そこで、私たちの新しい技術の主な利点は、濃縮と拡張を表すEプラスEと呼び、これらの細胞、まれな抗原特異的細胞を同時に豊かにし、拡大できることです。これにより、さまざまな設定や異なる臨床現場での役割を調査したり、免疫療法に利用できる多数の細胞を以前には不可能な方法で開発したりするのに十分な数の細胞を開発することができます。我々が開発した方法、このEプラスEは、ネオエピトープの存在に関する基本的な免疫学の重要な質問に答えるのに役立ちます。ネオエピトープとは何ですか?これらは、がん免疫療法の面で非常に重要であり、抗原特異的ダイナミクスの面でも、これらの要因が疾患状態と効果的な細胞療法にどのような影響を与えるかを知っているパーソナライズされたエピトープです。ナノ粒子人工抗原提示細胞は、個々の患者の免疫応答に合わせて容易に調整される既製の製品である可能性があるので、この技術の意味は免疫療法にまで及ぶ。ナノ粒子人工抗原提示細胞を生成するために、凍結乾燥した粒子を1ミリリットルの再懸濁液バッファーで再懸濁し、凝集体が見えないまで少なくとも15分間、懸濁液を激しく渦液にする。タンパク質との反応前に十分に渦を起こし、再懸濁されていない粒子は、解答することができないより大きな凝集粒子をもたらす。磁気スタンドに磁性粒子を置いて上清を取り除き、0.5ミリリットルのフレッシュリサスペンションバッファーで再懸濁します。凝集体が見えなくなるまで渦を起こすためにガラスシンチレーションバイアルに粒子懸濁液を移し、再懸濁粒子の1ミリグラム当たり0.1ミリグラムの総刺激信号を加える。ボルテックス後、懸濁液をローテーターで混合して室温で2.5時間反応させ、続いて0.1ミリリットルのクエンチバッファーを加え、混合して室温でさらに30分間インキュベーションします。2回目の反応の終わりに、シンチレーションバイアルを磁気スタンドに置き、1ミリリットルのリサスペンションバッファーで粒子を4回洗浄し、洗浄後に透明になったときに緩衝液を取り除き、その後の磁石で洗浄するためのフレッシュリサスペンションバッファに置き換えます。最後の洗浄後、溶液を吸引し、マグネットスタンドから粒子を取り除き、その後、最大6ヶ月間摂氏4度で新鮮な再懸濁液バッファーの1ミリリットルに粒子を保管します。抗原特異的CD8+T細胞を単離するには、野生のC57型黒6Jマウスの脾臓およびリンパ節を無菌、70マイクロメートルの細胞ストレーナーを介して頻繁にPBSリンスを介して浸食し、メーカーの指示に従って非CD8+T細胞分離キットを使用して非CD8+T細胞を排除します。カウント後、分離によって単離されたCD8+T細胞を収集し、0.5%牛血清アルブミンと2ミリモルEDTAを補ったPBSの100マイクロリットルのペレットを再中断し、ナノ粒子人工抗原提示細胞で細胞をインキュベートし、10回から11番目のペプチドをロードしたMHC-ig/、5ミリリットルポリスチレン丸底チューブを摂氏4度で1時間、継続的な混合を行う。インキュベーションの終わりに、示されているように、ナノ粒子細胞懸濁液を磁気スタンドで3回洗浄し、最後の洗浄後に1%T細胞増殖因子を有する新鮮な500マイクロリットルの人工抗原提示細胞および濃縮CD8+T細胞を再懸濁する。最大の細胞回収のためには、粒子と細胞を磁柱に少なくとも2分間放置し、磁場の影響を受ける粒子や細胞を破壊することなく、慎重にバッファを吸引することが重要です。カウント後、種子2.5倍の10倍から5番目の濃縮CD8+T細胞と96ウェルU底板に1%TCGFを加えた添加培地の160マイクロリットル当たりの磁性粒子混合物。3日目に、ウェルあたり2%T細胞増殖因子を含む80マイクロリットルの補充培地で細胞を供給し、さらに4日間、プレートを細胞培養インキュベーターに戻します。7日目には、刺激された細胞を5ミリリットルの丸底チューブに収穫し、遠心分離によって細胞を収集します。0.05%のアジ化ナトリウムと2%のウシ血清を加えたPBSの0.5ミリリットルでペレットを再懸濁し、5番目から5倍の5倍の10〜4〜5倍の5番目の細胞を5倍の5ミリリットルの丸底チューブに再懸濁して、抗原特異的染色用の新しい5ミリリットルの丸底チューブにします。バイオチン化MHC-igと抗マウスCD8aで適切なチューブに4°Cで1時間ラベルを付け、 続いて新鮮なPBSで遠心分離機を洗浄して過剰なビオチン化免疫グロブリンを除去し、適切なストレプトアビジン共役二次抗体と適切な生死性固定死細胞株を摂氏4度で15分間染色し、フローサイトメーターで細胞を読み取り、抗原特異的CD+Tプロトコルの特異性と数を決定する。ここで成功したタンパク質結合は、粒子へのタンパク質結合の3つの異なる方法を用いて達成された。AAPCリガンド密度が低すぎると、抗原特異的CD8+T細胞の有効な刺激が得られない。ビオチン化二量体の品質管理は、トランスジェニックCD8+T細胞で完了し、染色を検証することができます。例えば、これらの代表的な結果は、非抗原特異的b6 CD8+T細胞をバックグラウンドコントロールとして有するgp100特異的CD8+T細胞を有する陽性染色を示す。また、ビオチン化二量体の背景染色に関する知識は、このコグネイト染色チューブにおける染色よりも低い任意の割合が否定的な結果であると考えるべきであるため、重要である。7日間の濃縮および拡張の後、2回の間の10〜4番目と2回の間の第5の抗原特異的および5〜50%CD8+T細胞は、元の5倍の10から6番目の内因性CD8+T細胞開始集団から期待することができる。同じ方法論を使用して、富化後わずか1週間後に観察された抗原特異的CD8+T細胞の割合と数の割合と数が同様の増加を伴うヒト抗原特異的CD8+T細胞を単離し、刺激することができます。この手順を試みる際には、記載されているとおりに適切な試薬品質チェックを行うことを忘れないでください。この手順に従って、細胞内サイトカイン染色または表面タンパク質発現のためのフローサイトメトリーのような他の方法は、細胞の機能性または表現型に関する追加の質問に答えるために行うことができる。この技術は、基本的な免疫学分野の研究者が、がん、感染症、自己免疫研究などの他の生物医学分野における抗原特異的応答の広さ、深さ、ダイナミクスを探求する道を開きます。
