MUB40は、ラクトフェリンに結合し、これまでにテストされたヒトおよびすべての哺乳類組織サンプルで特異的な検出を可能にする新しい好中球マーカーである。MUB40で好中球を標識することは容易、速く、そして特定である。それは今、私たちの研究室で広く使用されており、この技術の意味は炎症性疾患の診断にまで及びます。まず、テキストプロトコルに従ってソリューション1、2、3を準備し、一晩無酸素キャビネットに入れます。次いで、採取した血液の管を650gで20分間遠心分離し、細胞を血漿分画から分離する。分離された血液を妨げずに、チューブを無酸素キャビネットに移し、プラズマ分を50ミリリットルの円錐形チューブに移します。この後、無酸素キャビネットからプラズマのチューブを取り出し、20分間2900 gで遠心分離します。ペレット血小板を破壊しないように注意し、チューブを無酸素キャビネットに移し、血小板の悪血漿を新鮮な50ミリリットルのチューブにピペットします。採血管を使用して、円錐50ミリリットルチューブに赤血球を結合する。その後、総容積が44ミリリットルに達するまで0.9%の塩化ナトリウム溶液を加え、6%デキストランの6ミリリットルを加えます。チューブを10〜20回反転させて血液とデキストランの混合物を穏やかに混合し、チューブを少なくとも30分間沈静化させます。チューブ堆積物の間に、4.2ミリリットルのパーコール溶液を5.8ミリリットルのプラズマに加えてパーコール勾配を調製し、チューブを反転して混合します。次に、デキストランの上部分画を含む好中球を採取し、赤血球をピペットしないように注意する。スクリューキャップを締め、無酸素キャビネットからデキストランチューブを取り外し、チューブを300gで10分間遠心します。ペレット化された細胞を邪魔しないように注意し、チューブを無酸素キャビネットに戻します。次に、ピペット処理により液体を取り除く。ペレットを1ミリリットルのプラズマで静かに再懸濁します。ゆっくりとパーコール溶液の上部に再中断セルを追加します。パーコール溶液チューブを無酸素キャビネットから慎重に取り出し、800 gで20分間遠心分離して、PBMCsを好中球から分離します。次に、テキストプロトコルに従って14ミリリットルの洗浄バッファー溶液を調製する。次に、パーコールチューブを無酸素キャビネットに入れる。ピペットを使用してパーコールおよびPBMCを除去し、ペレット含有ペレットを1ミリリットルの洗浄バッファー溶液中に再懸濁させます。次に、200マイクロリットルの磁気ビーズを再懸濁した細胞に加えます。細胞とビーズを少なくとも15分間穏やかに混ぜ合わせてインキュベートします。この後、分離カラムを2ミリリットルの洗浄バッファーで2回洗います。カラムの下に新しい15ミリリットルの円錐管を保持しながら、カラムを通して好中球の赤血球混合物をゆっくりとピペットする。カラムからのフロースルーは曇りで赤色を失い、残りの赤血球からの好中球の完全な分離を確認する。2ミリリットルの洗浄バッファーをカラムの上部に加え、フロースルーを同じチューブに集めます。この洗浄の終わりまでに、流れは明確であるべきです。コレクションチューブを軽く混ぜて、好中球が均等に分配されるようにします。次に、細胞計数から10マイクロリットルのフロースルーを採取した。この後、無酸素キャビネットから好中球チューブを取り出し、チューブを300gで20分間遠心分離します。沈み込んだ好中球を無酸素キャビネットに戻し、ピペットを介して洗浄バッファーを取り外します。次に、プラズマ中のペレットを再懸濁する。RI-MUB40-Cy5の1マイクロリットルをRPMI-1640培地の1ミリリットルに加え、フェノールレッドなしで、ピペットを介して混合します。次に、1マイクロリットルのFMLP溶液とピペットを加え、混合します。ガラス底顕微鏡皿に混合物を移します。その後、適切な量の精製細胞を皿に加えます。最後に、皿を反転蛍光顕微鏡に入れ、画像の取得を開始します。このプロトコルでは、MUB40は、FMLPによるシミュレーション時に好中球分泌を検出するために生細胞に使用された。ラクトフェリンを含む顆粒含有量の放出のダイナミクスは、マゼンタで経時に視覚化され得る。MUB40は、固定好中球に対して追加的に使用することができる。ここで、時効系列で蛍光ビーズを食食いする。好中球顆粒の内容は、青色でここにラベル付けされています。好中球は、初期の時点で細胞染色をほとんどまたは全く示さなかったが、徐々に、RI-MUB40標識は、原形質膜およびビーズ表面の両方で、時間の経過とともに増加した。MUB40はまた、テキスト図に示すように、特に好中球を検出するために固定された炎症性組織に使用され得る。ここでは、私たちの研究室で日常的に適用される好中球精製手順について説明します。他の方法は、適切である、MUB40-Cy5ラベル付けはまだ動作します。好中球は、精製後にMUB40で特異的に標識されてもよいし、患者または動物モデルから直接炎症組織に記載することができる。いくつかのMUB40誘導体は、その使用を広げるために設計されています。その開発後、この技術は、炎症の分野の研究者が炎症性疾患における好中球の募集と活性化を探求する道を開いた。
