Sinds de vroege beschrijving in de jaren 1950, het veld van apoptosis beschikt over geavanceerde aanzienlijk, steeds een prominente onderzoeksruimte. Ondanks de brede interesse en uitgebreide inspanningen, zijn bepaalde aspecten van apoptosis, in het bijzonder de vorming van ApoBDs, niet goed bestudeerd door het gebrek aan geschikte methoden. Deze omvatten met name de beperking in het bijhouden van apoptosis progressie en vorming van de ApoBD gelijktijdig met behulp van de traditionele stroom cytometry A5/PI analyse en d...

Een goed bestudeerde vorm van geprogrammeerde celdood, apoptosis, is vereist voor de fysiologische homeostase te handhaven en potentieel schadelijke cellen binnen het menselijk lichaam1verwijderen. Na de inductie van apoptosis, kunnen apoptotic cellen (ApoCells) een aantal morfologische veranderingen ondergaan en demonteren in kleine membraan-gebonden blaasjes genoemd ApoBDs. Globaal, is dit proces staat bekend als de apoptotic cellen demontage en kan worden onderverdeeld in 3 duidelijke stappen op basis van morfologie2,3. Stap 1 (plasmamembraan blebbing) wordt gekenmerkt door de vorming van ballon-achtige structuren aan het celoppervlak bekend als blebs4,5. Stap 2 (apoptotic uitsteeksel vorming) omvat de vorming van de lange membraan uitsteeksels zoals kralen-apoptopodia, apoptopodia en microtubulus spikes6,7,8. Stap 3 (ApoBD vorming) bevat ten slotte de versnippering van de apoptotic uitsteeksels en/of ApoCells voor het genereren van ApoBDs6,9. Eerdere bevindingen hebben voorgesteld een rol van ApoBDs in de medeplichtigheid van apoptotic cellen goedkeuring en bemiddelen van de intercellulaire communicatie. Bijvoorbeeld, wordt voorgesteld dat de fragmentatie van een ApoCell in de ApoBDs kleine 'hapklare' stukjes die gemakkelijk kunnen worden verwijderd door het omringende fagocyten2,10,11kan genereren. Bovendien kunnen ApoBDs een reeks van biomoleculen zoals DNA, RNA en eiwitten, die kunnen worden verhandeld naar omliggende cellen te vergemakkelijken van cel-cel communicatie12,13,14haven. Functioneel onderzoek doen naar deze processen, is het essentieel voor het bevestigen van drie essentiële parameters, met inbegrip van (i) validering van apoptose inductie en vorming van de ApoBD, (ii) isolatie van ApoBDs, en (iii) de bevestiging van ApoBD zuiverheid.
Eerder, een aantal methoden, met inbegrip van stroom cytometry en elektronenmicroscopie hebben gebruikt om apoptosis en ApoBDs15,16,17,18te studeren. ApoBD detectie en kwantificering zijn echter vaak moeilijk of over het hoofd gezien. Bijvoorbeeld, de meest routinematig gebruikte stroom cytometry gebaseerde apoptosis assay werken Annexine V (A5, een eiwit dat de externalized bindt ' eten-me' signaal fosfatidylserine (PtdSer)) en nucleïnezuur vlek propidium jodide (PI)19. Echter, met behulp van deze universele vlek combinatie, analyse is verondersteld dat er slechts drie soorten cel deelverzamelingen (levensvatbare cellen, ApoCells en necrotische cellen) in het monster zijn. Bovendien al beschouwd als "een goudstandaard" door vele onderzoekers voor apoptose, stroom cytometry testen en latere data-analyse vaak worden uitgesloten van ApoBDs door middel van een eerste gating stap FSC/SSCintermediair-hoge gebeurtenissen selecteren. Daarom hebben we onlangs een nieuwe stroom cytometry assay met A5 en aan-PRO-3 ontwikkeld, een ander stikstofbase vlek die kan selectief worden overgenomen door caspase-3/7-geactiveerde pannexin 1 (PANX1)7,,20kanalen. Zoals activering van caspase 3-geïnduceerde PANX1 voorafgaat PtdSer blootstelling aan het vroege stadium van apoptosis, vlekken aan-PRO-3 differentieel apoptotic en necrotische cellen. Bovendien, deze aanpak in combinatie met onze roman gating strategie omvat alle verworven evenementen tijdens gegevensanalyse en daardoor zes cel/deeltje deelverzamelingen zijn geïdentificeerd, met inbegrip van: (i) levensvatbare cellen (FSC/SSCintermediair/hoge, A5laag ,Lageaan-PRO-3), (ii) A5- vroege ApoCells (FSC/SSCintermediair/hoge,lageA5, aan-PRO-3intermediaire), (iii) A5+ ApoCells (FSC/SSCintermediair/hoge, A5hoog ,Tussenliggendeaan-PRO-3), (iv) necrotische cellen of late ApoCells (FSC/SSCintermediair/hogeA5hoog, aan-PRO-3hoog), (v) ApoBDs (FSC/SSClage, A5tussenliggende, aan-PRO-3laag / intermediate), en (vi) puin (FSC/SSClage,lageA5, aan-PRO-3lage)20. Onze aanpak benadrukt het belang van het analyseren van alle deelverzamelingen van de cellen/deeltje en, nog belangrijker, de scheiding van ApoBDs van cellen en puin20. Aldus, toont deze aanpak een efficiënte techniek voor het valideren van de inductie van apoptosis en de vorming van de ApoBD gelijktijdig.
Traditioneel zijn ApoBDs geïsoleerd door een verscheidenheid van differentiële centrifugeren benaderingen waarbij ApoBDs kan worden losgekoppeld van de cellen of andere extracellulaire blaasjes op basis van dichtheid. Dergelijke centrifugeren methoden zijn echter vaak beperkt door lage ApoBD zuiverheid, gebrek aan een kwantificering stap te bevestigen monster zuiverheid en/of onvermogen om te scheiden van de cel type-specifieke ApoBDs17,21,22. Dus ontwikkelde we recent twee benaderingen, een FACS gebaseerde en een nieuwe differentiële centrifugeren gebaseerde benadering, die kan worden gekoppeld aan onze eerder vastgestelde stroom cytometry methode voor het valideren van de inductie van apoptosis en monster zuiverheid23. ApoBD isolatie via onze FACS gebaseerde benadering kan verrijken ApoBDs tot 99% zuiverheid en kan gepaard gaan met een scala aan cel type-specifieke antilichamen tegen ApoBDs isoleren van gemengde cel populaties, weefselsteekproeven en lichaamsvloeistoffen23. Bovendien, onze herziene differentiële centrifugeren aanpak toont een efficiënte methode om ApoBDs te isoleren > 90% zuiverheid23.
In deze paper beschrijven we in detail onze experimentele procedure apoptose inductie, valideren en te detecteren en kwantificeren van de vorming van de ApoBD. De ApoBD isolatie werkstromen met behulp van FACS gebaseerde en differentiële centrifugeren gebaseerde methoden zijn ook uitgewerkt en vergeleken. De representatieve gegevens tonen aan dat de beschreven methodologie een effectieve geavanceerde hulpprogramma voor toekomstige ApoBD studies bevat.

Apoptotic organen (ApoBDs), microvesicles en exosomes zijn de belangrijkste leden van de familie extracellulaire blaasje met ApoBDs wordt één van de grootste type. Er is voorgesteld dat de ApoBDs cel goedkeuring, alsmede de intercellulaire communicatie via mensenhandel biomoleculen kan steun. Conventionele benaderingen gebruikt voor de identificatie en de isolatie van de ApoBDs zijn vaak beperkt door het gebrek aan nauwkeurige kwantificering en lage monster zuiverheid. Hier beschrijven we een werkstroom om te bevestigen de inductie van apoptosis, ApoBD formatie te valideren en isoleren van ApoBDs naar hoge zuiverheid. We zullen ook schetsen en vergelijk fluorescentie-activated cell sorting (FACS) en differentiële centrifugeren gebaseerde benaderingen te isoleren van de ApoBDs. Bovendien zal de zuiverheid van geïsoleerde ApoBDs worden bevestigd met behulp van een eerder stellen stroom cytometry gebaseerde kleuring en de analysemethode. Tezamen, met behulp van de beschreven benadering, THP-1 monocyt apoptosis en de apoptotic cellen te demonteren was geïnduceerd en gevalideerd, en ApoBD gegenereerd op basis van THP-1 monocyten werden geïsoleerd met een zuiverheid van 97-99%.
