胚の発達の間に、単一の細胞層は3つの別々の生殖層1、2、3に変換される。生体内で発生する現象の研究可能性を高めるために、便利なモデルとして三次元集合体(胚体)の生成が開発されました。このようにして形成された細胞凝集体は、細胞分化を引き起こす様々な条件にさらされ、胚4、5の発達を反映する。P19マウス胚癌細胞株(P19細胞株)は、ビトロ6、7、8における神経新生研究の細胞モデルとして一般的に使用される。P19細胞株は、典型的な多能性幹細胞の特徴を示し、細胞凝集中のレチノイン酸(RA)の存在下でニューロンに分化し、付着条件下で神経突起の成長を続けることができます。また、未分化P19細胞株は、ジメチルスルホキシド(DMSO)9、10、11、12の影響下で筋肉および心筋細胞様細胞を形成することもできる。
多くの方法13、14、15、16は、神経分化のために報告されているが、方法論は時々複雑であり、説明を読むだけでは理解しにくい。例えば、プロトコルは、ふくらはぎ血清(CS)と胎児ウシ血清(FBS)13の混合物を補充したダルベッコの修飾イーグル培地(DMEM)培地の組み合わせを必要とする場合がある。さらに、神経発達に使用される培地は、しばしばニューロベースサルとB27サプリメント13、14、15、16で構成されている。そのため、既存のメソッドは準備の複雑さが含まれており、ここでの目標はプロトコルを簡素化することです。本研究では、FbSを用いてDMEMをP19細胞株(DMEM+10%FBS)の維持、ニューロン発達(DMEM+5%FBS+RA)に利用できることを実証した。P19細胞株を用いて神経新生を簡素化したこの簡略化された方法は、ニューロンがどのように発達するかの分子機構を研究することを可能にする。また、アルツハイマー病等の神経変性疾患に関する研究もP19細胞株17,18を用いて行われ、本研究で開発した方法が解明に一役を果たそして、神経発達異常および神経変性疾患における分子機構。

マウス胚由来の奇形癌に由来するP19細胞株は、3つの生殖層に分化する能力を有する。レチノイン酸(RA)の存在下で、懸濁培養P19細胞株はニューロンに分化するように誘導される。この現象は、インビトロで神経新生モデルとして広範囲に研究されている。したがって、P19細胞株は、神経新生に関連する分子および細胞研究に非常に有用である。しかしながら、文献に記載されているP19細胞株の神経分化のためのプロトコルは非常に複雑である。本研究で開発した方法は簡潔で、神経発達異常や神経変性疾患の分子機構解明に一役を果たします。

P19細胞株はニューロンに分化することが知られている。しかし、P19細胞株のニューロン分化のためのプロトコルは、時には複雑である。この方法により、神経原性実験をユーザーフレンドリーな方法で行い、将来のP19細胞株の使用可能性を拡大することができます。まず、P19細胞を維持培地に維持し、DMEMで構成され、グルコース1リットル当たり4.5グラム、牛胎児血清10%、ペニシリン1ミリリットルペニシリン当たり100単位、ミリリットルストレプトマイシンあたり100単位を補う。細胞が約80%コンフルエントになるまで、37°Cおよび5%の二酸化炭素環境で細胞培養をインキュベートする。細胞培養フラスコから使用済み培地を取り出し、カルシウムとマグネシウムフリーPBSの2ミリリットルで細胞を洗います。0.25%トリプシン-EDTAの1ミリリットルを加えて、細胞の単層を完全に覆い、摂氏37度、炭酸ガス5%で2~3分間インキュベートします。顕微鏡下で、すべての細胞が剥離しているかどうかを確認します。トリプシンを中和するために、9ミリリットルの維持培地で細胞を再懸濁する。細胞を15ミリリットルのチューブに移し、遠心分離機をGの200倍、室温を5分間移して細胞をペレットします。その後、ペレットを乱さずに上清を吸引する。新鮮なメンテナンス培地の10ミリリットルにペレットを再中断し、メーカーの指示に従ってセル数を決定するためにセルカウンターを使用します。新しいT-25フラスコで1平方センチメートル当たり20,000細胞をシードし、最大10ミリリットルの維持培地を添加します。摂氏37度、二酸化炭素5%を2~3日間インキュベートします。トリプシン消化を開始するには、まず上清をゆっくりと吸引し、カルシウムとマグネシウムフリーPBSの2ミリリットルで細胞を洗います。その後、0.25%トリプシンEDTAの1ミリリットルを細胞に加え、摂氏37度で、5%の二酸化炭素を2〜3分間インキュベートします。インキュベーション後、トリプシンを中和し、9ミリリットルの分化培地で細胞を再懸濁し、高グルコースレベルのDMEMで構成され、5%のウシ胎児血清、ミリリットルペニシリン当たり100単位、およびミリリットルレンサマイシンあたり100単位を補った。細胞を15ミリリットルのチューブに移し、遠心分離機をGの200倍、室温を5分間移して細胞をペレットします。次いで、上清をゆっくりと吸引し、RAを含まない新鮮な分化培地の1ミリリットルでペレットを再懸濁する。セルカウンタを使用して、メーカーの指示に従ってセル番号を確認します。凝集生成のために、10ミリリットルの分化培地を加え、100ミリリットルの非処理培養皿に5マイクロリットルのRAを添加する。培養皿に100万個の細胞を播種し、凝集体形成を促進するために摂氏37度と5%の二酸化炭素で2日間インキュベートする。インキュベーション後、10ミリリットルのピペットを用いて、凝集物を含む培地を吸引し、室温で15ミリリットルのチューブに移す。集計が底部に落ち着くのを1.5分待ってから、上清を捨てます。0.5マイクロモルRAを補充した新鮮な分化培地の10ミリリットルを追加します。新しい100ミリメートルの非処理培養皿に凝集物を穏やかに播種し、摂氏37度と2日間の5%の二酸化炭素でインキュベートします。まず、10ミリリットルのピペットを使用して、細胞凝集体を15ミリリットルチューブに移します。凝集体が底部に落ち着くまで室温で1.5分待ってから、上清を捨てます。血清および抗生物質を含まないDMEMで凝集物を洗浄します。細胞の凝集体が底部に落ち着くのを待ち、上清を捨て、0.25%トリプシンEDTAの2ミリリットルを加えます。37°Cの水浴でチューブを10分間インキュベートし、2分ごとにタップして細胞を懸濁させておきます。次に、1ミリリットルのピペットを使用してトリプシン活性とピペットを20回上下させるために4ミリリットルのメンテナンス培地を加えます。最後に、細胞をGの200倍、室温で5分間遠心する。上清を取り除き、維持培地の5ミリリットルで細胞ペレットを再懸濁します。セルカウンターを使用して細胞数を決定し、細胞のめっきを行います。細胞をプレートするには、まず6ウェル培養プレートに維持培地のウェルあたり3ミリリットルを加える。その後、ウェルあたり500,000の密度で細胞を播種します。プレートを摂氏37度と炭酸ガス5%でインキュベートします。6ウェル培養プレートにカバーグラスの細胞を播種します。20%合流に達したら、抗MAP2抗体による免疫染色に進みます。次いで、RNAを単離し、MAP2、ヌエン、OCT4、ナノグ、およびGAPDHに対する逆転写PCRを行う。本研究では、神経分化の簡略化された方法を紹介する。P19細胞株は、5%FBSおよび0.5マイクロモルRAを有する非処理培養皿で培養される。4日後、細胞凝集体はトリプシンと解離し、次の4日間接着細胞培養プレートに播種される。この方法の効率は、未分化状態と神経新生中のP19細胞株のRTPCR分析との比較によって評価される。結果は、Oct4やNANOGなどの多能性遺伝子の発現の急速な減少、およびMAP2およびNeuNなどのニュールノマルマーカー遺伝子のアップレギュレーションを示す。今回の研究で開発した方法は単純で、神経発生の分子メカニズムや神経変性疾患の解明に一役立つと考えています。
