この方法は、遺伝子候補、ウイルス、または血液精巣バリア機能または完全性に影響を与える可能性のある環境毒素に関する生物学分野の血液検査障壁の主要な質問に答えるのに役立ちます。手術の3日前に、少なくとも3匹の8週齢のC57Black/6雄マウスを、腹腔内注射を介して1キログラムの体重1キログラム当たり5ミリグラムの塩化カドミウムで治療する。手術当日、75%エタノール殺菌手術領域の上にきれいな組織を置き、サーモスタットヒーターを摂氏37度に設定します。麻酔をしたマウスを組織に置き、つま先ピンチに対する反応の欠如を確認する。腹部の毛を剃り、75%エタノールで露出した皮膚を消毒する。腹壁を露出させるために前置腺の上に1センチメートルの皮膚切開を行い、腹膜に0.5センチメートルの切開を行うために皮膚を持ち上げるために小さな鉗子を使用します。鉗子を使用して、精巣上体と精巣の周りの脂肪パッドを検索し、慎重に脂肪パッドを引き出して1つの付着精巣を露出させます。脂肪パッドと精巣の下に9センチメートルの紙を置き、新たに調製したイヌリン-FITC溶液でマイクロインジェクションピペットをロードします。ピペットをマイクロマニピュレーターユニットに接続し、解剖顕微鏡を使用して、チュニカアルブギネアの下にマイクロインジェクションピペットチップをそっと挿入します。手術レバーをゆっくりと回転させて、20マイクロリットルのイヌリン-FITCを精巣のインタースティシウムに送ります。配信が成功した場合、精巣は明るい緑色と黄色に変わります。すぐに精巣を腹腔に戻し、20マイクロリットルのPBSで対側精巣を注入し、外科縫合糸で皮膚を閉じる。そして、サーモスタットヒーターのヒートパッドにマウスを置きます。注射の40分後、鋭いはさみを使用して各マウスから精巣を採取し、1ミリリットルの氷冷PBSで組織を洗浄する。次に、1回の洗浄につき1.5ミリリットルで3回の洗浄を行い、摂氏4度で4%パラホルムアルデヒドで12〜24時間精巣を固定します。一晩で30%スクロース脱水後、個々の埋め込みフレームにサンプルを移し、最適な切断温度化合物で組織を覆います。クライオスタットの温度をマイナス20°Cに設定し、OCTが凍結するまで約10分間クライオスタットにフレームを入れます。次に、各鋳片全体が各金型で覆われるまで、各埋め込みフレームに新しいOCTを充填します。OCTの第2層が凍結すると、各精巣の厚さ5マイクロメートルの断面を取得し、得られるガラス顕微鏡スライド上に組織切片を捕捉する。サンプルを画像化するには、まず室温で加湿ボックスでスライドを温めます。10分後、スライドを洗浄ごとに新鮮なトリスバッファ付生理液で3回洗浄し、セクションを光から保護して5分間乾燥させます。ダストフリーの紙を使用して残留生理的な生理を取り除き、サンプルをDAPIを補った取り付け媒体で取り付けます。次に、各サンプルを反転カバースリップで覆い、共焦点顕微鏡の20X目的の下でサンプルを画像化します。3日間の塩化カドミウム治療は、血液精巣バリアに損傷を与え、イヌリン-FITCが基底コンパートメントから半日本体上皮の尖体コンパートメントへの拡散を可能にする。対照的に、この拡散は、PBS処理精巣の制御で遮断される。血液検査バリア損傷の程度は、同じ半細管の半径に対するイヌリン-FITC拡散の距離の比を計算することによって決定される。さらに、ヘテロ接合性Rictor floxdノックアウトマウスの無傷の血液検査障壁は、障壁を越えて上端コンパートメントへのイヌリン拡散をブロックし、条件付きRictorノックアウトマウスはインリン拡散を可能にする侵害された血液検査障壁を有する。この手順を試みる間、実験当日に準備されたイヌリン-FITCを提供し、FITCは光感受性分子であるため、組織を光から保護しておくことが重要です。この技術の意味は、血液検査障壁の機能が精子形成におけるmeiosisの完了のための免疫特権的微小環境を提供することであるため、精子形成障害の治療に及ぶ。その開発後、この技術は、生殖医学の分野の研究者がマウスの不妊症と不妊症を探求する方法を提供します。パラホルムアルデヒドを使用することは非常に危険であり、この手順を実行する際には衣服やフェイスマスクを着用するなどの予防措置を常に取るべきであることを忘れないでください。
