Indsamling er afgørende i dyreforsøg. Target venerne for blod samling omfatter retro-orbital plexus, halsfedt, den saphenous vene, hale blodkar og halspulsåren1,2,3,4. Nogle gange, kunne blod indhentes fra den abdominale aorta, vena cava eller endda hjertet5,6,7. Ved sådanne lejligheder, dyrene skal ofres og kan ikke bruges til efterfølgende observationsperiode således, disse metoder der bruges mindre i daglige eksperimentelt arbejde. Indsamling fra hale vene af rotter er populære og mindre stressende i forhold til de ovenfor nævnte metoder8.
Men, blod samling fra hale vene er undertiden begrænset af en utilfredsstillende succesrate. Lejlighedsvis, er det også svært at få nok blodvolumen ved denne metode. Som subclavia vene er stor nok og er placeret lige under kravebenet knogle, kan subclavia vene punktering være en alternativ metode til blod prøveudtagning hvis rutinemæssig blod samling metoder er mislykket. Her, indfører vi en metode til indsamling af subclavia vene punktering i rotter.

Blod samling med nok blodvolumen er afgørende i dyreforsøg. Indsamling fra hale vene af rotter er populære og mindre stressende sammenlignet med andre mere aggressive metoder såsom retro-orbital plexus prøve samling. Men, dette blod indsamlingsmetoden er undertiden begrænset af en utilfredsstillende succesrate. Her, vil vi præsentere en metode for indsamling gennem subclavia vene punktering. Subclavia vene er placeret lige under kravebenet og denne vene er stor nok til at opfylde kravene volumen af blod samling. Vores resultater viser, at denne metode er sikker og gældende for blod stikprøvetagning med den nødvendige blodvolumen. Blod samling gennem subclavia vene punktering kunne tjene som en alternativ blod indsamlingsmetoden i tilfælde af fejlslagne hale vene blod prøveudtagning i rotter.

Ialt 20 SD rotter (mandlige n = 10, kvindelige n = 10) blev brugt her. Blod collectionwas udføres af kvalificerede operatører, som har udført mere end 20 blod prøveudtagninger fra subclavia vene i rotter, og driftslederne nybegynder, der har ingen subclavia vene punktering erfaring med rotter eller andre dyr. Før operationer set begyndere mindst 3 procedurer gjort af operatorer.

Subclavian Vene Punktering som en alternativ metode til blodprøvetagning i rotter. Efter generel anæstesi, placere rotter i liggende stilling og sterilisere infraklavicular plads med 75% ethanol. Identificer overlegen brystfosse før punktering.Find punkteringsstedet, caudal til den ydre tredjedel af kravebenet i samme side. Retningen af nålen var placeret mod den overlegne brystvejr fossa og bør næsten parallelt med kravebenet, bare posteriort til det. Efter blodtapning skal du trykke på punkteringsstedet i et til to minutter for at stoppe blødningen.Blodtapning med tilstrækkelig volumen er altid nødvendig i dyreforsøg. Blodtapning fra rats'tail vene er populær og mindre stressende end nogle aggressive metoder såsom retro orbital plexus prøvetagning. Denne metode er dog begrænset af succesraten.Her introducerer vi vores metode til blodtapning med den ønskede blodvolumen fra den subclavia vene. Vores resultater viser, at denne metode er sikker og praktisk, og kan overvejes i tilfælde blodprøvetagning fra hale vene er mislykkedes. For at starte, forberede tape, epileringsmiddel, 75% ethanol, blodopsamlingsrør, to punkt fem milliliter sprøjte, hår barbermaskine, elektronisk skala, heparin, og fysiologisk saltvand.Husrotter under standardforhold, med fri adgang til mad og drikkevand, opbevares i 530 kvadratcentimeter bure med træbarbering. Fremkalde generel anæstesi hos alle dyr gennem intraperitoneal injektion før punktering. Placer rotten i liggende stilling, fastgør baglestremerne i en behagelig position, og fastgør de overlemmer parallelt med kroppen, lige ved siden af stammen.Rengør begge sider af det infrarøde rum for at fjerne pelsen og eventuelt synligt snavs. Tør hals og thorax hud med 75% ethanol, og holde området rent og tørt med gaze. Begge sider af den subclavia vene er velegnede til punktering, og her vælger vi den rigtige side til punktering.Identificer placeringen og retningen af højre kraveben med operatørens venstre tommelfinger, og find den overlegne brystfosse forsigtigt med pegefingeren. Skyl vores sprøjte med 10 enhed pr. milliliter heparinopløsning. Find punkteringsstedet, punkt fem millimeter hale til den ydre tredjedel af højre kraveben.Flyt sprøjten forsigtigt ind i huden på punkteringsstedet. Når nålen kommer ind i den infrarøde hud, skal der trykkes aktivt gennem operatørens højre hånd. Retningen af nålen er placeret mod den overlegne brystvejr fossa, og bør næsten parallelt med kravebenet, bare posteriort til det.Normalt kunne den subclavia vene nås ved at indsætte nålen i omkring to millimeter i denne retning. Når nålen kommer ind i venen, vil blodet komme ind i sprøjten under et aktivt tryk. Derefter skal det aktive tryk opretholdes, indtil der trækkes en korrekt mængde blod.Efter blodtapning fjernes punktålen, og punkteringsstedet trykkes i et til to minutter for at stoppe blødningen. Vær opmærksom: Trykket skal være meget skånsomt for at forhindre choker. Blodprøven overføres til et antikoaguleret vakuumrør med EDTA og centrifuge ved 1600G i 10 minutter for at indsamle plasma.Aspirate supernatent at re-clean rør og opbevares på minus 18 Celsius grader. For at sammenligne succesraten mellem de dygtige operatører og begyndere, 20 SD rotter blev opdelt i to grupper tilfældigt. Resultaterne viser, at gennem den rigtige subclavia vene, ni rotter i den dygtige gruppe og otte rotter i begyndergruppen lykkedes, henholdsvis.Så blodtapning fra de resterende tre rotter lykkedes gennem venstre side. Succesraten havde ingen signifikant forskel mellem disse to grupper. Tidsforløbet var lidt anderledes i disse to grupper.Ingen alvorlige komplikationer, såsom pneumothorax, hemopneumothorax, og blødning opstod under eller efter operationen. Alle dyr overlevede efter blodprøvetagningen. For at vise vanskelighederne ved at lære punkturproceduren sammenlignede vi yderligere antallet af tilbagekøb mellem de dygtige og begyndergrupperne.Som det fremgår af tabel to, er det gennemsnitlige antal udbetalinger i faglært gruppe lidt mindre end begyndere, men der er ingen statistisk signifikans. Afslutningsvis viser vores resultater, at den subclavia vene punktering strategi for blodtapning er sikkert og muligt hos rotter. Denne metode er værdifuld, især i tilfælde, hvor store mængder blod er nødvendig.Denne metode kan være et optimalt alternativ, hvis den konventionelle blodprøvetagning mislykkes.
