この方法は、肺内の炎症性遺伝子を調節すると予測されるマイクロRNAの発現を評価する方法に関する肺免疫分野の重要な質問に答えるのに役立ちます。この技術は、肺マイクロRNA発現に対する循環ホルモンの寄与を特定するための簡単な方法を提供する。エストルースサイクルステージを評価するには、8〜9週齢の雌マウスを手動で拘束し、超純水で満たされたプラスチック10マイクロリットルピペットの先端を膣に導入する。サンプルを収集するために4〜5回静かに洗い流し、膣液の最終的なフラッシュをガラススライドに堆積させます。その後、20倍の倍率で、未染色膣フラッシュを光顕微鏡で観察します。オゾンおよび濾過空気露出の場合、最大4匹のマウスを、金網の蓋付きの2つの個々の1.2リットルガラス容器の1つに入れます。オゾンチャンバーに1つのガラス容器を入れ、1つはろ過空気暴露室に入れます。その後、オゾン濃度を100万分の2に調整し、3時間後に容器を取り除きます。暴露の4時間後、第1実験マウスの露出した皮膚表面を70%エタノールで消毒し、肋骨ケージを切り取って心臓と肺を露出させる。鉗子を使用して、液体窒素に1.5ミリリットルRNaseフリーのマイクロ遠心分離チューブを浸水させ、収穫された肺を液体窒素中で凍結させます。次に、ステンレス鋼の組織粉砕機を使用して肺全体をマスチメントし、粉砕された組織を2つの1.5ミリリットルのマイクロ遠心分離管の間で分割します。この同じ方法ですべての動物から肺を採取して粉砕した後、各サンプルに500マイクロリットルのグアニジニウム・チオシアネートを加え、各組織を18、21、および23ゲージの針で順次均質化する。最後の均質化の後、15秒間渦を出す前に、各サンプルに500マイクロリットルのエタノールを加えます。遠心分離のために個々のコレクションチューブの個々のスピン列に混合物をロードし、フロースルーを廃棄します。DNase 1処理の場合は、各カラムに400マイクロリットルのRNA洗浄バッファーを追加して別の遠心分離を行い、RNaseフリーチューブのサンプルあたり5マイクロリットルのDNase 1と75マイクロリットルのDNA消化バッファーを混合します。室温で15分間のインキュベーションのためにカラムマトリックスに直接ミックスを加え、続いて2つの400マイクロリットルRNAプリウォッシュ溶液を遠心分離によって洗浄します。RNAを溶出するには、DNase RNaseフリー水を35マイクロリットルのDNaseフリー水を各カラムマトリックスに直接加え、最終的な遠心分離を行い、分光光度計上の各サンプルの全RNA濃度と純度を測定します。その後、RNAをマイナス80°Cで保存します。小さなRNaseの転写の場合、1サンプルあたり逆転写反応ミックスの1つのチューブに10マイクロウォッシュ溶液の総RNAの200ナノグラムを加えます。混合後、サンプルを短く遠心分離し、インキュベーションまで氷の上に置きます。次に、チューブを摂氏37度で60分間インキュベートし、続いて摂氏95度で5分間のインキュベーションを行います。2回目のインキュベーションの最後に、20マイクロリットル逆転転写反応あたり200マイクロリットルのRNaseフリー水でCDNAを希釈し、プリロードされたマウス炎症反応と自己免疫マイクロRNA PCRアレイの各ウェルに各反応ミックスサンプルの10マイクロリットルを加えます。プロエストラスステージは、円形および良好に形成された有核上皮細胞の存在によって同定することができる。マウスがエロス期にある場合、スミアでは、核分裂、角質化、扁平上皮細胞の密詰めクラスターが観察される。メ精巣の間に、角膜上皮細胞および多形核白血球が見られる。ダイストルでは、白血球は一般的により一般的である。4つのマウス肺から抽出されたRNAは、実証されているように、1マイクロリットル当たり1198〜2178ナノグラムの核酸濃度と、2.01〜2.02の間の平均A260〜A280比、および2.139〜2.223の間の平均A260〜A230比を示す。この表では、オゾンと濾過された空気暴露マウスとの間のマイクロRNA発現の2倍の変化を示す。マイクロRNA標的フィルターおよびコア分析後、14マイクロRNAについて、有意な発現対数比およびP値を得るために、これらのデータは、マイクロRNA標的フィルターによってマッピングすることができる。コア分析は、正規経路、疾患および機能、調節装置、およびネットワークに関する情報も提供します。また、機能解析ソフトウェアは、目的のマイクロRNAと他の分子との関係を示すネットワーク解析を生成するために使用することができる。この手順を試みる間、実験を行う前に、少なくとも3つの連続したサイクルについて、毎日の発情周期をチェックすることが重要です。この手順に従って、他の方法は、発情周期全体の肺炎症性遺伝子発現に関する追加の質問に答えるために行うことができる。開発後、この技術は、肺免疫の分野の他の研究者が他のモデルを使用して性ホルモン調節のメカニズムを探求する道を開いた。
