreft hos mennesker og placental trophoblasts. I denne protokollen beskriver vi en metode for å syntetisere plCSA-BP-konjugerte lipid-polymer nanopartikler lastet med doksorubicin (plCSA-DNPs); metoden består av en enkelt sonication trinn og bioconjugate teknikker. I tillegg beskrives flere metoder for å karakterisere plCSA-DNPs, herunder fastsettelse av deres mekanisk-egenskaper og mobilnettet opptak av placental choriocarcinoma (JEG3) celler.

Denne metoden kan bidra til å svare på viktige spørsmål i narkotikaleveringsfeltet, for eksempel målrettet levering av terapeutiske midler til menneskelig kreft og morkaken cystoblast. Den største fordelen med denne teknikken er at det er en enkel og reproduserbar metode for å syntetisere peptider konjugert i andre partikler. Til å begynne, tilsett tre millimeter av fire prosent etanol til et sterilt 10 milliliter sentrifugerør.Deretter legger du til 90 mikrogram soyabønnelecitin lagerløsning, 210 mikrogram DSPE-PEG-karboksylsyrelagerløsning og 750 mikrogram DOX-lagerløsning. Bruk en en milliliter sprøyte til å pipette to milligram PLGA lageroppløsning. Plasser sentrifugerøret i et isbad på en ultralydprosessor og soniker i to minutter.Deretter legger PLGA lagerløsning drop-wise inn i sentrifugerøret. For å rense DNPs, bruk et sentrifugefilter til å vaske løsningen i 0,1 molar MES-buffer. Deretter sentrifuger oppløsningen ved 1000 G i tre minutter ved fire grader Celsius.For å begynne ester aktivering, legge til 0,4 milligram EDC til en milliliter dnps. Deretter legger du til 0,24 milligram NHS til reaksjonen. Deretter lar reaksjonen oppstå i opptil en time ved romtemperatur i mørket.Etter dette blander du reaksjonskomponentene grundig og plasserer reaksjonen på en shaker. Neste dag fyller du dialyseposer med konjugatløsningen.Rens pICSA DNPs med PBS buffer ved romtemperatur i 24 timer i mørket. Etter å ha kultivert celler i henhold til tekstprotokollen, fjern mediet og legg til en milliliter frisk, kald media med fem prosent FBS og pICSA DNPs eller DNPs. Deretter inkubere cellen og nanopartikkelblandingen i en time ved fire grader Celsius.Etter dette fjerner du mediet og vasker cellene tre ganger med PBS. Deretter legger du til en milliliter frisk media og inkubere cellene i 30 minutter ved 37 grader Celsius. Etter dette fjerner du mediet og vasker cellene tre ganger med PBS.Deretter legger du til to milliliter kaldt, fire prosent PFA og inkuberer blandingen ved romtemperatur i 10 minutter. Fjern PFA og vask cellene med to milliliter PBS. Deretter legger du til en milliliter PBS med DAPI for kjernefarging.La blandingen inkubere ved romtemperatur i 10 minutter. Til slutt legger du til monteringsmedium og bilde fluorescensen med et fluorescensmikroskop. Ved hjelp av denne protokollen ble DNPs utarbeidet og konjugert til pICSA BP. Etter bøyning økte diameteren til de primære DNPs til ca. 109 nanometer.TEM-bildene av DNPs og pICSA DNPs viste at partiklene var godt dispergert og generelt demonstrert sfærisk morfologi. Zetapotensialet til nanopartiklene var negativt 20,1 og 29,9 millivolt, noe som indikerer at nanopartiklene var svært stabile. JEG3-mobilopptaksanalysen indikerte pICSA DNPs bundet til JEG3-cellene innen 30 minutter.Derfor ble pICSA BP effektivt konjugert til overflaten. Mens du prøver denne prosedyren, er det viktig å huske å sonikere blandingen i ett til to minutter før du legger PLGA. Etter denne prosedyren kan våre metoder som HPLC-analyse utføres for å bestemme bøyningseffektiviteten mer nøyaktig.Etter utviklingen banet denne teknikken vei for forskere innen reproduktiv biologi for å utforske muligheten for placenta-målretting behandling i graviditet komplikasjoner.
