Hiperandrogenismo es el síndrome de ovario poliquístico (SOP) según los criterios del NIH y del exceso de andrógenos y SOP (AE-SOP) la sociedad. Las mujeres con SOP tienen dificultad para quedar embarazadas y han aumentado el riesgo de embarazo complicaciones1. Incluso si consiguen embarazadas, su descendencia femenina tiene una de2,de los resultados adversos para la salud3. Se han desarrollado modelos animales usando varias estrategias4,5,6,7,8,9,10,11 , 12 y exhibe muchas características del SOP (anovulación, y tolerancia deteriorada de la glucosa y la insulina) con mayor peso corporal y obesidad había asociada con el tamaño del adipocito agrandado y aumentó el peso del adipocito. Hay dos estrategias principales para producir modelos de animales que se utilizan para el estudio de PCOS. Uno es el tratamiento con altos niveles de andrógenos directamente (inyección de andrógenos exógenos, inserción) o indirectamente (por ejemplo, bloqueando la conversión de andrógenos a estrógenos con inhibidor de la aromatasa) después nacimiento13. Otro es por hyperexposure fetal de andrógenos durante la gestación14,15 estudiar la descendencia. Por ejemplo, la hembra crías de mono rhesus16,17, ovejas18y roedores expuestos a niveles masculinos de andrógenos durante el período intrauterino desarrollan SOP-como rasgos más adelante en la vida. Estos modelos habían mejorado significativamente nuestra comprensión de los efectos de andrógenos elevados y programación fetal y uterinos efectos ambientales. Sin embargo, estos modelos tienen sus propias limitaciones: 1) los animales desarrollan obesidad y por lo tanto es difícil separar los efectos del hyperandrogenemia reproductiva inducida por la obesidad y la disfunción metabólica; 2) antes del embarazo, las mujeres con SOP ya exhiben altos niveles de andrógenos, así ovocitos han sido expuestos a andrógenos en exceso antes de la fecundación; 3) las dosis farmacológicas de la testosterona (T) o dihidrotestosterona (DHT) usado después del nacimiento o durante la gestación pueden no reflejar el ambiente de andrógeno de PCOS. Se han medido los niveles de testosterona y DHT en suero y en líquido folicular ovárico y testosterona y los niveles DHT son 1.5 a 3.9 veces mayor en mujeres con SOP5,19,20,21 ,22,23 en comparación con las mujeres. Hemos creado un ratón adulto modelo23,24,25 que se desarrolla la disfunción reproductiva y metabólica dentro de dos semanas de la iniciación de la exposición crónica de DHT de la inserción de un pellet con 4 mm de longitud de polvo de cristal DHT (longitud total de la pelotilla es 8 m m). Este modelo produce suero DHT niveles sobre 2 superior (denominado 2xDHT) que el de los ratones de control sin tratamiento de DHT. Los ratones 2xDHT no presentan alteraciones del estradiol del suero basal, testosterona, LH y no no desarrollar obesidad y muestran similar peso ovárico, los niveles séricos de colesterol, ácidos grasos libres, leptina, TNFα y IL-623,24, 25 relativas a controles hasta 3,5 meses después de DHT inserción23,24,25. Además, por el apareamiento de las hembras que ya han desarrollado características del SOP, podemos estudiar el impacto de un ambiente materno hyperandrogenic en la salud metabólica y reproductiva de la descendencia15.
Este nuevo paradigma (pertinente a los criterios NIH y sociedad AE-SOP) modelos de la enfermedad mediante la producción de niveles relativamente similares de andrógenos a las mujeres con SOP 2 - 3-fold mayor testosterona o niveles de DHT en comparación con las mujeres. Sin embargo, este modelo se mantiene por DHT exógena continua y no de hiperandrogenismo endógeno programada, una vez que se retira el DHT. El objetivo general de este artículo es concentrarse en 1) Cómo hacer que el perdigón DHT; 2) Cómo generar un SOP lean como modelo de ratón; 3) estrategias para evaluar la descendencia femenina de estas represas. a hyperandrogenic mouse model to study polycystic ovary syndrome
