この方法は、前立腺癌の遺伝子操作マウスモデルの開始と、生体内およびインビトロ分析を通じた前立腺癌メカニズムの研究に役立つ。この技術の主な利点は、組織解剖から細胞分離および培養まで、前立腺癌マウスモデルの完全な分析を可能にすることです。泌尿生殖器系(UGS)を暴露した後、尿膀胱を中型の鈍い鉗子でしっかりとつかみ、マウス腹部からシステム全体を持ち上げる。膀胱と前立腺の下にハサミを滑り込まして脊髄を切開し、残りの接続を切り裂いてUGS全体を取り除きます。2~6ミリリットルのPBSを含む6センチメートルのペトリ皿に、解剖顕微鏡でUGSを移します。そして、細かい鉗子とマイクロ解剖はさみを使用して、前立腺組織を切り取ることなく、組織システムの側側と側腹側の両方から脂肪のすべてを慎重にクリアします。すべての脂肪が取り除かれたら、鉗子で膀胱を引っ張り、膀胱をそのベースで切除する。残りの組織腹側を上に置き、鉗子で1つのダクトの端を保持し、はさみでそのベースに容器に従い、そのベースで容器を切除します。反対側の同じ容器を取り外し、必要に応じて隣接する結合組織の切り取りができるように、精嚢と前立腺の間に鉗子を挿入します。その後、尿道の基部に精嚢をトレースし、それらを穿刺することなく、血管を除去します。両方の小胞が取り除かれたら、蝶の羽に似た後部葉が見えるように組織の裏側を上に置きます。各背部葉を鉗子で保持し、そのベースの組織をはさみで切断して背側葉を採取し、組織を腹側に回す。小さく、通常は側に尿道を包み、同じように前葉、腹側葉、および側葉の間にくさび状にする横葉を収集します。側葉より大きく、尿道腹腔に横たわる腹側葉を集める。次に、尿道を切って捨てて前葉を収穫する。3D培養のために前立腺組織を処理するには、ローブを2〜3ミリリットルのDMEMを含む10センチメートル皿に移し、メスを使用して前立腺管管をできるだけ細かく均等にミンチします。組織断片を滅菌組織培養フードに移し、組織溶液を新しい15ミリリットルチューブに加える。新鮮な培地でチューブ内の体積を最大9ミリリットルにし、TenXコラゲナーゼストック溶液を1ミリリットル加えます。ボルテックス後、37°Cで2時間揺れでチューブをインキュベートし、細胞外マトリックスを劣化させます。インキュベーションの終わりに、遠心分離によって組織を収集し、EDTAの暖かい05%ストリップの2ミリリットルにペレットを再中断します。セル対細胞および細胞間接着の切断を容易にするために摂氏37度の5分後、広いボードチップを持つP1000パイプヘッドを使用して、組織を8〜10回三分して塊を分割します。P200 パイプヘッドチップで解離を繰り返します。その後、完全な培地の3ミリリットルでトリプシンを中和します。500単位のDNASE1を組織スラリーに混ぜ、18ゲージの針を装備した5ミリリットルの注射器を通して溶液を5〜10回、続いて20ゲージの針を通して5回渡します。これ以上大きな組織片が見えない場合は、40マイクロメートルのフィルターを通して50ミリリットルの円錐管に懸濁液をろ過し、遠心分離によって細胞を収集する。細胞めっきおよび培養の場合、ペレットを完全な前立腺上皮細胞増殖培地の0.5ミリリットルで再懸濁し、前立腺上皮細胞増殖培地濃度の1ミリリットル当たり5倍10〜5番目の細胞に細胞をアリュードした。細胞を細胞外基質と混合し、溶液比を2~3体積で、適切な実験細胞培養容器に細胞懸濁液をプレートします。その後、細胞を細胞培養器に30分間入れます。細胞外マトリックスが固まった場合、前立腺上皮細胞増殖培地を温め込んだ完全な前立腺上皮細胞で培養液を覆い、基質膜細胞外マトリックスプラグを乱さない様に注意する。その後、細胞を5〜10日間培養器に戻し、2〜3日ごとに培地の半分をリフレッシュする。球体を収穫するために、基底膜細胞外マトリックスゲルプラグを乱さずに培地を慎重に吸引し、基底膜外細胞マトリックスの100マイクロリットルに対して1ミリリットルのディスベース溶液を添加する。セルスクレーパーを使用して、プレートの底に沿って掻き取り、プレートの底から上澄み板に細胞外ゲルを持ち上げ、溶液全体を1回パイプアップしてプラグを小さくします。細胞培養インキュベーター内の細胞外マトリックス片を細胞外基質マトリックスが完全に溶解するまで1~2時間インキュベートする。その後、細胞懸濁液を15ミリリットルのチューブに移します。さらに下流操作のために遠心分離により細胞を収集する。マウス前立腺は、陰部および腹腔内に位置する4組のローブから、精嚢および尿道に位置する。前立腺葉は、複数の腺プロファイルで構成され、それぞれが分泌上皮細胞に囲まれた内腔で構成される。葉の形、細胞の組織、分泌の性質はローブによって異なります。単離された前立腺細胞は、基原膜細胞外マトリックス培養の4日後にオルガノイドに成長し始める。次の1〜6日間で、細胞のほとんどは部分的または完全な内腔を示す球の一部を有する固体球体に成長する。スフェロイドはまた、F-アクチン染色と共局化する細胞間接合部における強力なベータカテニン染色を示す。この手順を試みる間、細かく微細解剖の手順に従うことは非常に重要です。例えば、前立腺葉がまだ尿道に付着している間に分離するので、尿道に関してどのローブがどのローブに基づいているかを知ります。解剖手順に従って、単離されたローブは、RMAシーケンシング、ウェスタンブロッティングまたは免疫染色などの下流分析に使用できます。主要な3D培養技術は、形態および行動の変化のためにスフェロイドで生命を監視し、同じ中の腫瘍学的変化を識別することができるので、前立腺癌メカニズムに関するさらなる洞察を得ることを可能にする。要約すると、この前立腺解剖および培養方法は、遺伝子組み換えマウスモデルを用いて前立腺癌のメカニズムを調査するための貴重な情報を提供するために、様々な下流アプリケーションに組み込むことができる。
