Vi har beskrevet en metode der gør brug af kvantitative immunoblotting (IB) for at påvise nytten af immunofluorescens (IF) for at vurdere den relative forekomst af en target proteinet i FFPE vævsprøver. Er nødvendigheden at kvantificere proteiner i formalin-fast, paraffin-embedded (FFPE) biopsi vævsprøver findes i mange kliniske områder. For eksempel, bruges kvantificering af biomarkør proteiner i rutinemæssig biopsi prøver til at belyse prognose og informere behandling for kræft patienter1. Nuværende metoder er dog typisk subjektiv og mangler validering.
Immunhistokemi (IHC) anvendes rutinemæssigt i patologi laboratorier og generelt afhænger af en primær antistof rettet mod målet protein og en sekundær antistof konjugeret med en enzymatisk etiket som peberrodsperoxidase2. Konventionelle IHC er følsomme, kan gøre brug af minut prøver og bevarer vævsprøver dermed tillader vurdering af protein udtryk inden for dens relevante histologiske forbindelse morfologiske integritet. Men fordi kromogent signalet genereret af IHC er subtraktiv, det lider af en relativt snævre dynamikområde og giver et begrænset potentiale for multiplexing2,3,4. Matrix assisted laser desorption/Ionisation massespektrometri imaging (MALDI-MSI) bevarer morfologiske integritet. Men denne udvikling teknologi er forbundet med beskedne morfologiske opløsning og kræver betydelige kalibrering og normalisering, forringer dens gennemførlighed for rutinemæssig klinisk brug5,6,7. Alternative teknikker til at kvantificere protein i vævsprøver omfatter immunoblotting8, massespektrometri9,10,11og enzym-forbundet immunosorbent assay (ELISA)12, hver af som begynder med en homogeniseret lysate af vævsprøve. Primære vævsprøver er heterogene, idet de indeholder en lang række celletyper. Derfor, teknikker, der medfører homogenisering prøverne ikke tillader kvantificering af et protein i en bestemt celle population af interesse, såsom kræftceller.
Ligesom IHC, hvis gælder for lille FFPE prøver og tillader lagring af histologiske integritet13. Men takket være den additive natur af fluorescens signaler, hvis er åbne over for anvendelsen af flere primære antistoffer og fluorescerende etiketter. Heri beskriver vi anvendelse af kvantitative immunoblotting ved hjælp af etablerede cellelinier som en guld standard for at undersøge kvantitativ art, hvis kombineret med computerstøttet billedanalyse til at bestemme den relative forekomst af et protein af interesse i histologiske snit fra FFPE vævsprøver.
