DOPA er en aminosyre som er involvert i biosyntesen av katekolaminer i dyr og planter. Denne aminosyren er syntetisert fra Tyr av tyrosin hydroksylase og molekylære oksygen (O2)1. Fordi DOPA er en forløper til dopamin og kan trenge gjennom blod - hjerne barrieren, har det vært brukt i behandling av Parkinsons sykdom2. DOPA er også funnet i blåskjell vedheft proteiner (kart), som er ansvarlig for egenskapene selvklebende blåskjell i våte forhold,3,,4,,5,,6,,7. genetic incorporation of biosynthesized l-dihydroxyphenylalanine (dopa) and its application to protein conjugation

L-dihydroxyphenylalanine (DOPA) er en aminosyre som finnes i biosyntesen av katekolaminer i dyr og planter. På grunn av bestemt biokjemiske egenskaper har aminosyren flere bruker i biokjemiske programmer. Denne rapporten beskriver en protokoll for genetisk inkorporering av biosynthesized DOPA og sin søknad til protein Bøyning. DOPA er biosynthesized av en tyrosin fenol-lyase (TPL) fra catechol, pyruvate og ammoniakk aminosyren er direkte tatt inn proteiner av genetisk innlemmelse metoden bruker en utviklet aminoacyl-tRNA og aminoacyl-tRNA synthetase par. Direkte tilknyttet systemet inkorporerer effektivt DOPA med lite innlemmelse av andre naturlige aminosyrer og bedre protein avkastning enn forrige genetisk innlemmelse systemet for DOPA. Protein Bøyning med DOPA inneholder proteiner er effektiv og områdespesifikke og viser nytten for ulike applikasjoner. Denne protokollen gir protein forskere med detaljerte prosedyrer for effektiv biosyntesen av mutant proteiner som inneholder DOPA på ønskede områder og deres Bøyning for industri og farmasøytiske.

Uttrykket systemet direkte inkorporering av DOPA biosynthesized fra en TPL er vist i figur 1. Genene for utviklet aa-tRNA og aaRS paret er plassert i en plasmider og GFP genet (GFP-E90TAG) som inneholder et gult codon posisjonen 90 ligger i en annen plasmider evaluere inkorporering av DOPA ved GFP fluorescens. TPL genet er plassert i det samme uttrykket plasmider inneholder GFP genet og constitutively uttrykk for å maksimere avkastningen av DOPA biosyntesen....
