Complicaciones del embarazo mediada por la placenta, incluyendo la preeclampsia, la pérdida del embarazo, desprendimiento prematuro de placenta y pequeño edad gestacional (SGA), son comunes y conducen a importante morbilidad fetal y materna y mortalidad1,2, 3y muy pocos fármacos han demostrado para ser eficaces para tratar el embarazo trastornos de4,5. El desarrollo de estrategias para la administración de fármacos dirigidos a placenta más selectivo y seguro durante el embarazo sigue siendo un desafío en la farmacoterapia moderna.
En los últimos años, varios informes se han centrado en la entrega dirigida de fármacos a los tejidos uteroplacentaria por capa nanopartículas con anticuerpos o péptidos como instrumentos dirigidos a placenta. Estos incluyen un anticuerpo de anti-factor de crecimiento receptor (EGFR)6 , tumor-homing péptidos (CGKRK y iRGD)7, péptidos dirigidos a placenta8, péptidos orientada en la vasculatura placentaria9 y anticuerpos contra el receptor de oxitocina10.
Aquí, demostramos que se puede utilizar un péptido sintético placentaria condroitín sulfato A enlace (plCSA-BP) para la entrega específica de nanopartículas y sus cargas de droga a la placenta11. Las nanopartículas plCSA BP guiada son complementarias a la uteroplacentaria informó a métodos debido a el trofoblasto placentario de destino.
Como un método no invasivo, en vivo la proyección de imagen se ha utilizado para monitorizar la expresión génica específica de la placenta en ratones12y verde del indocyanine (ICG) ha sido ampliamente utilizado para seguimiento de nanopartículas usando la fluorescencia sistemas13, la proyección de imagen 14,15. Así, nos inyecta por vía intravenosa plCSA BP conjugado nanopartículas cargadas con ICG (plCSA-INPs) para visualizar la distribución plCSA INP en ratones embarazadas con un reproductor de imágenes de fluorescencia. Entonces inyectamos intravenoso methotrexate (MTX)-carga plCSA NPs en ratones embarazadas. Ultrasonido de alta frecuencia (HFUS), otro no-invasiva, en tiempo real imaging herramienta16,17 fue utilizada para supervisar el desarrollo fetal y placentario en los ratones. Por último, se utilizó cromatografía líquida de alto rendimiento (HPLC) para cuantificar la distribución de MTX en las placentas y fetos.
En este protocolo, describimos en detalle el sistema tres-método para evaluar la eficacia de la administración de fármacos dirigidos a placenta nanocarriers plCSA BP guiada.

No hay tratamientos eficaces existen para complicaciones del embarazo asociada a placenta, y desarrollo de estrategias para la entrega específica de drogas a la placenta y reducir al mínimo efectos secundarios fetales y maternos sigue siendo desafiante. Portadores de nanopartículas dirigidas ofrecen nuevas oportunidades para tratar los trastornos placentarios. Recientemente hemos demostrado que un péptido sintético placentaria condroitín sulfato A enlace (plCSA-BP) podría utilizarse para guiar las nanopartículas para administrar medicamentos a la placenta. En este protocolo, describimos en detalle un sistema para evaluar la eficacia del fármaco a la placenta por plCSA-BP que emplea tres métodos separados en combinación: en vivo la proyección de imagen, ultrasonido de alta frecuencia (HFUS) y alto rendimiento cromatografía líquida (HPLC). En vivo imagen, guiada por la BP plCSA nanopartículas fueron visualizadas en las placentas de animales vivos, mientras que HFUS y HPLC demostraron que las nanopartículas conjugadas de BP plCSA eficientemente y específicamente entregan metotrexato a la placenta. Por lo tanto, una combinación de estos métodos puede utilizarse como una herramienta eficaz para la entrega específica de drogas a la placenta y el desarrollo de nuevas estrategias de tratamiento para varias complicaciones del embarazo.
