世界的には、非特異的な臨床プレゼンテーションによる確認のための外科的介入と診断検査のパフォーマンスが悪いため、肺外結核の診断が遅れることが多い。結核菌抗原の血清診断試験を改善するため、肺外結核を検出するための超常磁性酸化鉄ナノプローブを開発しました。TB-SPIOナノプローブは、自己免疫応答を誘導することなく、常磁特性のために低濃度で正確な磁気共鳴画像を提供することができます。TB-SPIOナノプローブは、結核菌感染の標的化および検出を可能にする。それらは他の疾患の検出のためのMRI造影剤としても適用することができる。実験の各部分は特定のコア技術を利用するため、プロトコルを試す前にプロトコルを慎重に検討してください。この方法の視覚的なデモンストレーションは、特により複雑な実験のいくつかについて、生徒がプロセスを実装する方法を理解するのに役立ちます。デキストランコーティングされた酸化鉄磁性ナノ粒子を調製するために、デキストランT40の5ミリリットルを45ミリグラムの塩化第一六水和物水溶液と32ミリグラムの塩化四水和物を室温で精力的に組み合わせます。水酸化アンモニウムを10ミリリットル速く加え、攪拌棒を使用して、得られた黒い懸濁液を室温で1時間かき混ぜる。攪拌インキュベーションの終わりに、溶液を遠心分離し、凝集体を取り除く。最終的なSPIO製品を非結合デキストランT40から分離するには、5ミリリットル反応混合物全体を2.5 x 33センチメートルのゲルろ過クロマトグラフィーカラムにロードし、酢酸ナトリウム0.1モルと塩化ナトリウム緩衝液の0.15モルでデキストランを溶出します。次に、精製されたデキストラン被覆酸化鉄ナノ粒子を空隙体積に集め、標準プロトコルに従って330および490ナノメートルで鉄およびデキストラン用にカラムが溶出するアッセイを行う。SPIO共役結核菌表面抗体を作成するために、まずSPIO-EBDEのアルカリ溶液10ミリリットルをコハク酸無水物1グラムと室温で24時間混合してSPIO-EDBEコハク酸コハク酸無水物を合成する。翌日、12,000~4,000分子量カットオフ分子多孔質膜チューブを使用して、1回の変化につき2リットルの蒸留水を26時間変化して溶液を透析する。最後の変更後、得られた溶液の100マイクロリットルを1ミリリットルTB表面抗体あたり4.5ミリグラムの400マイクロリットルに加え、続いて1-ヒドロキシベンゾトリアゾールおよびベンゾトリアゾール-1-イロキシトリコリジノホスホニウムを攪拌しながら室温で触媒として添加する。溶液を、PBSをバッファーとして使用してゲルろ過クロマトグラフィーを介して非結合抗体から分離します。平均粒径、形態、およびサイズ分布を決定するには、複合分散体を200メッシュの銅格子に投下し、室温でサンプルを空気乾燥してから、100キロボルトでイメージングする透過電子顕微鏡にローディングします。ナノプローブの緩和時間値を測定するには、20メガヘルツと37°Cのプラスマイナス1度の核磁気共鳴リラクソメーターを使用します。ナノプローブパルス細胞イメージングでは、まず標準細胞培養プロトコルに従ってヒトTHP-1単球を培養します。細胞が活性化の適切な段階に達した場合、1時間のマイコバクテリウムボビスBCGの6番目のコロニー形成単位に10回で7つの単球に1回10回プレインキュベートする。インキュベーションの終わりに、細胞を1ミリリットルマイクロ遠心分離チューブに移し、SPIOマイコバクテリウム結核表面抗体共役ナノプローブを2ミリモルずつ各チューブに加え、摂氏37度と5%の二酸化炭素で1時間培養します。インキュベーションの終了時に、遠心分離によって細胞を収集し、新鮮な培地の200マイクロリットルでペレットを再懸濁します。次に、3.0-T磁気共鳴画像法を介して高速勾配エコーパルスシーケンスを使用してサンプルをスキャンし、ナノプローブの特異性と感度を決定します。実験動物にM.bovis BCGを接種するには、まず、乾燥したワクチンまたは細菌ストックをSauton培地中で再構成し、再構成したストック溶液を生理食塩水で希釈する。次に、1匹の動物につき1ミリリットルのシリンジに溶液の100マイクロリットルをロードし、各マウスの左または右の肩甲骨皮膚に皮内細菌の全容を注入する。生きたナノプローブ注入動物の生体内磁気共鳴画像処理では、各マウスの尾静脈に200マイクロリットルの生理食塩水に懸濁したSPIO-TB抗体プローブのナノモルを2ナノモルに注入する前に、各麻酔実験動物のベースラインT2加重高速スピンエコー画像を取得する。その後、注射直後に動物を再び画像化し、次の30分間5分ごとに画像を作成します。画像処理セッションの終了時に、M.結核肉芽腫センターと肉芽腫領域に隣接する背筋の同等の場所での定義された関心領域の測定として、すべての磁気共鳴画像を定量的に分析します。次に、この式を用いて、造影剤の注入前及び0〜3時間後の信号強度測定を用いて相対的な信号増強を計算する。接種の1ヶ月後、各動物の皮内接種部位から組織を採取し、ヘマトキシリンおよびエオシン、酸性菌用Ziehl-Neelsen染色、鉄の鉄のベルリンブルーを含む5〜10マイクロメートルの厚い組織サンプルを埋め込んだホルマリン固定パラフィンを染色する。SPIO-TB表面抗体ナノプローブの透過電子顕微鏡イメージングは、ナノプローブがコア当たり平均サイズ3.8プラスマイナス0.4ナノメートルの良好に分散した外観を示すことが明らかになっています。M.ボビスBCGおよび酸の速い細菌株はZiehl-Neelsen染色によって検出することができる。鉄を含むプローブで単離および培養した後、細菌はベルリンブルー染色を介して同定することができる。SPIO-TB表面抗体ナノプローブのTBターゲティング度合いは、濃度依存的に発生するナノプローブの存在下でのシグナル強度の低下によって示されるように、T2重み付け磁気共鳴画像法を介して決定することができる。ナノプローブ注入動物では、M.結核肉芽腫領域におけるシグナル低減のT2加重増強は、コントロール部位の約14倍高い。感染の1ヶ月後、組織化された皮下肉芽腫は、新しい血液血管化およびリンパ球および上皮系マクロファージ凝集体を有するナノプローブ注入C57黒6マウスにおいて、これらの病変内に記載されている。正のM.結核発現は、病変部位において陽性の酸性菌染色を有する肉芽腫領域においても観察される。ベルリンブルーは、鉄鉄陽性染色体であり、TBに対するプローブの感度を決定するためにも使用できる。生体内磁気共鳴画像の場合、ナノプローブ注入動物はSPIO-TB抗体プローブを注入する前に、麻酔付き実験動物ごとにベースラインT2加重高速スピンエコー画像を取得します。肺外結核を検出するSOP技術を確立したので、これらの抗体は肺外結核を特異的に診断するために使用することができます。
