Seit seiner frühen Beschreibung in den 1950er Jahren avancierte das Feld der Apoptose deutlich, immer ein prominenter Forschungsgebiet. Trotz der breiten Interesse und umfangreiche Bemühungen sind bestimmte Aspekte der Apoptose, insbesondere die Bildung von ApoBDs, aufgrund des Fehlens geeigneter Methoden nicht gut untersucht worden. Dazu gehören vor allem die Einschränkung bei der Verfolgung von Apoptose Progression und ApoBD Bildung gleichzeitig mit der traditionellen Durchflusszytometrie A5/PI-Analyse und die Veru...

Apoptose, eine gut untersuchte Form des programmierten Zelltods, ist erforderlich, um die physiologischen Homöostase aufrechtzuerhalten und potenziell schädliche Zellen im menschlichen Körper1zu entfernen. Nach der Induktion der Apoptose können apoptotische Zellen (ApoCells) durchlaufen eine Reihe von morphologischen Veränderungen und Zerlegen in kleinen Membrane-springen Vesikel bezeichnet ApoBDs. Insgesamt ist dieser Prozess ist bekannt als apoptotische Zelle Demontage und kann in 3 verschiedene Schritte anhand der Morphologie2,3geteilt werden. Schritt 1 (Plasmamembran Blebbing) zeichnet sich durch die Bildung von Ballon-ähnliche Strukturen auf der Zelloberfläche, bekannt als Blebs4,5. Schritt 2 (Apoptotic Vorsprung Bildung) umfasst die Bildung von langen Membran Vorsprünge z. B. Apoptopodia, Perlen-Apoptopodia und Mikrotubuli Spitzen6,7,8. Schritt 3 (ApoBD Bildung) umfasst schließlich die Fragmentierung des apoptotischen Vorsprünge und/oder ApoCells ApoBDs6,9erzeugen. Bisherigen Erkenntnisse haben eine Rolle von ApoBDs in Beihilfe apoptotische Zelle Abstand und Vermittlung interzelluläre Kommunikation vorgeschlagen. Zum Beispiel wird vorgeschlagen, dass die Zersplitterung der ein ApoCell in ApoBDs kleine "mundgerechte" Stücke generieren kann, die durch Phagozyten2,10,11Umgebung leicht entfernt werden konnte. Darüber hinaus können ApoBDs beherbergen eine Reihe von Biomoleküle wie DNA, RNA und Proteine, die zu den umliegenden Zellen Zellezelle Kommunikation12,13,14zu erleichtern gehandelt werden können. Um diese Prozesse funktional zu untersuchen, ist es wichtig, die drei wichtigsten Parameter einschließlich (i) Validierung der Apoptose-Induktion und ApoBD Bildung, (Ii) Isolierung von ApoBDs und (Iii) die Bestätigung der ApoBD Reinheit zu bestätigen.
Bisher wurden eine Reihe von Methoden, einschließlich der Durchflusszytometrie und Elektronenmikroskopie zur Apoptose und ApoBDs15,16,17,18zu studieren. Allerdings sind ApoBD Erkennung und Quantifizierung oft schwierig oder übersehen. Zum Beispiel routinemäßig verwendet Flow Cytometry-basierte Apoptose assay beschäftigt annexin V (A5, ein Protein, das die externalisierte bindet "Essen-mich" Signal Phosphatidylserin (PtdSer)) und Nukleinsäure Flecken Propidium Jodid (PI)19. Jedoch mithilfe dieses universelle Fleck Kombination Analyse setzt voraus, dass es nur drei Arten der Zelle Teilmengen (lebensfähige Zellen, ApoCells und nekrotische Zellen) in der Probe gibt. Darüber hinaus obwohl von vielen Forschern für die Apoptose als "Goldstandard" betrachtet, Durchflusszytometrie assays und anschließende Datenanalyse schließt oft ApoBDs durch gating zunächst FSC/SSCZwischenprodukt hoher Ereignisse auswählen. Also, vor kurzem haben wir einen neuartige Flow Cytometry Assay mit A5 und TO-PRO-3, ein weiteres Nuclein-Fleck, der selektiv durch Caspase 3/7-aktivierte Pannexin 1 (PANX1) aufgenommen werden kann Kanäle7,20. Wie Caspase 3-induzierte PANX1 Aktivierung PtdSer Exposition in der Frühphase der Apoptose vorausgeht, Flecken TO-PRO-3 differentiell apoptotische und nekrotischen Zellen. Darüber hinaus enthält dieser Ansatz kombiniert mit unserem Roman Anspritzung Strategie alle erworbenen Ereignisse während der Datenanalyse und infolgedessen sechs Zelle/Partikel Teilmengen werden identifiziert, einschließlich: (i) lebensfähige Zellen (FSC/SSCMittelstufe/hohe, A5niedrig , TO-PRO-3niedrige), (Ii) A5– frühe ApoCells (FSC/SSCMittelstufe/hohe, A5niedrig, TO-PRO-3Mittelstufe), (Iii) A5+ ApoCells (FSC/SSCMittelstufe/hohe, A5hoch , TO-PRO-3Mittelstufe), (iv) nekrotische Zellen oder späten ApoCells (FSC/SSCMittelstufe/hohe, A5hoch, TO-PRO-3hohe), (V) ApoBDs (FSC/SSCniedrig, A5zwischen-, TO-PRO-3niedrig / Fortgeschrittene), und (vi) Schutt (FSC/SSCniedrig, A5niedrig, TO-PRO-3niedrige)20. Unser Ansatz betont die Bedeutung der Analyse alle Zellen/Partikel Teilmengen und noch wichtiger ist, die Trennung der ApoBDs von Zellen und Schutt20. So zeigt dieser Ansatz eine effiziente Technik um die Induktion der Apoptose und ApoBD Bildung gleichzeitig zu überprüfen.
ApoBDs traditionell durch eine Vielzahl von Ansätzen Differentielle Zentrifugation isoliert wobei ApoBDs von Zellen oder anderen extrazellulären Vesikel anhand der Dichte getrennt werden können. Allerdings sind solche Zentrifugation Methoden oft durch niedrige ApoBD Reinheit, Mangel an eine Quantifizierung Schritt zur Probe Reinheit und/oder Unfähigkeit, Zelle typspezifischen ApoBDs17,21,22trennen bestätigen begrenzt. Daher entwickelten wir vor kurzem zwei Ansätze, ein FACS-basierten und einen neuen Differentielle Zentrifugation basierenden Ansatz, welche mit unserer vorher festgelegten Flow-zytometrie-Methode, die Induktion der Apoptose und Probe Reinheit23validieren gekoppelt werden kann. ApoBD Isolierung über unsere FACS basierenden Ansatz kann ApoBDs bis zu 99 % Reinheit bereichern und koppelbar mit einer Vielzahl von Zelle-spezifischen Antikörper gegen ApoBDs von gemischten Zellpopulationen, Gewebeproben und Körperflüssigkeiten23zu isolieren. Darüber hinaus zeigt unsere überarbeitete Differentielle Zentrifugation Ansatz eine effiziente Methode, um ApoBDs zu isolieren > 90 % Reinheit23.
In diesem Papier beschreiben wir ausführlich unsere Versuchsanordnung um Apoptose-Induktion, zu validieren und zu erkennen und ApoBD Bildung zu quantifizieren. Auch die ApoBD Isolierung Workflows mit FACS-basierte und Differentielle Zentrifugation basierenden Methoden ausgearbeitet und im Vergleich. Die repräsentativen Daten zeigen, dass die beschriebene Methodik ein effektives modernstes Werkzeug für zukünftige ApoBD Studien bietet.

Apoptotic Bodies (ApoBDs), Microvesicles und exosomen gehören die extrazelluläre Vesikel-Familie, mit ApoBDs als eines der größten Art aus. Es wurde vorgeschlagen, dass ApoBDs Zelle sowie interzellulären Kommunikation durch Menschenhandel Biomolekülen unterstützen kann. Herkömmliche Ansätze zur Identifizierung und Isolierung von ApoBDs sind häufig durch das Fehlen der genaue Quantifizierung und niedrigen Probe Reinheit beschränkt. Hier beschreiben wir einen Workflow zur bestätigen die Induktion der Apoptose, ApoBD Bildung zu überprüfen und ApoBDs zu hoher Reinheit zu isolieren. Wir werden auch zu skizzieren und vergleichen Fluoreszenz-aktivierte Zelle Sortieren (FACS) und Differentielle Zentrifugation basierte Ansätze, ApoBDs zu isolieren. Darüber hinaus wird die Reinheit der isolierten ApoBDs bestätigt werden, mit einer zuvor Flow Cytometry-basierte Färbung und analytische Methode zu etablieren. Zusammengenommen mit dem beschriebenen Ansatz wurde THP-1 Monocyte Apoptose und apoptotische Zelle Demontage induzierte validiert und ApoBD generiert aus Monozyten THP-1 wurden isoliert zu einer Reinheit von 97-99 %.

Hier beschriebene Verfahren verwenden, THP-1 Monocyte Apoptose induziert wurde und ApoBDs wurden erkannt und isoliert entweder über eine FACS-basierte oder eine Differentielle Zentrifugation Ansatz (Abbildung 1).
