1. 仪器和材料的制备

</ol>2. 切除肺组织
<ol><li>在4°c 的 dmem f-12 培养基中立即储存新鲜的无肿瘤肺组织的叶切除切除术, 直至第3步。</li>

</ol>3. 琼脂糖填充前的检测和选择已切除的组织

	<li>根据表 1中定义的肺琼脂填充评分的标准对组织质量进行评分。</li>

</ol>4. 琼脂糖填充物引起的肺组织膨胀
<ol><li>从存储介质中提起组织, 并从组织中排出多余的介质。将肺组织转移到1.1.2 制备的15厘米培养皿中。</li>
	<li>用1.1.3 的低熔点琼脂糖填充一个30毫升的注射器。</li>

</ol>5. 精确切割肺切片

</ol>6. pcls 冲床的产生

</ol>7. 组织培养和样品采集

根据导管的位置, 大部分组织可以用液体琼脂糖填充 (图 2d)。或者, 环状的肺组织的部分, 反映肺的实质通风被穿透的支气管, 可能会充满琼脂糖 (图 2c)。在这两种情况下, 都可以观察到固体填充组织区域的特征模式: 第一, 组织的主要部分是在楔形中填充的 (图 2d), 其次是出现一个较小的突出圆形区域的完全填充组织区域 (图 2c)。如果部分气道由于琼脂糖凝块或其他原因而阻塞, 部分组织可能无法正确填充琼脂糖。因此, 只有部分组织可能适用于切片。在琼脂糖灌装过程中出现泄漏的情况下, 填充的呼吸树的部分可能会穿孔, 肺组织的填充几乎不可能, 然而, 可能的解决方法包括通过更外围支气管的灌装, 更深的插管进入远端气道 (图 2g), 或潜在夹紧泄漏区域 (图 2G)。
精密切肺切片 长度和宽度为 1-1. 5 厘米的组织块从完全充满凝固琼脂糖的组织区域切除 ( 图 3a-3b)。接下来, 单个组织块粘附在振动体的组织支架上 (图 3c)。生成了500μm 厚的 pcls, 而振动的组织块以3-12μms 的速度向前推进 (图 3d-3f)。最后, 将 pcls 淹没在含有0.1% 胎牛血清的细胞培养培养基中, 并在标准细胞培养条件下进行培养, 如步骤7所述。
