ここでは、抗菌活性を阻害する台湾のグリーンプロポリスを抽出し、特徴付ける溶媒としてエタノールを用いるプロトコルを紹介する。このプロトコルは、台湾のグリーンプロポリスの品質を決定するための信頼性と再現性の高い方法です。デモンストレーションの手順は、私の研究室の博士課程の学生であるチュンティン・チェンとYi'Uan Chienです。まず、台湾の緑のプロポリスの100グラムを計量します。スパイスグラインダーを使用して、プロポリスを粉砕し、大きな粒子を含まない細かい粉末に粉砕します。5つのフラスコをセットし、それぞれに水にエタノールの様々な濃度の100ミリリットルを追加します。各フラスコのエタノール溶液に10グラムのプロポリスを混ぜ、250RPMで48時間振ると摂氏25度でインキュベートします。この後、25マイクロメートルの細孔サイズのフィルターペーパーを使用してエタノール抽出物を濾過します。容積フラスコを使用して、95%エタノールで100ミリリットルの元の体積に濾液を再構成する。使用する準備ができるまで、再構成された抽出物をマイナス20°Cで保管してください。HPLC用の抽出物を準備する準備ができたら、真空蒸発を使用して、各抽出物を10ミリリットル摂氏40度で10分間濃縮します。乾燥した物質を摂氏45度で24時間焼きます。次いで、95%エタノールの10ミリリットルで乾燥物質の各サンプルを再構成する。0.22マイクロメータの細孔サイズのシリンジフィルターを使用して、各抽出物を再フィルタし、HPLCで直接分析するために濾液を収集します。標準曲線の確立を開始するには、88.8~11.2の比で水溶液に対する移動相メタノールの1リットルを調製します。移動相溶液を溶媒として使用し、テキストプロトコルで概説されているようにプロポリン標準濃度の逐次希釈液を調製する。各標準濃度の20マイクロリットルを、最も低い濃度から最高濃度まで順次逆相カラムに注入します。次に、HPLCカラムを摂氏30度に設定し、UV検出器の免除長を280ナノメートルに、流量を1分あたり1ミリリットルに設定し、レコーダー時間を20分に設定する。少なくとも3回は、規格を分析してください。この後、計算シートソフトウェアを使用して、測定応答を濃度に対して差し込み、その結果、決定された方程式とR-2乗値を持つ標準曲線を得る。抽出物の分析を開始するには、各抽出物の20マイクロリットルを逆相カラムに注入します。HPLCカラムを摂氏30度の温度に設定し、1分あたり1ミリリットルの流量を設定します。UV検出器の免除長を280ナノメートルに、レコーダー時間を20分に設定します。抽出を少なくとも3回分析する。次に、標準曲線を用いて、各エタノール抽出物中のプロポリンの濃度を決定する。試験生物とエタノール抽出物を調製した後、希釈エタノール抽出物を10マイクロリットル加え、1ミリリットル当たり0.156~640マイクログラムの範囲で96ウェルプレートのウェルに入れる。各ウェルのボリュームを100マイクロリットルに調整し、すべての希釈液に5%DMSOを維持するようにするために、スープを使用してください。次に、細菌培養物の100マイクロリットルを96ウェルプレートの各ウェルに接種する。摂氏37度で48時間培養。590ナノメートルのマイクロプレートリーダーを使用して、濁度に応じた細菌の増殖を分析し、最小阻害濃度を決定します。この後、寒天板上に成長を示した各ウェルから10マイクロリットルの液体培養液を接種する。摂氏37度で24時間インキュベートします。細菌の活性を決定するために、目に見える細菌の増殖を明らかにしなかった最も低い濃度を特定します。細胞増殖を完全に排除した濃度は、最小細菌濃度であると考えられる。本研究では、台湾の緑のプロポリスをエタノールで抽出した。高濃度のエタノールを使用すると乾燥物質の収率が最も高く、水を使用すると最低であると考えられる。これらの結果は、有機溶媒がこの抽出中に最も優れたパフォーマンスを発揮し、その収率がエタノール濃度と正に関連していることを示している。同様に、プロポリンの濃度は抽出時のエタノール濃度と積極的に関連していると思われ、95%および99.5%エタノール抽出物で得られるプロポリンの最高収率が得られる。次に、Sアウレウス及び大腸菌に対するエタノール抽出物の抗菌作用を調査する。Sアウレウスの平均最小阻害濃度は1ミリリットル当たり10〜20マイクログラムの間であると思われ、平均細菌濃度は1ミリリットル当たり20マイクログラムであると思われる。しかし、水抽出物は、Sアウレウスに対して抗菌性に影響を及ぼさなかった。水またはエタノール抽出物を使用した場合、大腸菌に対する抗菌影響は認められていません。重要な実験手順の1つは、粉砕中に台湾のグリーンプロポリスの均一性を取り入れる方法です。
