关于对该议定书可能的修改, 其他嵌入树脂和金属污渍可以代替 LR 白色丙烯酸和 PTA 分别使用。Embed812 是一种常用于电子显微镜的环氧树脂, 具有高粘性、更难转移的特性, 可能引起试样的畸变, 并与丙烯酸或乙二醇甲基丙烯酸酯树脂相比具有较高的背景。而 JB4 加, 乙二醇甲基丙烯酸酯, 比 EMbed812 粘性低, 具有较低的背景, 随机形成的气囊经常出现在附近的标本。如前所述, 空气袋降低了图像质量, 对于贵重样品尤其有问题。值得注意的是, Technovit 7100, 另一个乙二醇甲基丙烯酸酯, 干扰 PTA 染色, 导致在最终图像的对比度较差。相对地, LR 白色丙烯酸具有水样粘度, 低背景, 不干扰 PTA 染色。在我们的手中, 成功率嵌入在 LR 白色没有样品损坏或气泡是大于95%。因此, 我们提倡将其用作组织显微 CT 的嵌入树脂。在污渍方面, 在微 CT 成像中对各种金属污渍如锇四氧化二、碘和 PTA 的结果进行了比较, 并在别处讨论了15、16、17 , 超出了这一范围。手稿。
样本大小是当前协议的一个主要限制。由于我们使用吸力将试样转移到油管中, 因此观察试样的能力对定位至关重要, 并确保它确实在油管中。因此, 像缓步动物(i. e, 水熊) 这样的亚毫米样品很难嵌入, 因为它们需要使用显微镜进行可视化。一旦吸入被应用, 显微标本很容易从焦平面丢失, 并成为挑战重新发现, 使它很难确定, 如果他们通过连接末端的油管。更大的样本, 如小鼠胚胎, (3-10 毫米) 可以嵌入对嵌入协议的细微修改 (图 4D)。特别地, 聚酰亚胺管被填充到1/3 与液态树脂, 这是在嵌入之前聚合。固定和染色样品放置在预聚合树脂的顶部。然后, 油管完全充满了未聚合树脂, 其次是第二聚合。

表型是一个有机体的可观察的特征, 代表其遗传背景和环境之间独特相互作用的后果。这些特征可以包括行为、生化、形态学、发育和/或生理特性。重要的是, 野生型生物体和遗传突变体之间的性状差异可以为受影响基因的机制和功能提供重要的见解。在形态学方面, 组织病理学是评估细胞级表型的金标准, 但受机械工件的影响, 不允许准确的定量体积分析1。我们的实验室的动机是克服在亚微米分辨率下, 组织学对全组织体积的诊断能力的障碍。
对现有技术的一项调查表明, X 射线微计算机断层扫描 (显微 CT) 成像可以为毫米级全动物3维 (3D) 组织学提供理想的能力。微 CT 可实现非破坏性、各向同性、3D 可视化和对组织结构进行定量分析的能力1,2。近年来, 微 CT 对未染色和金属染色斑马鱼的3D 成像得到了越来越多的牵引, 并被用于多种组织的容积分析, 包括肌肉、牙齿、骨骼和脂肪组织3、4 ,5,6,7,8,9,10。其他模型生物体和组织标本也可用于显微 CT 成像。例如, 通过同步辐射显微 CT11, 引入了用于量化小鼠大脑致密中尺度神经解剖学的管道。同样, 伊红的染色协议已被证明适合于软组织显微 CT 成像的整个小鼠器官12。未染色人 L3 椎骨的形态学分析和用显微 CT 显示银染人肺的方法证明了该技术对人体样品13、14的实用性。
为了实现微 CT 对完整的小动物和组织标本的完全形态学分型的巨大承诺, 需要克服许多障碍, 如产量、分辨率、视野、细胞综合染色、和长期保存。虽然这些方面都是至关重要的, 但本手稿的重点是嵌入程序的优化, 还有关于样品固定和染色的补充说明。重金属染色是很有用的, 因为微 CT 图像中不同软组织的内在对比度较低。研究了四氧化二、碘、磷钨酸 (PTA) 和 gallocyanin chromalum 等各种金属污渍的功效, 以增强显微 CT 成像的对比度15、16、17。铀酰醋酸酯也被用作骨和软骨显微 CT 成像的对比剂18,19。PTA 在我们的协议中使用, 导致整个斑马鱼标本中几乎所有组织和细胞的一致染色, 产生的图像可能与组织学类似的研究, 通过全卷的组织。
在显微 CT 图像采集过程中, 2 维 (2D) 投影被视为样品旋转的程度的一小部分和重复, 直到样品完成180°或360°旋转, 产生一系列的2D 投影用于3D 卷20的重建。在此过程中, 试样的任何扰动都会导致相应的2D 投影脱离对齐, 从而导致3D 体积的重建效果不佳。固定样品是纠正此问题的一种方法, 目前的一些策略包括将样品浸入酒精中, 或在聚丙烯管中嵌入琼脂糖或微量吸管技巧3、5、15、16,21,22. 液体浸泡方法并不理想, 因为图像采集过程中的样本移动可能发生在成像集之间, 从而导致3D 体积23的移位重建。此外, 众所周知, 由于化学不稳定性和表面磨损与样品和容器壁之间接触点的移动相关, 液体储存的组织样品的物理稳定性在数月到数年的时间尺度上较差 (与固定动物和人体组织样本的个人观察)。
为了减少成像过程中样品移动的可能性, 可以将样品嵌入琼脂糖24、25、26, 但这种做法与染料扩散到琼脂糖的风险有关, 从而减少了图像对比度26。此外, 液体或琼脂糖制剂容易受到物理损伤, 并随着时间的推移而降解, 使其不适合长期样品贮存。我们推断, 一个潜在的成功和简单的样品固定化方法可以从用于电子显微标本的。如 EPON 812 (停产并被嵌入812取代) 等聚合树脂通常用于提供为电子显微术27、28生成超薄截面所需的硬度。事实上, x 射线成像和电子显微镜的一些比较研究表明, 在树脂中嵌入的样品可以通过 X 射线显微术29,30成像。然而, 电子显微镜的一些标准做法并不直接转化为显微 CT 成像。例如, 典型树脂块的方形树脂边缘样品的显微 CT 成像和这些方块切割的样品与可能干扰成像的边缘衍射伪影相关。平滑树脂边缘, 虽然可能, 是费力和耗时。
虽然在电子显微镜中使用了各种塑料嵌入树脂, 但与硬树脂 (如嵌入 812) 相关的高背景使我们能够测试其他材料。由于其粘度低、收缩率低、聚合过程中形成气泡的可能性低、背景较低等原因, 我们选择了 LR 白。为了创建无边缘样品, 并尽量减少样品周围树脂的数量, 我们开发了在聚合前将液态树脂中的试样绘制成聚酰亚胺管的协议。选择聚酰亚胺具有较高的热稳定性和高 x 射线透射率, 因此无需进行成像即可去除油管。最后, 我们为样品嵌入技术设计了一个定制的微量吸管适配器, 以可靠地握住油管, 防止微的污染。适配器可以从 CAD 映像文件打印3D。结合起来, 这里介绍的样品制备方法的目标是使小样本的嵌入更简单, 实现增强的染色对比度, 刚性固定化, 并长期储存完整的毫米尺度标本。
