1. 动物习惯

5. 数据分析

cpp 范式以经典 (pavlovian) 条件原则为基础, 通常用于衡量吗啡4、5、9、10 等滥用药物的奖励效果, 因为吗啡很容易在实验室中的可用性。典型的 cpp 范式包括三个阶段: 预调理、调理和测试阶段。在调理阶段, 药物的奖励特性 (无条件刺激) 与一个或多个中性上下文刺激配对, 这些刺激在几个调理配对后获得奖励属性, 并作为条件反射。在测试阶段, 动物对药物配对隔间的偏好被测量和反映为他们的奖励记忆。因此, cpp 范式也被用来研究学习和记忆的机制。为了建立吗啡的长期奖励记忆, 对影响建立 cpp 的变量进行了调整和优化, 并采用了以下描述的协议11。上下文刺激是视觉、触觉和气味暗示的组合。根据不同的研究目的, 对上下文刺激和测试时间点进行了变化。例如, 在我们之前的研究12中, 体感提示 (不同的地板纹理) 主要用作上下文刺激, 以研究岛屿皮层的尾端部分, 一个具有体感功能的大脑区域, 在习得中的作用和长期奖励记忆的维护。该装置的设计和大鼠对首选方的分配影响了 cpp 的建立。下面的协议中使用了三腔装置而不是双室装置, 以避免强制选择。两个侧室的平衡和动物的初始分配影响 cpp4,13的获取。因此, 本协议采用了侧室的平衡设计和动物的偏颇分配, 以获得强烈的位置偏好。长期奖励记忆的维护需要在调理阶段进行更多的调理试验。为了降低调理阶段的时间成本, 每天进行两次调理试验, 并平衡吗啡注射, 以避免吗啡在盐水配对试验中的急性戒断作用4。
建立了加强动物模型14 , 对吸毒成瘾周期的前置/预期阶段进行了建模。还利用 cpp 研究了模拟成瘾行为复发的恢复 5.复发是戒毒治疗的障碍之一。渴望是反复复发的原因之一。在禁欲期间, 对药物的渴望不断增加, 这种现象被称为对渴望的孵化, 即 15,16。在药物自我管理范式中, 对渴望的孵化进行了广泛的研究, 而不是对 cpp 范式进行了研究。李等人首先提出了使用 cpp 范式来反映渴望的孵化.17. 其结果17显示, 在禁欲的头 14天, 低剂量吗啡 (1 mg/kg 和 3 mg kg) 但不是高剂量 (10mg/kg) 的 cpp 分数逐渐增加。同样, 注射低剂量可卡因的小鼠的 cpp 得分随着时间的推移有所增加18。低剂量吗啡 cpp 得分的增加被用来反映孵化后的渴望 17.建议在药物相关环境中使用的持续时间和进入相关环境的次数, 以反映 cpp 范式19中强化的有条件的反应。最近的大多数研究都使用 cpp 分数来直接或间接地反映在药物配对腔中花费的时间, 而作为参数进入侧室的数量往往被忽略。由于这些原因, 使用了 10 mg/kg 的吗啡剂量, 并在我们的研究11中测量了进入量。结果表明, 在禁欲期间,1 1号期间, 老鼠进入侧室的情况显著增加。这表明, cpp 分数和条目数量都是评估对药物的渴望增加的有用参数, cpp 范式适用于对渴望现象的孵化模型。然而, 为了减少所需的动物数量, 对一组有吗啡经验的动物进行了反复试验;因此, 灭绝的影响不能排除在主题设计中的当前。在具有相同协议的主题设计之间, 可以克服这一限制。

a conditioned place preference protocol for measuring incubation of craving in rats

吗啡组的21只大鼠和盐水组的10只大鼠被用来测试长期有益记忆的表达。混合方差分析显示, 组与测试之间的相互作用有显著差异 (f (3, 87) = 4.973, p = 0.003), 测试 (f (3, 87) = 18.237, p & lt; 0.001) 和组 (f (1, 29) = 11.413, p = 002)。吗啡组的试验 (f (3, 60) = 46628, p & lt; 0.001) 之间存在显著差异, 但未检测到盐水 group(F(3,27) = 1.576, p = 0.001)。通过与邦费罗尼调整的多重对比分析, 显示三种试验与吗啡组 t0 有显著差异。如图 2所示, 吗啡奖励记忆至少持续了 18天, 并在时间依赖的趋势下减少。数据来自先前发表的研究报告11。

在 t1 (t(1,21) =-2.833 时, 大鼠进入吗啡配对隔间的频率高于盐对的隔间, p = 0.010) 和 t2 (t(1,21) =-4.458, p = 0.0002), 但不是 t3 (t(1,21) =-0.471, p = 0.642) (双尾配对 t 测试)。
盐水组 与吗啡组相似, 采用了混合方差分析, 并显示组与测试 (F(3,54) = 0.345, p = 0.793)、测试 (F(3,54) = 1.793、p = 0.345) 和组 (F(1,18) = 0.345, p = 0.345) 之间的相互作用没有显著差异。因此, 随着时间的推移, 盐水组侧室的入口是稳定的。
