La importancia de este método radica en su reproducibilidad y la eficacia del tejido cardiaco resultante de varias capas. El objetivo de la actual ingeniería de tejido cardiaco es desarrollar una terapia para reemplazar o reparar la estructura y función del tejido miocárdico lesionado1. Métodos para crear modelos 3D tejido cardiaco exhibe las propiedades importantes de contráctiles y electrofisiológicas del tejido cardiaco nativa han estado ampliando rápidamente2,3. Una variedad de estrategias han sido exploradas y utilizadas en los estudios4,5. Estos métodos entre el uso de específicos hidrogeles bioactivos naturales y sintéticos, tales como gelatina, colágeno, fibrina y péptidos6, y deposición del bio-tinta tecnologías tecnologías2 y bioprinting7.
Se ha demostrado que métodos libres de andamio pueden producir tejidos comparables como métodos basados en el biomaterial, sin los inconvenientes de la incorporación de material de andamio exterior8. Oren Caspi et al demostraron que la incorporación de varios tipos de células permite la generación de tejido cardiaco ingeniería humana altamente vascularizado9. Chin et al. desarrolló un método de impresión 3-d para la creación del parche cardiaco de esferoides. Parches que se componen de cardiomiocitos, fibroblastos y células endoteliales en un cociente de 70:15:1510. Esferoides han demostrado ser eficaces "building blocks" de la creación de tejido cardíaco libre de andamios, ya que son resistentes contra la hipoxia y poseen suficiente integridad mecánica para la implantación de11,12. Estudios anteriores han demostrado varios métodos de fabricación para la creación de esferoide, incluyendo el uso de las que cuelgan de la gota método, spinner frascos13, sistemas de microfluidos14y cultura no adherente superficies sin revestir o revestidos con micro-moldes de agarosa15. En este protocolo, se utiliza el colgante gota dispositivo, que contiene microporos para cientos de esferoides de condensación a la vez.
Este estudio presenta un novedoso y eficaz método de andamio-libre para la creación de tejido cardíaco, que incluye siembra manualmente los esferoides en la cavidad de un molde cuadrado y el tejido en un agitador de incubación para la maduración. Bajo condiciones de cultivo estático habitual, difusión de oxígeno está limitada a los aspectos exteriores de la construcción de tejido, resultando en necrosis central. Sin embargo, con el molde de red, todos los esferoides sembradas en el molde están inmersos en los medios de comunicación con un constante movimiento fluídico, permitiendo para la mayor difusión de nutrientes y oxígeno. Además, este método basado en el molde permite la creación simultánea de parches de tejido de diferentes tamaños con el mínimo esfuerzo manual y el tejido resultante se puede quitar fácilmente del molde. Este novedoso método permite la creación eficiente y reproducible de parches cardíacos libre de andamio, de varias capas.

Este protocolo describe un novedoso y fácil método neto basado en el molde para crear tejidos cardiacos de tridimensional (3D) sin material de andamio adicional. Cardiomiocitos de derivados de células madre humanas pluripotentes (iPSC-CMs), fibroblastos cardiacos humanos (HCF) y las células endoteliales de vena umbilical humano (HUVECs) son aisladas y utilizadas para generar una suspensión con 70% iPSC-CMs, HCF de 15% y 15% HUVECs. Son cultivadas conjuntamente en un sistema de fijación ultra baja "gota colgante", que contiene microporos para cientos de esferoides de condensación a la vez. Las células agregan y espontáneamente forman esferoides paliza después de 3 días de cocultivo. Los esferoides son cosechadas, sembrados en una cavidad de molde novela y cultivadas en un agitador en la incubadora. Los esferoides se convierten en un tejido funcional maduro aproximadamente 7 días después de la siembra. En nuestros experimentos, se utilizó una suspensión de iPSC-CMs 70%, 15% HCF y HUVECs 15% en medios de comunicación de células RPMI/B-27 a una concentración de 2.475.000 células por mL.
