Apoptose, et godt undersøgt form af programmeret celledød, er forpligtet til at opretholde fysiologiske homeostase og fjerne potentielt skadelige celler i den menneskelige krop1. Efter induktion af apoptose, kan apoptotiske celler (ApoCells) underkastes en række morfologiske ændringer og adskille i lille membran-bundet vesikler betegnes ApoBDs. Samlet set denne proces er kendt som apoptotiske celle demontering og kan opdeles i 3 særskilte foranstaltninger baseret på morfologi2,3. Trin 1 (plasma membran blebbing) er karakteriseret ved dannelsen af ballon-lignende strukturer på cellens overflade kaldes blebs4,5. Trin 2 (apoptotiske fremspring dannelse) omfatter dannelsen af lange membran fremspring såsom apoptopodia, beaded apoptopodia og mikrotubulus pigge6,7,8. Endelig, trin 3 (ApoBD dannelse) omfatter fragmentering af de apoptotiske fremspring og/eller ApoCells til at generere ApoBDs6,9. Tidligere resultater har foreslået en rolle for ApoBDs i medvirken apoptotiske celle clearance og mægle intercellulær kommunikation. Derudover kan ApoBDs havnen en serie af biomolekyler DNA, RNA og proteiner, som kan være smugles til omkringliggende celler til at lette celle-celle kommunikation12,13,14. Tidligere har været brugt en række metoder, herunder flowcytometri og elektronmikroskopi at studere apoptose og ApoBDs15,16,17,18. ApoBD påvisning og kvantificering er dog ofte vanskelig eller overset. For eksempel, mest rutinemæssigt anvendes flow flowcytometri-baserede apoptose assay beskæftiger annexin V (A5, et protein, der binder de externalized ' spise-mig ' signal Phosphatidylserin (PtdSer)) og nukleinsyre pletten propidium Iodid (PI)19. Men ved hjælp af denne universelle pletten kombination, analyse forudsætter, at der er kun tre typer af celle subsets (levedygtige celler, ApoCells og nekrotiske celler) i prøven. Derfor, vi har for nylig udviklet en roman flow flowcytometri assay bruger A5 og til-PRO-3, en anden nukleinsyre plet, der selektivt kan tages op af caspase 3/7-aktiveret pannexin 1 (PANX1) kanaler7,20. Vores tilgang understreger vigtigheden af at analysere alle celler/partikel delmængder og endnu vigtigere, adskillelsen af ApoBDs fra celler og debris20. Således, denne tilgang viser en effektiv teknik for at validere induktion af apoptose og ApoBD dannelse samtidigt.
ApoBDs har traditionelt været isoleret gennem en række differentierede centrifugering tilgange hvorved ApoBDs kan adskilles fra celler eller andre ekstracellulære vesikler baseret på tæthed. Men sådanne centrifugering metoder er ofte begrænset af lav ApoBD renhed, manglen på en kvantificering trin at bekræfte prøve renhed, og/eller manglende evne til at adskille celle typespecifikke ApoBDs17,21,22. Derfor, vi for nylig udviklet to tilgange, en FACS-baseret og en ny differentieret centrifugering-baseret tilgang, som kan kobles til vores tidligere etablerede flow flowcytometri metode til validering af induktion af apoptose og prøve renhed23. ApoBD isolation via vores FACS-baserede tilgang kan berige ApoBDs til op til 99% renhed, og kan være kombineret med en bred vifte af celle type-specifikke antistoffer til at isolere ApoBDs fra blandet cellepopulationer, vævsprøver og kropsvæsker23. Desuden vores reviderede differential centrifugering tilgang viser en effektiv metode til at isolere ApoBDs til > 90% renhed23.
I dette papir beskriver vi i detaljer vores forsøgsmetoden at validere apoptose induktion, og for at påvise og bestemme ApoBD dannelse. ApoBD isolation arbejdsprocesser ved hjælp af FACS-baseret og differentieret centrifugering-baserede metoder er også udarbejdet og sammenlignet. Apoptotiske organer (ApoBDs), microvesicles og exosomes er de vigtigste medlemmer af familien ekstracellulære vesikel, med ApoBDs at være en af de største type.
