DNA-avhengige endring av NK celler har vært utfordrende4,9. Vi derfor introdusert direkte en syntetisk preformed ribonucleoprotein (RNPs) kompleks og Cas9 protein som renset protein i primære og utvidet NK celler8. Denne metoden tillatt oss å eliminere capping, tailing, og andre transcriptional og translasjonsforskning prosesser startet av RNA polymerase II, som kan forårsake NK celle apoptose forbundet med DNA-avhengige signaltransduk...

Kreft immunterapi har vært avanserte i de siste årene. Genetisk endret chimeric antigen reseptor (bil) T-celler er et utmerket eksempel på utvikling immunceller distribuert i kreft immunterapi. Disse cellene var godkjent av FDA for behandling mot CD19 + B celle malignitet, men suksess hittil vært begrenset til sykdommer bærer noen targetable antigener og målretting slik begrenset antigen repertoar er utsatt for svikt immun flukten. Videre har bil T celler vært fokusert på bruk av autologous T-celler på grunn av risikoen for graft - versus - host sykdom forårsaket av allogene T-celler. I kontrast, NK celler kan drepe svulst mål i en antigen-uavhengig måte og forårsaker ikke GvHD, noe som gjør dem en god kandidat for kreft immunterapi6,7,8,9.
CRISPR/Cas9 teknologien har vært brukt nylig i engineering immunceller, men genetisk omprogrammering NK celler med plasmider har alltid vært utfordrende. Dette er på grunn av problemer i transgene levering på en DNA avhengige måte som lentiviral og retroviral signaltransduksjon forårsaker betydelig prosedyre-assosiert NK celle apoptose og begrenset produksjon av genmodifiserte NK celler4 , 9.
Mange medfødte immunceller express høye nivåer av reseptorer for patogen-forbundet molekylær mønster som Retinoic acid-induserbart genet jeg (RIGG-jeg), som aktiverer økt anerkjennelse av utenlandske DNA. Undertrykkelse av disse veiene har aktivert høyere signaltransduksjon effektivitet i NK celler ved DNA-baserte metoder for genetisk modifisering10.
Her beskriver vi metoden for å bruke en DNA-fri genomet redigering av primære og utvidet menneskelige NK celler utnytte Cas9 ribonucleoprotein komplekser (Cas9/RNPs). Cas9/RNPs består av tre komponenter, rekombinant Cas9 protein kompleksbundet med syntetisk single-guide RNA består av en kompleksbundet crRNA og tracerRNA. Disse Cas9/RNPs er stand til å spalte genomisk mål med høyere effektivitet sammenlignet med utenlandske DNA-avhengige tilnærminger på grunn av deres leveringsmidler som funksjonell komplekser. I tillegg kan rask klarering av Cas9/RNPs fra cellene redusere effekten av mål som induksjon av apoptose. Dermed kan de brukes til å generere fortrenging eller knock-ins kombinert med DNA for homologe rekombinasjon6,7. Vi viste at electroporation av Cas9/RNPs er en enkel og relativt effektiv metode som overvinner foregående begrensninger av genmodifisering i NK celler.
TGFβ er en stor suppressive cytokin, som hemmer aktivisering og funksjoner av NK celler. Det har blitt foreslått at målretting TGFβ veien kan øke immunforsvaret celle funksjoner. Vi målrettet regionen koding TGBR2 ectodomain som binder TGFβ11. Representant resultatene viser en betydelig reduksjon i nivået av mRNA uttrykk for dette genet og ytterligere demonstrere at endrede NK cellene blir motstandsdyktig mot TGFβ. I tillegg beholde de endrede cellene levedyktighet og proliferativ potensial, som de kunne være utvidet innlegg-electroporation benytter bestrålt mater celler. Derfor følgende metode er en lovende tilnærming til genetisk manipulere Celler NK for ytterligere kliniske eller forskning.

CRISPR/Cas9 teknologi er akselererende genomet engineering i mange celletyper, men så langt, gen levering og stabil genet modifikasjon har vært utfordrende i primære NK celler. For eksempel resulterte ved lentiviral eller retroviral signaltransduksjon transgene levering i en begrenset avkastning av genmodifiserte NK celler på grunn av betydelig prosedyre-assosiert NK celle apoptose. Her beskriver vi en DNA-fri metode for genomet redigering av menneskelig primære og utvidet NK celler ved hjelp av Cas9 ribonucleoprotein komplekser (Cas9/RNPs). Denne metoden tillatt effektiv knockout av TGFBR2 og HPRT1 gener i NK celler. RT PCR data viste en signifikant nedgang i gene expression nivå, og en cytotoksisitet analysen representant celle produktet foreslo at RNP endret NK cellene ble mindre følsomme til TGFβ.
