获得有用的 iscat 数据的最关键的一个方面是能够在覆盖层表面找到正确的焦点位置, 此外, 还能够长时间保持这一位置。如果做不到这一点, 就会导致 psf 扩大, iscat 信号变弱, 动态分析中与漂移相关的伪影。事实证明, 在干净、裸露的盖板表面上找到焦面并不是一件容易的事情, 因为在大参考光束背景下看不到表面特征 (见图 4d)。

在该协议中, 我们描述了 iscat 的广域照明方案, 因为它快速、易于实现, 并允许在大面积14 上进行并行检测。另一种常见的方法是使用声光偏转器 (aod) 并扫描样品 12、17的共聚焦光束。这种方法避免了对高质量波前的需求, 但比传统的广域成像在实验上更为复杂。此外, 共聚焦照明的速度受到 aod 照明速度的限制。根据所需的实验参数, 原则上可以利用共聚焦或广域照明方案来检测活细胞分泌的单一蛋白质。
正如整个协议所讨论的, 必须最大限度地减少显微镜样品阶段的横向机械波动。即使是样品位置上的纳米偏差也会导致连续相机帧的变化, 并在差分图像中产生明显的外部噪声。因此, 建议在实验期间使用机械稳定的显微镜级和阻尼光学工作台 (步骤 1.1.1), 并在实验期间用光学窗帘或面板覆盖设置 (步骤 3.5)。
还可以考虑为长期测量制定积极的重点稳定计划。在这种方法中, 第二个激光被整合到显微镜中的一个总内部反射 (tir) 安排, 然后成像到一个象限光电二极管。系统焦距的变化转化为象限二极管上 tir 激光光斑的侧向位移, 然后可在有源反馈回路中使用该位移来控制压电第26阶段的 z 轴.因此, 消除了长期的垂直漂移效应。
