Hyperandrogenism is het waarmerk van polycysteus ovarium syndroom (PCOS) volgens de criteria van de NIH en de overtollige androgeen en PCOS (AE-PCOS) samenleving. Vrouwen met PCOS hebben moeite om zwanger en hebben een verhoogd risico van zwangerschap complicaties1. Zelfs als ze zwanger zijn, hebben hun vrouwelijke nakomelingen een gezondheidsrisico resultaten2,3. Dierlijke modellen zijn ontwikkeld met behulp van verschillende strategieën4,5,6,7,8,9,10,11 , 12 en vertonen veel kenmerken van PCOS (anovulatie en of verminderde glucose en insuline tolerantie) met verhoogd lichaamsgewicht en obesitas verbonden aan de grootte van de uitgebreide adipocyte en adipocyte gewicht toegenomen. Er zijn twee belangrijke strategieën om te produceren van dierlijke modellen die worden gebruikt voor het bestuderen van PCOS. Een is behandeling met hoge niveaus van androgenen rechtstreeks (exogene androgeen injectie/invoeging) of niet indirect (zoals het blokkeren van androgeen conversie naar oestrogeen met aromatase remmer) na geboorte13. Een andere is door foetale hyperexposure van androgenen tijdens dracht14,15 te bestuderen van de nakomelingen. Bijvoorbeeld, ontwikkelen vrouwelijke nakomelingen van rhesus monkey16,17, schapen18en knaagdieren blootgesteld aan mannelijke androgeen uteriene deperiode PCOS-achtige eigenschappen later in het leven. Deze modellen aanzienlijk verbeterd ons begrip van verhoogde androgeen effecten, en foetale programing en uteriene milieu-effecten. Echter, deze modellen hebben hun eigen beperkingen: 1) dieren ontwikkelen obesitas en daarom is het moeilijk te scheiden van de gevolgen van de hyperandrogenemia van obesitas geïnduceerde reproductieve en metabole dysfunctie; 2) vóór zwangerschap, vrouwen met PCOS vertonen al hoge niveaus van androgeen, aldus eicellen zijn blootgesteld aan androgeen overtollige vóór bevruchting; 3) de farmacologische doses van testosteron (T) of dihydrotestosteron (DHT) gebruikt na de geboorte of tijdens de zwangerschap kunnen niet overeen met de androgeen-omgeving van PCOS. Niveaus van testosteron en DHT zijn gemeten in ovariële folliculaire vloeistof en/of serum en testosteron en DHT niveaus zijn 1,5 tot 3,9 vouw hoger bij vrouwen met PCOS5,19,20,21 ,22,23 t.o.v. onaangetast vrouwen. We hebben een volwassen muis model23,24,25 dat reproductieve en metabole dysfunctie binnen twee weken na de opening van chronische DHT blootstelling ten gevolge van het inbrengen van een pellet met 4 mm lengte van ontwikkelt gemaakt Crystal DHT poeder (totale lengte van pellet is 8mm). Dit model produceert serum DHT niveaus die over 2-fold hoger (hierna: 2xDHT) dan die van controle Muizen zonder DHT behandeling. De 2xDHT muizen geen wijzigingen van de basale serum estradiol, testosteron en LH Ken vertonen niet ontwikkelen van obesitas, en Toon vergelijkbaar ovariële gewicht, serum niveaus van cholesterol, vrije vetzuren, leptine, TNFα en IL-623,24, 25 ten opzichte bepaalt zelfs maximaal 3,5 maanden na DHT invoeging23,24,25. Bovendien door de paring vrouwtjes die kenmerken van PCOS al ontwikkeld, kunnen we bestuderen het effect van een hyperandrogenic moeders omgeving op de metabolische en reproductieve gezondheid van de nakomelingen15.
Dit nieuwe paradigma (relevante NIH en AE-PCOS samenleving criteria) modellen de ziekte door het produceren van de relatief vergelijkbare niveaus van androgenen aan die van de vrouwen met PCOS 2 - 3-voudig hoger testosteron of DHT niveaus ten opzichte van onaangetast vrouwen. Dit model wordt echter gehandhaafd door de voortdurende exogene DHT en niet van geprogrammeerde endogene hyperandrogenism zodra DHT wordt ingetrokken. Het algemene doel van dit artikel is gericht op 1) hoe maak je de DHT pellet; 2) hoe te genereren een lean-PCOS zoals muismodel; 3) strategieën te evalueren van de vrouwelijke nakomelingen van deze dammen. a hyperandrogenic mouse model to study polycystic ovary syndrome

Hyperandrogenemia speelt een cruciale rol in functie van de reproductieve en metabole bij vrouwtjes en is het waarmerk van polycysteus ovarium syndroom. Het ontwikkelen van een mager PCOS-achtige muismodel dat nabootst van vrouwen met PCOS is klinisch zinvol. In dit protocol beschrijven we een dergelijk model. Door het invoegen van een 4 mm lengte van DHT (dihydrotestosteron) crystal poeder pellet (totale lengte van pellet is 8 mm), en vervangt het maandelijks, zijn wij in staat om te produceren een muismodel van PCOS-achtige met serum DHT niveaus 2 vouw hoger dan muizen niet geïmplanteerd met DHT (neen-DHT). Reproductieve en metabole dysfunctie waargenomen zonder gewicht en lichaamssamenstelling. Terwijl vertoont een hoge mate van onvruchtbaarheid, een kleine ondergroep van deze vrouwelijke muizen van PCOS-achtige kan zwanger en hun nakomelingen laten zien van vertraagde puberteit en toegenomen testosteron als volwassenen. Dit mager muismodel van PCOS-achtige is een nuttig instrument om te studeren de pathofysiologie van PCOS en de nakomelingen van deze PCOS-achtige dammen.

Serum DHT niveaus en Glucose tolerantie test
DHT niveaus werden gemeten vanaf verzamelde serum door beide ELISA en LC-MS volgens protocol 1,24-1,25, 2.9 en 3.0. Deze methode kan helpen bij het beantwoorden van belangrijke vragen op het gebied van reproductie. En de endocrinologie. En zijn van toepassing op de studie van polycystend eierstoksyndroom of PCOS en hyperandrogenisme.Het voordeel van deze techniek is dat het een consistente, uniforme, lange termijn afgifte van hormonen op een kostbare effectieve manier vergemakkelijkt. JHUs de behandeling van de procedure zal Ping Xue, een technicus uit mijn laboratorium. Voor dihydrotestosteron of DHT pellet voorbereiding, gebruik een scheermesje om een 15 millimeter stuk siliconen buizen te snijden.En gebruik een drie milliliter spuit uitgerust met een afgestompte 20 meter naald om twee tot vijf millimeter medische lijm siliconen injecteren in een uiteinde van de buis. Laat de slang 's nachts drogen. En controleer op luchtbellen aan het verzegelde uiteinde.Het dragen van de juiste persoonlijke beschermingsmiddelen giet DHT poeder in een plastic wayboat in de flowhood. Maak indruk op het geopende uiteinde van de met lijm bedekte buis in het poeder. Met behulp van een grote rechtgetrokken paperclip, tamp het poeder naar beneden in de slang, totdat de DHT bereikt de juiste experimentele diepte.Sluit vervolgens de geopende kant van de buis 's nachts af met siliconen. Bewaar vervolgens de pellets in de 50 milliliter conische buis verpakt met folie op kamertemperatuur voor maximaal drie maanden.Voor PCOS muis model generatie equilibrate tot 20 DHT of controle pellets in aparte 50 milliliter conische buizen met 30 milliliter van 0,9 procent zoutlijn gedurende 24 uur bij 37 graden Celsius. De volgende dag, bevestigen gebrek aan reactie op teen knijpen in een verdoofde twee maanden oude vrouwelijke muis en gebruik steriel gaas om sequentiële Povidone-jodium en 70 procent ethynol scrubs toe te dienen aan de chirurgische gebied. Maak vervolgens een incisie van vijf millimeter door de huid in de buurt van de nek.En gebruik een trocar van tien meter om een 15 millimeter tunnel te maken in de rostral-richting. Gebruik de trocar om een pellet dorsally in de tunnel te steken en de incisie af te dichten met de chirurgische lijm. Bespree de wondranden handmatig met tangen en gebruik zachte borstelstreken om drie dunne filmlagen vloeibare lijm aan te brengen tot één centimeter van elke kant van de tegengestelde wondranden.Plaats de muis vervolgens alleen in een kooi op een warmtekussen met controle tot volledige recumbency. Het vervangen van de pellet om de vier weken om een constant niveau van androgeen blootstelling te handhaven. Drie dagen na het inbrengen van pellets, gebruik een p10 pipet te spuiten tien microliters van steriele zoutoplossing in de vaginale holte en gebruik de pipet tip om onmiddellijk te herstellen van de zoutoplossing.Verdeel het vaginale celzoutmonster over een gelabelde dia. En laat de zoutoplossing drogen op kamertemperatuur. Bevestig de gedroogde glijbanen in een container van 100 procent ethynol gedurende ten minste vijf minuten.Vlek vervolgens de dia's voor een minuut elk in buffer B en buffer C van een dif-quick kleuring kit. Aan het einde van de buffer C incubatie was de glijbanen met kraanwater gedurende drie seconden en droog ze op kamertemperatuur. Om de estrous cyclicity te bepalen bekijken de dia's onder de tien keer doel van een lichte microscoop.Tel de dagen in elke fase en deel door het totale aantal dagen om procentuele tijd in elke fase te berekenen. Om de glucosetolerantie te testen, 20 dagen DHT inbrengen, snel de muizen 's nachts. Weeg de muizen de volgende ochtend, en gebruik een glucometer en teststrips om basilicum glucose niveau van staart ader bloed te meten.Injecteer de muizen met twee gram per kilogram lichaamsgewicht van 20 procent dextrose. Meet vervolgens het glucoseniveau van staartaderbloedmonsters op 15, 30, 60, 90, 120 minuten na dextrose toediening. Om vrouwelijke nakomelingen van DHT ingebrachte dames te verwerven, verplaatst u de vrouwelijke muis om in de kooi te worden getest met een bewezen vruchtbaar mannetje op dag 15, na het inbrengen van pellets.Documenteer de dames lichaamsgewicht elke week om te bepalen of de muis zwanger is. Op postnatale dag zeven, dip een lancet tip in tattoo plakken en markeer elke pup met een tatoeage op de teen. Weeg de gemarkeerde muizen om de zeven dagen tot 70 dagen oud.Tussen postnatale datum 12 tot 16, oor punch de muizen om ze te nummeren volgens het systeem zoals geïllustreerd in de figuur. Om het geslacht te onderscheiden, onderzoekt u de ventrale kant van elke getagde muis. De vrouwtjes hebben zichtbare tepels.Voor het verzamelen van bloedmonsters voor verschillende downstream-analyses gebruikt u een lancet om het geschatte experimentele volume van bloed tussen negen en tien uur te verzamelen door submandibulaire aderbloedingen. En centrifugeren de monsters om serumverzameling mogelijk te maken. Bewaar vervolgens de serummonsters in 1,5 milliliter microcentrifugebuizen bij min 80 graden Celsius tot hun analyse.Om de puberteit bij de DHT vrouwelijke nakomelingen te beoordelen, controleer elke dag na het spenen op postnatale dag 21 de vaginale opening door visuele inspectie. En gebruik kalibers om de ano-genitale afstand te meten. Hoewel de absolute waarden van serum DHT verschillen tussen massaspectrometrie en Elisa-analyse, is de relatieve volksverandering van DHT versus no-DHT-invoeging vergelijkbaar met zowel tests als verschillende experimenten.DHT niveaus zijn aanzienlijk verhoogd van pre-conceptie tot zwangerschap in zowel DHT en geen DHT muizen. Dht-niveaus zijn echter tweevoudig hoger bij DHT-muizen in vergelijking met geen DHT-muizen op zowel pre-conceptionele als zwangerschapstijdpunten. Volwassen DHT-dochters vertonen verschillen in reproductieve functie.Bijvoorbeeld, verstoorde eestrous cyclicity in vergelijking met volwassen geen DHT dochters ervaren aanzienlijk langere tijden in metestrus diestrus en minder tijd in pro-esterous en esterous. Tijdens een poging deze procedure is het belangrijk om te onthouden om de pellets uit te broeden voor 24 uur voordat ze worden gebruikt voor uniforme afgifte van het hormoon. Na deze procedure kunnen ook de reproductieve en metabolische gevolgen van DHT-blootstelling worden onderzocht.Na de ontwikkeling maakte deze techniek de weg vrij voor onderzoekers op het gebied van endrochronologie en moederlijke foetale interacties om PCOS en de impact van hyperandrogenisme op laag niveau bij vrouwelijke muizen te bestuderen. Vergeet niet dat het werken met DHT zeer gevaarlijk kan zijn en de voorzorgsmaatregelen zoals het dragen van handschoenen en een masker en een bril, en werken in een bio-veiligheidskap, moet altijd worden genomen tijdens het uitvoeren van deze procedure.
