De technologie van gen richten door HR in lymfkliertest ES-cellen zorgt voor een krachtig hulpmiddel voor het ontleden van de cellulaire gevolgen van specifieke genetische mutaties1,2. Het belang en de betekenis van deze technologie worden weerspiegeld in de erkenning ervan door 2007 de Nobelprijs voor de Fysiologie of Geneeskunde3,4; Ondertussen, het vertegenwoordigt de komst van het moderne tijdperk van gene engineering5. Gen richten via HR kan worden gebruikt om ingenieur vrijwel elke wijziging variërend van puntmutaties tot grote chromosomale herschikkingen in het genoom van muis ES cellen6,7. Het is algemeen bekend dat, vóór de opkomst van de zogenaamde genoom bewerkingsgereedschappen, de generatie van een gen knockout muis de toepassing van gentargeting technologie in ES cellen8,9,,10 vereist. Tijdens de afgelopen twee decennia, werden meer dan 5.000 gen-gerichte muizen geproduceerd door deze benadering voor het modelleren van ziekten bij de mens of studeren gene functies11. De inspanning van een genoom-brede knock-out is opgezet voor het distribueren van gentargeting vectoren, gerichte ES cel klonen en levende muizen naar de wetenschappelijke gemeenschap2,12,13,14 , 15. ongetwijfeld ES cel-gebaseerde HR-gemedieerde gentargeting technologie zich sterk ontwikkeld ons begrip van de functies van genen in fysiologische of pathologische verband gespeeld.
Omdat HR is een relatief zeldzame gebeurtenis in zoogdieren16,17, de belangrijke cellen en volgende stap na is gentargeting in lymfkliertest ES-cellen voor het analyseren van de talrijke kolonies van de ES voor het identificeren van een paar klonen met mutaties die voortvloeien uit HR met de targeting vector18. De gouden methoden voor het identificeren van HR gebeurtenissen omvatten Southern blotting en PCR19,20. De voordelen van de benaderingen omvatten dat Southern blotting correct gerichte ES klonen identificeren kunt en toelaat van onderzoekers voor het analyseren van de structuur van het gen-gerichte evenement, zoals een verificatie van de invoeging van een enkel exemplaar van de constructie, terwijl een PCR gebaseerde strategie vergunningen meer snelle screening voor HR gebeurtenissen21,22. Al deze methoden nadelen, zoals hebben dat ze tijdrovend en kunnen hebben valse positieven, de combinatorische gebruik van hen is breed geaccepteerd en toegepast door de meeste laboratoria in het identificeren van HR gebeurtenissen, vooral in ES-cellen, voor genereren van genetisch gemodificeerde dieren.
Drie isoforms van nonmuscle myosin II (NM II) in zoogdieren, elk bestaande uit twee identieke NMHC IIs die zijn gecodeerd door drie verschillende genen (met de naam Myh9, Myh10, en Myh14) en twee paren van lichte kettingen, worden aangeduid als NM II-A en II-B II-C23. Eerdere studies hebben aangegeven dat ten minste de isoforms van NM II-A en II-B zijn essentieel voor de ontwikkeling van de muis, omdat de in-vivo -ablatie van deze isoforms in embryonale letaliteit24,25,26 resulteert. Om dit probleem te omzeilen en het verkrijgen van nieuwe inzichten in de isovorm-specifieke functies van NM II-A en II-B in de latere stadia van de ontwikkeling van de muis, een genetische vervanging strategie met behulp van ES cel-gebaseerde HR-gemedieerde gentargeting technologie werd aangenomen het genereren van een serie van muis modellen27. In de loop van het identificeren van de gewenste ES klonen, zowel Southern blotting en PCR methodes werden gebruikt, en dit bleek te zijn efficiënt en betrouwbaar27,28.

identification of homologous recombination events in mouse embryonic stem cells using southern blotting and polymerase chain reaction

Ten opzichte van de kwesties van off-target effecten en de moeilijkheid van het invoegen van een lang fragment van DNA in de toepassing van ontwerper nucleasen voor genoom editing, embryonale stamcellen (ES) cel-gebaseerde gentargeting technologie beschikt niet over deze tekortkomingen en wordt op grote schaal gebruikt voor het wijzigen van dier/muis genoom variërend van grote verwijderingen/ingevoegde tot één nucleotide vervangingen. Met name, identificatie van de relatief weinig homologe recombinatie (HR) gebeurtenissen nodig zijn om de gewenste ES klonen is een belangrijke stap, die vraagt om accurate en betrouwbare methoden. Southern blotting en/of conventionele PCR worden vaak gebruikt voor dit doel. Hier beschrijven we de gedetailleerde procedures van die twee methoden gebruiken voor het identificeren van HR gebeurtenissen die hebben plaatsgevonden in muis ES cellen waarin het endogene Myh9-gen is bedoeld om te worden verstoord en vervangen door cDNAs codering van andere nonmuscle myosin zware ketting IIs (NMHC IIs). De hele procedure van Southern blotting omvat de bouw van targeting vector(s), electroporation, selectie van de drug, de uitbreiding en de opslag van ES cellen/klonen, de voorbereiding, de spijsvertering en van genomic DNA (gDNA), de kruising bevlekken en wassen van probe(s), en een laatste stap van autoradiografie op de X-ray films. PCR kan worden uitgevoerd rechtstreeks met bereid en verdunde gDNA. Ideaal om resultaten te verkrijgen, moeten de sondes en restrictie-enzym (her) snijden sites voor het Zuidelijke bevlekken en de inleidingen voor PCR zorgvuldig worden gepland. Hoewel de uitvoering van de Southern blotting is tijdrovend en arbeidsintensief en PCR-resultaten hebben valse positieven, de juiste identificatie door Southern blotting en de snelle screening door PCR de uitsluitende of gezamenlijke toepassing van deze methoden toestaan in deze paper wijd gebruikt en geraadpleegd te worden door de meeste laboratoria in de identificatie van genotypen van ES-cellen en genetisch gemodificeerde dieren.

Identification of Homologous Recombination Events in Mouse Embryonic Stem Cells Using Southern Blotting and Polymerase Chain Reaction

Hier presenteren we een gedetailleerd protocol voor het identificeren van homologe recombinatie gebeurtenissen die hebben plaatsgevonden in muis embryonale stamcellen met behulp van Southern blotting en/of PCR. Deze methode wordt geïllustreerd door de generatie van nonmuscle myosin II genetische vervanging Muismodellen met behulp van traditionele embryonale stamcel gebaseerde homologe recombinatie-gemedieerde targeting technologie.

Deze methode kan helpen bij het beantwoorden van belangrijke vragen in moleculaire en cellulaire biologie veld, zoals het identificeren van homologe recombinatie gebeurtenissen in de muis embryonale stamcellen. Het belangrijkste voordeel van deze techniek is dat het nauwkeurig, betrouwbaar en veel gebruikt is. Deze methode kan dus inzicht geven in het identificeren van homologe recombinatiegebeurtenissen in de embryonale stamcel van muizen.Het kan ook worden toegepast op andere systemen, zoals genetisch gemodificeerde cellen of dieren. Over het algemeen, individuele nieuw voor deze methode zal worstelen, omdat het lang duren en gaat om meerdere stappen. Voor elke gDNA, meng 30 microliter van 10 x buffer voor DRA een, drie microliters van DRA een en 10 microgram gDNA's per monster.Voeg water toe tot het uiteindelijke volume 30 microliters is en broed 's nachts uit tot 37 graden Celsius om de gDNAs te verteren. Bereid een 1%agarose elektroforese gel met ethidium bromide. Breng de gel vervolgens over in een lade met een DNA-neutraliserende oplossing. En schud het zachtjes gedurende 20 minuten bij kamertemperatuur.Breng met behulp van een snel neerwaarts overdrachtssysteem de NC's van de gel naar het membraan over. Monteer vervolgens de turble blotter en blotting stack zoals beschreven in de instructies van de fabrikant. Neem hierna het membraan eruit en was het met twee X zout natriumcitraat gedurende één minuut.Absorbeer de vloeistof met weefsels, en gebruik vervolgens een UV-crosslinker om het DNA te kruisen met het membraan. Om te beginnen met het labelen van de DNA-sondes met radioactiviteit, voeg de warmte gedenatureerde sonde DNAs aan de buis met de ready to go DNA-etikettering kralen. Pipette op en neer om te mengen.En voeg vijf microliters van radioactief gelabeld deoxycytodine triphosfate. Incubeer bij 37 graden celsius gedurende 15 minuten. Zuiver de gelabelde sondes met behulp van G 50 micro kolommen volgens de instructies van de fabrikant.Gebruik een scintillatieteller om de radioactiviteit te meten. Om te beginnen met het hybridiseren van de membranen met de gelabelde sondes, plaats het membraan in de hybridisatiebuis. Voeg de gemengde pre hybridisatieoplossing toe.Plaats de buis in hybridisatieoven en laat de pre hybridisatie 30 minuten op 42 graden Celsius gaan. Haal vervolgens de buis uit de oven en giet de pre-hybridisatieoplossing in een buis van 50 milliliter. Voeg de gedenatureerde sonde toe en meng voorzichtig.Om de niet-gehybridiseerde sondes te verwijderen, plaatst u het membraan in een lade met één X zoutoplossing natriumcitraat met 0,1%SDS en schud voorzichtig op 55 tot 60 graden Celsius gedurende 10 minuten. Verpak het membraan hierna met plastic folie en bevestig het in de belichtingscassette. In een donkere kamer, bloot het membraan aan twee vellen van x-ray film.Ontwikkel de film om de resultaten te visualiseren. Bepaal of een overeenkomstige ES-kloon de gewenste is met het doel van recombinatie of niet, afhankelijk van de grootte van de DNA-banden die door de sondes worden gedetecteerd. In deze studie, zuidelijke blotting en PCR wordt gebruikt om HR-gebeurtenissen die zich voordoen in de muis ES cellen voor de generatie van NM twee genetische vervangende muis modellen met behulp van ES cellen gebaseerd HR gemedieerde targeting technologie te identificeren.Omdat de genomische NIT's in vele fragmenten met verschillende lengtes zijn gesneden, vertonen ze een uitstrijkjeachtige status op de DNA-gel. Dit suggereert een volledige vertering van de genomische NC's. Als laatste stap van zuidelijke blotting, worden de signalen van een radioactiviteit gelabeld sonde hybridiseren met een doel-DNA-fragment worden getoond op de film.Het verschijnen van verwachte banden weerspiegelt het optreden van HR-gebeurtenissen in de ES-klonen. Volgens het voorontwerp in de studie, ES klonen met gemuteerd allel hebben twee verschillende grootte banden. Terwijl wilde type ES klonen hebben slechts een band, suggereert de gewenste ES klonen zijn heterozygoot.Na de PCR-reactie kunnen de PCR-producten worden geanalyseerd op de DNA-gel. Als een specifieke NPCR-band van de juiste grootte wordt waargenomen, suggereert dit het optreden van HR-gebeurtenissen die verder kunnen worden bevestigd door die PCR-band te sequencingen. De implicatie van deze techniek strekte zich uit tot de therapie van het identificeren van puntmutatie omdat de stichting identiek is.Tijdens het proberen van deze procedure is belangrijk om te onthouden om geduldig en voorzichtig te zijn. Na deze procedure kan andere methode zoals microinjectie van ES-cel in blastocysten worden uitgevoerd om aanvullende vragen te beantwoorden, zoals de vorming van chimerische dieren. Achter de ontwikkeling ervan maakte deze techniek de weg vrij voor de onderzoeker op hun gebied van moleculaire en cellulaire biologie om de functie van een specifiek gen of het gevolg van een gemuteerd gen bij muizen of daarbuiten te onderzoeken.Vergeet niet dat het werken met radioactiviteit labeler P32 DCTB kan zeer gevaarlijk zijn en voorzorgsmaatregelen zoals afscherming met bescherming boord moet altijd worden genomen tijdens het uitvoeren van deze procedure.
