以前は、ケージ NLS7,8,9を使用して、細胞の核蛋白質の光トリガー転流を実現しています。NLS は、6細胞質輸送タンパク質に結合して細胞核に移行します。報告の方法でケージの NLS を軸受ゲスト蛋白質はマイクロインジェクション8で細胞質に組み込まれてまたは遺伝コードの拡張手法9を使用してターゲット細胞で発現直接。したがって、細胞内取り込みと光誘起核移行を実現できる手法は実用的なアプリケーションのために有利です。

この方法は、特定の細胞粒状に薬物を送達する方法など、薬物送達の分野における重要な質問に答えるのに役立ちます。この技術の主な利点は、ガスの核転位が光によって誘導され得る点である。この手順を実演するのは、私のグループの卒業生であるモガキ・リナと有坂昭夫です。他の場所でより詳細に記載されるようにケージ状の分子接着剤を結合したジベンゾシクロオクチンを合成して量子ドット連結ケージ分子接着剤の調製を開始する。乾燥したDMSOでジベンゾシクロオクチンに結合したケージ分子接着剤の10ミリモルのストック溶液を調製する。次に、まずアスアイを機能化した量子ドットを作ることによって、量子ドット連結ケージ分子接着剤を調製する。これを達成するために、アミン機能化ペグでコーティングされた量子ドットの500ナノモルを含むDMF溶液の400マイクロモルaseye PEG4 NHSエステルの100マイクロモルaseye PEG4のエステルを加えます。室温で1時間かき混ぜます。得られた溶液を再生セルロース膜に入れ、800mlのDMFに対して24時間透析します。次に、ジベンゾシクロオクティンをDMFで50マイクロモルに結合したケージ分子接着剤のストック溶液を希釈し、その200マイクロリットルを透析後溶液に加える。混合物を室温で3時間かき混ぜます。次に、得られた混合物を、25000の分子量を遮断した再生セルロース膜を用いて800mlの新鮮なDMFに対して24時間透析する。24時間後に得られた溶液をDMFで200ナノモルに希釈した。ヒト肝細胞癌HEP3 B細胞を、標準的な培養条件下で10%FBSを含むイーグルの最小必須培地で維持します。実験の1日前に、培養培地の200マイクロリットルの5000HEP3 B細胞を8チャンバーガラススライドの各ウェルに入れた。その後、スライドを覆い、24時間インキュベーターに入れます。翌日、培養培地を取り除き、100マイクロリットルの予温PBSで細胞を2回リンスします。8チャンバースライドの各ウェルに、蛍光染色ケージ分子接着剤の10マイクロモルを含むFBSフリー培地200マイクロリットルを加えます。3時間培養後、培養培地を取り出し、100マイクロリットルのPBSで細胞試料を2回リンスする。Endoソームを視覚化するには、LysoTracker Redなどの追加の赤色蛍光色素の100ナノモルを含む培養培地の200マイクロリットルを追加します。得られた細胞サンプルを20分間インキュベートする。その後、培養液を取り出し、100マイクロリットルのPBSで細胞試料を2回リンスする。その後、細胞を200マイクロリットルの培養培地に戻す。蛍光染色ケージ分子接着剤の核転位を誘導するために、細胞を365ナノメートルのバンドパスフィルタを装備した100ワットのキセノン光源の下に置く。細胞をUV光に2分間照射します。この間、コントロールセルサンプルは、暗闇の中で紫外線を曝させずに保ちます。ガラス基板の蓋は効率的な紫外線暴露のために取り除くことができますが、UV光への長時間の暴露は細胞死を引き起こす可能性があるため注意してください。核を可視化するために、培養液に1マイクロリットルのホーシュスト染色を加える。得られた細胞試料を10分間インキュベートする。次に、サンプルを共焦点レーザー走査顕微鏡のステージに置きます。蛍光染色ケージ分子接着剤、LysoTracker Red、および核染色の顕微鏡写真を記録します。先に示したように、種子5000ヒト肝細胞癌HEP3B細胞を、標準条件下で8ウェルチャンバースライド及び培養の各ウェルに24時間用いた。蛍光染色ケージ分子接着剤を含有するFBSフリー培養培地200マイクロリットルを細胞試料に供給する。得られた細胞試料を3時間インキュベートする。その後、培養液を取り出し、100マイクロリットルのPBSで細胞試料を2回リンスする。培養培地を200マイクロリットルに戻す。次に、サンプルの共焦点レーザー走査顕微鏡を受け、付随するテキストプロトコルに記載されているように顕微鏡写真を記録します。先に示したように、種子5000ヒト肝細胞癌HEP3 B細胞は、標準条件下で8ウェルチャンバースライドおよび培養の各ウェルに24時間用いた。量子ドット連結ケージ分子接着剤の10ナノモルを含むFBSフリー培養培地の200マイクロリットルを細胞サンプルに供給する。得られた細胞試料を3時間インキュベートする。その後、培地を取り出し、100マイクロリットルのPBSで細胞サンプルを2回リンスします。細胞をすすった後、培養培地の200マイクロリットルにそれらを返す。その後、365ナノメートルのバンドパスフィルターを装備した100ワットのキセノン光源の下にプレートを置きます。細胞サンプルをUV光に2分間照射し、コントロールサンプルを暗く保ちます。核を可視化するために、培養液に1マイクロリットルのHoechst染色を加える。得られた細胞試料を10分間インキュベートする。次に、共焦点レーザー走査顕微鏡を使用して細胞を画像化します。実験の前日にヒト肝細胞癌HEP3 B細胞を、1ウェルあたり5000個の細胞に96個のウェル培養プレートに播種します。その後、培養培地の200マイクロリットルで細胞をカバーし、24時間インキュベーターにプレートを置きます。翌日、培養培地を取り出し、100マイクロリットルのPBSで細胞サンプルを2回リンスする。次に、0.1~100マイクロモルの範囲の濃度で蛍光染色ケージ分子接着剤を含むFBSフリー培養培地を200マイクロリットル加えます。プレートを3時間インキュベートし、UV光源の下にプレートを置きます。細胞サンプルをUV光に2分間照射し、コントロールサンプルを暗く保ちます。培養液に対する細胞計数キット-8試薬の10マイクロリットルでのUV露光後、得られた細胞試料を2時間インキュベートする。次にマイクロプレートリーダーを用いて450ナノメートルでサンプルの吸収を読み取る。蛍光染色ケージ分子接着剤でインキュベートされた光放射HEP3 B細胞の前に、内部からの穿刺蛍光放出を示すLysoTracker Redを示す類似の顕微鏡写真は、蛍光染色ケージ分子接着剤が内因性に局在したことを示す。したがって、蛍光染色ケージ分子接着剤に割り当て可能な蛍光発光は、UV照射前に細胞核の外で観察された。紫外線放射後、蛍光体は、蛍光染色ケージ分子接着剤が欠取され、細胞質および細胞核に移行したことを示唆している。このような蛍光染色ケージ分子接着剤の核翻訳は、2光子近赤外光により選択的に部位を誘導することができる。非照射領域における蛍光染色ケージ分子接着剤は、内欠から逃れらず、蛍光穿刺として残った。樹状ケージ分子接着剤タグは、量子ドットなどの高分子ゲストを細胞核に送り込むこともあります。これらのコンジュゲートは細胞内に取り込まれ、UV曝露後2分間、量子ドットの蛍光放出が核内に見える。この開発の後、この技術は、遺伝子治療の分野の研究者が低い副作用治療を探求する道を開いた。
