この謎は、葉芽細胞へのタンパク質入力などの細胞生物学の分野でいくつかの質問に答えるのに役立ちます。この技術の主な利点は、葉包体へのタンパク質入力がen vivo条件下で研究できることです。手順を実証するには、私の研究室から2つのポストアウトであるジュンホ・リーとヒョンジュ・カンです。まず、1から2つのB5プレートに2週齢の植物から無傷の葉組織を収穫します。外科用ナイフを使用して葉を収穫し、25ミリリットルの酵素溶液を含む50ミリリットルの円錐形チューブに入れます。チューブをロータリーシェーカーの上に水平に置き、ペチオールと茎だけが溶液に残るまで、暗闇の中で摂氏22度で穏やかな攪拌で8〜12時間インキュベートします。インキュベーションを完了した後、新鮮なペトリ皿に140マイクロメートルの細孔サイズのメッシュを介して放出されたプロトプラストを含む酵素溶液をフィルター処理します。その後、慎重に50ミリリットルの円錐管に21%スクロース溶液の15ミリリットルの上に溶液を重ねます。98 gsのスイングバケットローターでチューブを10分間遠心し、最も低い加速と減速設定を行います。その後、パトゥラピペットを使用して、酵素溶液を含む最上層および酵素溶液とスクロース溶液の界面から慎重にプロトプラストを除去します。この溶液をW5溶液の30ミリリットルを含む50ミリリットルの円錐管に移し、チューブを反転して混合します。チューブを51 gsで6分間遠心分離します。ピペットを使用して、ペレット内のプロトプラストを邪魔することなく、慎重にこの上清を捨てます。25ミリリットルのW5溶液を加え、プロトプラストを静かに再中断します。安定化のために、最低1時間、摂氏4度の冷蔵庫でインキュベートします。摂氏4度インキュベーション後、プロトプラストを46gsで2分間遠心して完全にペレット化する。慎重にスーパーナテントを完全に取り出し、PROTOPLASTペレットにMANG溶液を加え、1ミリリットル当たり6倍に10倍を達成します。ピペットを使用して、10マイクログラムのプラズマDNAと300マイクロリットルのプロトプラスト溶液を新鮮な13ミリリットルの丸底チューブに加えます。チューブを手で穏やかに回転させることで混合し、すぐに作りたての40%PEG溶液の300マイクロリットルを加えます。チューブをほぼ水平に傾け、手で数回穏やかに回転させることで完全に混ぜます。室温で30分間インキュベートします。その後、W5溶液の1ミリリットルを追加し、以前のように手で軽くチューブを回転させることによって完全に混合します。試料を室温で10分間インキュベートする。最初の1.5ミリリットルを加え、次に2ミリリットルのW5を加え、最終的な添加と混合の後、30分間インキュベートしてさらに2回繰り返します。46 gsでチューブを4分間遠心し、上清を捨てます。ペレットに2ミリリットルのW5溶液を加え、以前と同じように穏やかに、しかし完全に混ぜます。暗い部屋で摂氏22度で18時間インキュベートします。蛍光顕微鏡を用いてタンパク質のインポートを分析するには、トリミングされた先端を持つピペットを使用して、ガラススライド上にプロトプラスト溶液の10マイクロリットルを配置します。カバースリップを使用して慎重に解決策をカバーし、プロトプラストを損傷しないようにします。緑色蛍光タンパク質とクロロフィルの自己蛍光を観察するために設定された励起入フィルターを使用して、蛍光顕微鏡のステージにスライドを置きます。冷却された電荷カップルデバイスカメラを使用して画像をキャプチャし、イマジン編集ソフトウェアを使用して疑似カラー画像を生成して処理します。総タンパク質抽出と免疫ブロッティングの場合は、プロトプラスト溶液から1ミリリットルを取り出し、残りの1ミリリットルを混合します。このプロトプラスト溶液を遠心分離管に移し、46 gsで遠心分離機を4分間移します。遠心分離が完了した後に上清を取り除き、80マイクロリットルの変性バッファーを追加します。5秒間勢的に渦を打ち出す。5つのXDSサンプルバッファーを追加し、よく混ぜ、変性するために10分間沸騰させます。標準STSページと抗GFP抗体による免疫ブロット法を進めます。RbcSおよびTGFTは、GFPに融合したトランジットペプチドを含むRBCSの79末端末端アミノ酸残基をコードする融合構築物であり、蛍光顕微鏡および免疫ブロット法による葉包体へのタンパク質の輸入を研究するために使用された。蛍光顕微鏡で調べた場合、標的タンパク質からの緑色蛍光シグナルは、葉緑体へのタンパク質輸入を示すクロロフィル自己花序からの赤色蛍光シグナルと結合した。総タンパク質を単離した後、免疫ブロットでは2つのタンパク質バンドが観察され、タンパク質が葉白に適切に輸入されたことを示す。上のバンドは、葉芽突きにインポートした後、処理された形式に全長前駆体と下帯に対応しています。タンパク質の処理された形態の量は時間依存的に増加し、タンパク質が葉白質に輸入されることを示唆している。この開発では、この技術は、細胞生物学の分野の研究者がシロイヌナズナでタンパク質の疲れや膜糸摘出を探求する道を開いた。
