Kanker immunotherapie heeft in de afgelopen jaren naar voren geschoven. Genetisch gemodificeerde chimeer antigeen receptor (auto) T-cellen zijn een uitstekend voorbeeld van gemanipuleerde immuuncellen geïmplementeerd in kanker immunotherapie. Deze cellen zijn onlangs goedgekeurd door de FDA voor behandeling tegen CD19 + B cel maligniteiten, maar succes tot nu toe beperkt gebleven tot ziekten dragen van een paar targetable antigenen en richten van dergelijke beperkte antigene repertoires is gevoelig voor storing door immuun ontsnappen. AUTO T cellen hebben bovendien gericht geweest op het gebruik van autologe T cellen vanwege het risico van graft - versus - host-ziekte veroorzaakt door allogene T-cellen. In tegenstelling, NK-cellen zijn in staat om te doden van tumor doelen op een antigeen-onafhankelijke manier en niet leidt tot GVGH, waardoor ze een goede kandidaat voor kanker immunotherapie6,7,8,9.
CRISPR/Cas9 technologie heeft onlangs zijn gebruikt in engineering immune cellen is, maar genetisch herprogrammering NK-cellen met plasmiden altijd uitdagend. Dit is vanwege problemen in de transgenic levering in een DNA-afhankelijke manier zoals lentivirale en retrovirale transductie veroorzaakt aanzienlijke procedure-geassocieerde NK cellen apoptosis en de beperkte productie van genetisch gemanipuleerde NK cellen4 , 9.
Vele ingeboren immune cellen express hoge niveaus van receptoren voor pathogen-geassocieerde moleculaire patronen zoals Retinoic acid-afleidbare gene ik (RIG-ik), waardoor verhoogde erkenning van buitenlandse DNA. Onderdrukking van deze trajecten heeft hogere efficiëntie van de signaaltransductie in NK-cellen, bij het gebruik van DNA gebaseerde methoden voor genetische modificatie10.
Hier beschrijven we de methode voor het gebruik van een DNA-vrije genoom bewerken van primaire en uitgebreide menselijke NK-cellen met behulp van Cas9 ribonucleoprotein complexen (Cas9/RNPs). Cas9/RNPs bestaat uit drie onderdelen, recombinant Cas9 eiwit complexvorm met synthetische single-gids RNA bestaat uit een complexvorm crRNA en tracerRNA. Deze Cas9/RNPs zijn geschikt voor het splijten van genomic doelen met hogere efficiëntie ten opzichte van buitenlandse DNA-afhankelijke benaderingen toe te schrijven aan hun levering als functionele complexen. Daarnaast, snelle goedkeuring van de Cas9/RNPs van de cellen kan het verminderen van de gevolgen van de af-target zoals inductie van apoptosis. Dus kunnen ze worden gebruikt voor het genereren van knock-outs of knock-ins wanneer gecombineerd met DNA voor homologe recombinatie6,7. We toonden dat electroporation van Cas9/RNPs is een makkelijk en relatief efficiënte methode die de eerdere beperkingen van genetische modificatie in NK-cellen overwint.
TGFβ is een grote immunosuppressieve cytokine, die de activering en de functies van NK-cellen remt. Er is gesuggereerd dat het gericht op de TGFβ-pathway immuun cel functies kan verhogen. We gericht de regio codering TGBR2 ectodomain die TGFβ11. bindt De representatieve resultaten tonen een significante afname in het niveau van mRNA uitdrukking van dit gen en verder aantonen dat het gewijzigde NK-cellen resistent voor TGFβ geworden. CRISPR/Cas9 technologie versnelt genoom engineering in vele celtypes, maar tot dusver, gene levering en stabiele genetische modificatie geweest uitdagende in primaire NK-cellen. Bijvoorbeeld resulteerde transgenic levering met behulp van lentivirale of retrovirale transductie in een beperkte opbrengst van genetisch gemanipuleerde NK cellen als gevolg van substantiële procedure-geassocieerde NK cellen apoptosis. Hier beschrijven we een DNA-gratis methode voor het bewerken van het genoom van menselijke primaire en uitgebreide NK cellen met behulp van Cas9 ribonucleoprotein complexen (Cas9/RNPs). Deze methode mag efficiënte knock-out van de TGFBR2 en HPRT1 genen in NK-cellen. RT-PCR gegevens toonden een significante afname in gen expressie niveau, en een bepaling van de cytotoxiciteit van een representatieve cel product gesuggereerd dat de RNP gemodificeerde NK-cellen werd minder gevoelig voor TGFβ. Genetisch gemodificeerde cellen zou uitgebreide post-electroporation door stimulatie bestraalde mbIL21-tot uitdrukking te brengen feeder cellen.
