直接血统重新编程从成纤维细胞到红细胞祖细胞。这是我们在实验室中开发的新方法，以便更好地研究决定红细胞命运的转录调控过程。使用直接顺性重新编程来研究开发的主要优点是，它是一种非常直接的技术。因此，与敲除小鼠功能研究的损失相比，在这里我们可以非常容易地操纵因子，在特定时间点过度表达，然后很容易获得足够的材料来研究下游效应，如染色质修饰或基因表达变化。虽然此方法侧重于将小鼠尾尖成纤维细胞重新编程为红细胞祖细胞，但也可以应用于其他成纤维细胞类型，以及包括人类在内的其他生物体。这项技术的影响延伸到输血医学，因为它为体外红血球的产生铺平了道路。演示这个程序将是来自我的研究小组的奥尔班·约翰逊。首先用0.1%明胶覆盖盘子的表面。在37摄氏度下孵育约20分钟。吸明胶溶液，让盘子干燥。用剪刀在安乐死鼠标尾部底部剪下来，将其取出。将尾部放在 DPBS 中，用 2%FBS，直到准备好使用。在无菌条件下的组织培养罩中，在 DPBS 中准备 0.02% trypsin EDTA 的稀释尝试素溶液，并将 5 毫升加入未涂覆的 10 厘米培养皿中。在组织培养罩中，用足够70%乙醇在15毫升管中清洗尾巴，以覆盖它，然后在DPBS中清洗。将尾部平放在盘子中，用钳子固定到位。使用手术刀沿其从底座到尖端的纵向轴在尾部上切口。使用一对钳子抓住尾巴并垂直握住它，然后用第二对钳子抓住尾巴底部切口旁边的皮肤，然后剥回。在切口的两侧执行此操作，直到皮肤可以通过向下向尾部尖拉去剥落。用钳子将剥落的尾巴放在含有三辛溶液的盘子上。使用解剖剪刀将尾巴切成小块。使用剪刀将 trypsin 溶液中的这些碎片切成更小的碎片，然后在 37 摄氏度下孵育 10 分钟。要淬火，加入两卷FEX介质，DMEM与补充剂和抗生素。将整个盘子的内装物收集到50毫升的管子中。在4摄氏度下将管在350倍G下离心5分钟。吸制上一液，并在10毫升新鲜FEX介质中重新暂停含有尾部碎片的颗粒。将此悬浮液转移到明胶涂层的盘中。在37摄氏度的5%二氧化碳和4%的氧气中孵育，每两天加入一次新鲜的FEX介质。观察五到七天后的文化，其中尾部碎片附着在菜的底部，成纤维细胞正在远离它们。一旦一组成纤维细胞可见，轻轻摇动盘子，以驱逐尾片。用所有的骨片吸气介质，使成纤维细胞附着在板上。加入10毫升的新鲜FEX介质，培养成纤维细胞，直到汇合。为确保成纤维细胞培养中造血源的祖细胞没有污染，使用手稿中描述的 trypsin EDTA 将细胞从盘子里分离。加入介质并收集细胞，并加入含有抗体的磁珠。使用磁性分离系统去除表达造血标记的细胞。包括此步骤是绝对重要的，因为它消除了文化中可能存在的任何 HSPC，因此消除了 FACS 分析当天出现任何误报的可能性。种子逆转录病毒包装细胞在大约25，000个细胞每平方厘米在组织培养处理的菜。在37摄氏度和高葡萄糖DMEM中孵育细胞，在一夜之间孵育5%的二氧化碳。第二天早上，取出介质，添加一半的体积用于培养无添加剂的DMEM过夜。下午，检查细胞的转染所需的70%至80%的汇合。开始转染，准备一个2到1的表达载体PMX的六微克的混合物和三微克的帮助器载体，其中包含Gag，Pol和包络基因，每个重新编程因子。接下来，对于每个重新编程因子，在无菌聚苯乙烯管中加入300微升室温DMEM。然后小心地将27微升的室温商用转染试剂直接加入介质，避免与管壁接触。将质粒混合物加入含有管的转染试剂中。涡流在室温下短暂孵育混合物15分钟。涡流转染试剂DNA混合物再次。将其滴滴添加到逆转录病毒包装细胞中，以便其均匀地分布在培养体上，并在37摄氏度下孵育。转染后24小时，取出介质，加入DMEM与20%FBS和100单位每毫升青霉素链霉素。转染后48小时，收集上清液，并通过0.22微米孔径注射器过滤器过滤。开始转导，在FEX介质中，每平方厘米以10，000个细胞播种尾尖成纤维细胞，在0.1%的明胶预涂层盘上。在37摄氏度下孵育24小时。执行此步骤时，必须以相等的比例添加四种逆转录病毒超级体，以确保所有四个转基因基因对重新编程过程的贡献相等。第二天，准备转导混合物，首先添加一卷病毒上流剂，每次重新编程因子，并辅以每毫升四微克的逆转录病毒感染试剂到六卷FEX介质。之后，从成纤维细胞培养中吸出FEX介质，加入转导混合物。在缺氧条件下，在5%的二氧化碳和4%的氧气下，在37摄氏度下孵育转导反应4小时。四小时后，吸气转导混合物，并加入新鲜的重新编程介质。在缺氧条件下孵育37摄氏度8天，每两天加入一次新的重新编程介质。观察从板分离出的细胞团成功重新编程。要从培养皿中收集重新编程的细胞以作进一步分析，请轻轻移液，然后上下移液，然后收集细胞。在成功重新编程 iEPs 后，YFP 阳性细胞早在转导后五天就可观察到。它们变成圆形，从板块表面抬起来，开始形成聚类，并表现出红体前体状形态。一些细胞的血红蛋白通过积极的本齐丁染色是明显的。一小部分表示红机器人特异性表面标记TER-119。到了第八天，可以看到大型 YFP 阳性聚类。第 8 天 iEPs 的红细胞表型比第 5 天 iEPs 更分化。它们明显更小，积累了更多的血红蛋白，并表现出TER-119的表达力增加。qPCR对第8天收集的eEPs的基因表达分析表明，它们几乎关闭了成纤维细胞基因的表达，并调节了许多红细胞基因。在对重新编程的细胞进行BFU-e菌落形成分析后，第8天第8天即聚体形成两种类型的菌落，明显为红色，而不是明显的红色。虽然来自红菌落的细胞表现出红细胞形态，但来自非红菌落的细胞则没有。大约每1000天有1个，5个eEP形成红色殖民地，而只有大约1万个殖民地形成第8天（第8天）的一个。尾尖成纤维细胞或TTF的重新编程效率受通道编号的影响。与经过三次的细胞相比，经过九次的 TTF 显示出产生 iEPs 集群的能力急剧下降。此外，不同的文化条件会影响重新编程的效率。在诺莫夏培养的转导 TTF 重新编程的速度要慢得多，IEP 集群在 10 天后而不是 5 到 8 天后观察到。按照这个程序，可以应用蜂群形成分析、qPCR和FACS分析等方法来确定细胞分化状态。自开发以来，这项技术为红细胞体领域的研究人员在理解小鼠和人类原始和确定性红细胞细胞之间的转换方面为人们铺路。
