</ol>3. 膜分数的分离和溶解

</ol>4. 免疫沉淀

植物依靠叶绿体进行光合作用和光自养生长1。绝大多数叶绿体蛋白被编码在细胞核中, 在细胞溶胶中合成, 并通过包膜中的 tic 和 toc 复合物导入叶绿体2。toc 复合体的核心包括 toc75 蛋白传导通道和两个受体 gtpase Toc75 和 Toc753,4,5。目录159是叶绿体的生物发生所必需的, 并介导光合作用相关蛋白的大量积累.tic 综合体由 tic20 蛋白传导通道以及 tic110 和 tic407、8组成。最近, 一个1md 蛋白移位复合物在内包膜被隔离, 并包含以前未被识别的成分9, 其中之一是 tic56。我们最近使用 tap-toc159 亲和力纯化协议共分离了 1个 mda 复合体中的 tic56。数据表明, "规范" toc函配合物与 1 mda 复合物10之间存在结构重叠。以前未知的组件, 如 koc1, 也被确定为新的交互伙伴的 toc 和 tic 复合体使用这里描述的 tap 方法 10,11.
因此, tap-标记纯化是一种有效的方法, 用于分离蛋白质复合物和通过随后的质谱分析12 识别相互作用的伙伴。tap 标记由两个 igg 结合重复从钙调素结合肽 (cbp) 分离的烟草蚀刻病毒 (tev) 蛋白酶裂解位点。最初的方法, 包括 igg 亲和力纯化步骤, 其次是 tev 裂解和随后的钙调联亲和层析, 允许本地纯化大型和高度纯蛋白质复合物13,14.我们简化了程序, 并证明, 含有 toc159 的蛋白质复合物也可以有效地纯化, 只使用 igg 亲和力纯化步骤, 然后 tev 裂解洗脱15。根据我们的经验, 钙调素亲和力步骤的遗漏导致了更高的产量, 因此可能适合于低丰度蛋白质。

affinity purification of chloroplast translocon protein complexes using the tap tag
