Die oben beschriebenen Versuchsverfahren präsentieren eine Methode zum überwinden der intrinsisch niedrigen Aktivität der einzelnen Umsatz Protease PCSK9 und seine proteolytische Funktion in einer robusten Weise zu bewerten. Der Schlüsselbegriff des Assays stützt sich auf ein Single-Umsatz-Ereignis in einer enzymatisch verstärkte Auslesen umzuwandeln. Die Stärken des Assays gehören die relativ kurzen Zeitrahmen und Benutzerfreundlichkeit der Luciferase Reporter, als auch die Skalierbarkeit für Hochdurchsatz-Ans?...

PCSK9 richtet den LDL-Rezeptor (LDL-R) für den Abbau, Erhöhung von LDL-Cholesterin (LDL-C) und fahren atherosklerotische Herzkrankheit1,2. Therapeutika targeting PCSK9 robust niedrigere LDL-C und Verbesserung der Herz-Kreislauf-Ergebnisse für Patienten, auch wenn eine aggressive lipidsenkenden Therapie mit Statinen3,4hinzugefügt. Derzeit zugelassene Therapien beschränken sich auf Antikörper-basierte Ansätze, und leiden unter mangelnder Wirtschaftlichkeit5,6. Um dieses Problem zu lösen, sind weniger kostspielig therapeutischen Alternativen, ein Mittel zur Identifizierung von Patienten wahrscheinlich größere Vorteile zu erlangen oder beides erforderlich.
Klein-Molekül Ansätze könnten gezielt intrazellulären PCSK9, bieten eine verbesserte Art der Verabreichung und Kosten reduzieren, so dass sie den "Heiligen Gral" in diesem Bereich7. Allerdings ist durch kleine Moleküle PCSK9 schwierig, Drogen nachgewiesen. Als eine Protease ist Ausrichtung PCSK9s proteolytischen Funktion eine attraktive Strategie, wie selbst-Proteolyse der Rate-Begrenzung von PCSK9 Reifung8 ist und für seine maximale Wirkung auf LDL-R9erforderlich ist. Bislang jedoch diese Strategie hat nicht erfolgreich gewesen, wahrscheinlich aufgrund PCSK9s einzigartige Biochemie: PCSK9 spaltet nur selbst10, Durchführung einer Einzel-Umsatz-Reaktion und nach selbst-Spaltung, die PCSK9 prodomain bleibt im aktiven Zentrum als eine Auto-Inhibitor11, verhindert das Auslesen der jede weitere Protease-Aktivität.
Dieser Artikel stellt eine Methode zur Evaluierung PCSK9 proteolytischen Funktion im Hochdurchsatz-Mode8. Durch Site-verwiesene Mutagenese können Ermittler dieser Assay untersuchen die Auswirkungen der Codierung SNPs gefunden in der Klinik, um sie für Auswirkungen auf Proteolyse, die Bandbreitenbegrenzung Schritt des PCSK9 Reifung zu bewerten. Darüber hinaus werden diese Methode nützlich bei der Gestaltung von Hochdurchsatz-Bildschirmen Modulatoren des PCSK9 Proteolyse zu identifizieren, die voraussichtlich letzten Endes stören die Präsentation des PCSK9 auf die LDL-R (und modulieren PCSK9s normalem Effekt) . Zu guter Letzt kann dieses Protokoll andere Proteasen mit intrinsisch geringer Aktivität angepasst werden, vorausgesetzt, dass (i) ein paar spezifische Substrat-Protease gefunden werden kann, und (Ii) ein geeigneter intrazellulärer Anker für das Substrat hergestellt werden kann.

Obwohl PCSK9 Proteolyse für seine volle normalem Wirkung erforderlich ist, die Bewertung ihrer proteolytischen Funktion ist anspruchsvoll: PCSK9 ist nur bekannt, sich Spalten, erfährt nur einen einzigen Umsatz und nach Proteolyse, behält seine Substrat in seiner aktiven Seite als ein Auto-Inhibitor. Die hier vorgestellten Methoden beschreiben eine Probe, die diese Herausforderungen überwindet. Der Test konzentriert sich auf intermolekularen Proteolyse in einem Zell-basierte Kontext und Links erfolgreiche Spaltung die sekretierten Luciferase-Aktivität, die leicht im konditionierten Medium gelesen werden können. Über aufeinander folgenden Schritten Mutagenese, Transiente Transfektion und eine Luciferase auslesen, kann der Test PCSK9 Proteolyse unter Bedingungen von genetischen oder Molekulare Störung in gewissem Sinne Hochdurchsatz-Sonde. Dieses System eignet sich gut für beide die biochemische Auswertung der klinisch entdeckte Missense Einzel-Nukleotid-Polymorphismen (SNPs), als auch für das Screening von kleinen Molekül-Inhibitoren des PCSK9 Proteolyse.

Hochdurchsatz-Proteolyse Assay stützt sich auf drei große Herausforderungen zu überwinden. Erstens ist zur Überwindung der intrinsisch geringen Leistung eines Single-Umsatz PCSK9 Protease, ein PCSK9 Protease fehlt die hemmenden prodomain mit der Spaltung Sequenz am Ende der prodomain verwendet, verbunden mit einer Luciferase, die sekretierten14sein können.
