この方法は、断食や肥満などの異なる代謝条件で細胞間相互作用と脂肪組織生理学がどのように変化するかなど、脂肪生物学分野の重要な質問に答えるのに役立ちます。この技術の主な利点は、元の脂肪3D構造を最適に保存し、通常、従来の免疫組織化学に必要な長い処理ステップを回避することです。この方法は脂肪組織の構造に関する洞察を提供することができるが、脳全体のマウント染色によって神経系などの他の器官系にも適用することができる。一般的に、この方法に新しい個人は、特性組織部位を得ること、適切な固定期間を見つける、および最適な抗体濃度を見つけるのが困難のために苦労します。解剖が完了した後、1%パラホルムアルデヒド中の組織サンプルを含む皿を氷から室温に1時間移す。次に、組織を12または24ウェルの細胞培養プレートに移し、より迅速に洗浄します。マウント全体の染色手順は、解剖直後に開始する必要があり、各ステップ間に一時停止ポイントがないことを覚えておくことが重要です。PBS-0.3Tで組織を22度傾けたシェーカーで、室温で毎分20〜25回の傾きで5分間洗浄します。この洗浄を2回繰り返します。この後、5%動物血清を含むブロッキングバッファーの約0.5を1ミリリットルに加えます。前の条件を使用してシェーカーのプレートを1時間インキュベートします。ブロッキング溶液を吸引し、PBS-0.3Tで希釈した一次抗体を1%動物血清ずつ1%の一次抗体を各ウェルに加える。22度の傾きと毎分20〜25の傾きの4°Cのシェーカーに一晩プレートをインキュベートします。翌日、PBS-0.3Tを使用して、室温で5分間洗浄します。この洗浄を2回繰り返します。次いで、適切な希釈二次抗体溶液をそれぞれウェルに0.5ミリリットルから1ミリリットルの間に加えます。プレートをアルミホイルで包み、室温でシェーカーに1時間インキュベートします。この後、PBS-0.3Tを室温で2回洗浄して、1回5分間洗浄します。中性脂質染色時に液滴の可視化が必要な場合は、1回の洗浄で1回PBSで2回洗浄し、5分間持続します。PBSで希釈した中性脂質染色を加え、室温で30分間インキュベートする。イメージングを開始するには、鉗子を使用して、24 x 60ミリメートルのガラスカバースリップに組織を平らに置きます。DAPIによる核染色が望ましい場合は、DAPIを含む取り付け培地を1~2滴加えて組織を完全に水没させ、乾燥を防ぎます。次に、反転した共焦点レーザー顕微鏡システム上にスライドを置きます。複数の焦点面で全マウント染色された組織の画像を取得するには、所望の倍率で4〜6マイクロメートルのステップサイズで深さ100〜150マイクロメートルのZスタックを実行します。本研究では、脂肪組織アーキテクチャを維持するために、全マウント染色が使用される。脂肪組織形態の変形につながる複数の処理ステップおよび断面を伴う方法とは異なり、全マウント染色法は脂肪細胞の形態を保存することができ、結果を解釈する際の精度を確保する。脂肪組織の過固定は固定性誘発蛍光を導き、これはチロシンヒドロキシラーゼおよびPECAM-1シグナルのような同一の領域を重ねることによって示すことができる。しかし、適切に固定された全マウント染色サンプルは、個別の異なる信号を示し、これらの信号が自己蛍光ではないことを示す。全マウント染色された脂肪組織をイメージングすると、異なる実験条件下で形態変化を定量化することができます。特に脂肪組織は、エネルギーレベルの急激な変化時に代謝性恒常性を媒介する上で重要である、高血管化される。例えば、C57黒6Jマウスは、24時間の断食を受けるが、脂肪分解を示す有意に小さい脂肪細胞サイズを示し、連続的に供給されたマウスと比較して血管密度の上昇傾向を示す。iDISCO+で脂肪組織をクリアすると、脂肪組織が光学的に透明になります。組織クリアを経た脂肪組織の免疫標識は、同じ倍率で組織をクリアすることなく、全体のマウント染色で観察されたものと比較して、はるかに密度の高い神経耕動を示す。この手順を試みる間、PFAの室温で1時間以上サンプルをオーバーフィックスしないように注意することが重要です。しかし、サンプルが摂氏4度に固定されている場合、この固定期間は長引く可能性があります。この手順に従って、ImageJを用いた定量のような他の方法は、細胞構造、例えば、血液血管密度、様々な生理学的条件に応じて変化する方法のような追加の質問に答えるために行うことができる。
