De experimentele procedures die hierboven beschreven presenteer een methode om te overwinnen de intrinsiek lage activiteit van de single-omzet protease PCSK9 en zijn proteolytische functie wordt geëvalueerd in een robuuste manier. PCSK9 richt de LDL-receptor (LDL-R) voor afbraak, verhoging van LDL cholesterol (LDL-C) en rijden atherosclerotische hart-en vaatziekten1,2. Therapeutiek targeting PCSK9 krachtig lagere LDL-C en verbeteren van cardiovasculaire uitkomsten voor patiënten, zelfs wanneer toegevoegd aan een agressieve lipide-verlagende therapie met statines3,4. Momenteel goedgekeurde therapieën zijn beperkt tot antilichaam-gebaseerde benaderingen, echter, en lijden onder een gebrek aan kosteneffectiviteit5,6. U kunt dit probleem oplossen, zijn minder dure therapeutische alternatieven, een middel om het identificeren van patiënten kunnen profiteren van meer voordelen, of beide, nodig.
Kleine-molecuul benaderingen kunnen richten intracellulaire PCSK9, bieden een betere wijze van toediening en kostenverlaging, waardoor ze de "Heilige Graal" in dit gebied7. PCSK9 heeft echter bewezen moeilijk te drug door kleine moleculen. Tot op heden echter deze strategie niet geslaagd, waarschijnlijk als gevolg van de PCSK9 van unieke biochemie: PCSK9 cleaves alleen zelf10, uitvoeren van een reactie van single-omzet, en na zelf-decolleté, de PCSK9 prodomain blijft gebonden in de actieve site als een auto-remmer11, voorkomen van de uitlezing van elke verdere protease-activiteit.
Dit artikel presenteert een methode om te evalueren van de Proteolytische functie PCSK9 in high-throughput mode8. Via plaats-geleide mutagenese, kunnen onderzoekers deze test gebruiken om de effecten van codering SNPs gevonden in de kliniek om te beoordelen voor effecten op proteolyse, de snelheidslimieten stap van PCSK9 rijping sonde. Bovendien, zal deze methode nuttig zijn in het ontwerp van hoge-doorvoer schermen te identificeren modulatoren van PCSK9 proteolyse, die worden verwacht om uiteindelijk het verstoren van de presentatie van PCSK9 de LDL-r (en moduleren van PCSK9 van hypercholesterolemic effect) . Proprotein convertase subtilisin/kexin type 9 (PCSK9) is een single-omzet protease die serum low-density lipoprotein (LDL) niveaus regelt en, bijgevolg, hart-en vaatziekten.
