1. 准备聚阳库存解决方案

</ol>2. dna 折纸纳米结构的纯化
<ol><li>将 dna 折纸样品的 100μl (在 5 mm tris、1 mm edta、5 mm 氯化碳缓冲液 (称为 tb) (包括 18 mm mgcl2) 中混合, 等效体积为 22 mm mgcl 2, 补充 tb (100μl) 在 1.5 ml 管中.</li>

</ol>3. 琼脂糖凝胶电泳 (年龄)
<ol><li>加入1.6 克琼脂糖和 80 ml 0.5 tae 缓冲液 (20 mm tris, 0.5 mm edta, 10 mm 乙酸, ph 8), 制成玻璃烧杯, 制备2% 琼脂糖凝胶。微波炉 5分钟, 在水浴中旋转烧杯的内容物1分钟。加入352μl 的 mgcl2 (2.5 m), 用 11 mm mgcl2制备凝胶。用10μl 的 dna 凝胶染色对凝胶进行预染色。</li>

</ol>4. 负污渍透射电子显微镜 (nstem)
<ol><li>在发光排放的碳涂层 cu400 tem 栅格上应用5μl 的稀释纳米结构或聚面。让它吸收大约1分钟。</li>
	<li>用一张滤纸从网格边缘排出多余的液体。</li>
	<li>使用5μl 的2% 乌兰醋酸水溶液, 等待1分钟。</li>

</ol>5. 聚丙烯形成
注: 在 n/p0.01-8 比处制备由 dna 折纸和壳聚糖组成的多面体的示例。
<ol><li>使用公式-1 (表 1) 计算每聚阳离子的胺数量。</li>
	<li>使用紫外线分光光度计在260纳米处测量纯化 dna 折纸的浓度。使用2μl 的 tb 缓冲液作为空白来校准机器。使用公式-2 计算 "磷酸盐的 nmol"。</li>
	<li>制备15μl 的 dna 折纸结构, 包括 1 nmol 磷酸盐。</li>

	<li>使用公式-5, 以预定义的 ap 比计算聚面体分解所需的糊精硫酸盐体积。a-p 电荷比是多阴离子 (a) 的硫酸盐基团与 dna 折纸磷酸盐基团 (p) 之间的比率。为了有效地分解多层化合物 (例如500-1000), 需要过量的硫酸右旋糖酐。例如, 要分解在第5节 (聚色含有1个磷酸盐 nmol) 中制备的聚 plex, 在 aop 1, 000, 需要3.6μl 的硫酸糊精 40 kda (50 mg/ml) (ap 为 1, 000, 磷酸盐的 nmol 为 1, 硫酸盐糊精的分子量为 40, 000 g/mol, 糊精硫酸盐库存溶液浓度为 50 mgml, 硫酸盐的含量为 220)。在步骤5.4 的多面体中加入3.6μl 的硫酸糊精 (50 mg/ml), 搅拌均匀。</li>

</ol>7. 对镁消耗的稳定性
<ol><li>用超滤柱 (mwco ~ 3 kda) 用4x600μl 的 mg-零缓冲液 (5 mm tris、1 mm edta 和 30 mm nmol) 清洗20μl 的 dna 折纸或相应的聚 plex (包括 2 nmol 磷酸盐)。每次旋转在 14, 000xg 的 rt. 3-5分钟。</li>
	<li>收集剩余的溶液 (80μl), 并在37°c 下孵育一天, 在650转/分的温度。</li>
	<li>如果测试 lpei 聚面, 请添加 2.5μl mgcl2 (500 mm), 最终浓度为 16 mm mgcl2。然后, 加入7μl 的硫精糊精 (50 mg/ml) (相当于 aop 1, 000) 来分解核心 dna 纳米结构 (分离, 见步骤 6)。 注: 如果使用壳聚糖聚体或裸 dna 折纸, 请跳过此步骤。</li>
	<li>按照步骤3中所述 (图 2) 进行 nstem 成像。</li>
</ol>8. 裸体 dna 折纸纳米结构的 dnase i 滴定测定
<ol><li>将3μl 的裸 dna 折纸, 包括 1 nmol 磷酸盐和2μl 的 10x dnase i 缓冲液 (100 mm Tris-HCl, 25 mm mgcl2, 5 mm cacl2, ph 7.6), 1μl mgcl2 (250 mm), 在 0.2 ml pcr 管中的水的12μl。加入2μl 的 dnase i (10倍)。调整 dnase i (10倍) 的库存浓度, 使最终的 dnase i 浓度达到 0.25-2 uml。</li>
	<li>在37°c 下将样品在热环器中培养1-2小时。</li>
	<li>将样品浸入冰中, 通过添加 2μl 500 mm 二硫醇 (dtt) 和 2.5μl egta (33 mm) 来使其失活。</li>
	<li>使用步骤3中所述的年龄分析示例。</li>
</ol>9. 多聚体向 dnase i 方向的稳定性
<ol><li>将聚氨酸 (包括在不同 nsp 比下制备的 1 nmol) 的4μl 与 dnase i 缓冲液 (10x) 的1.5 微米、tb 的8μl 和 dnase i (100 uml) 的1.5 微米与 0.2 ml pcr 管中混合。</li>
	<li>在37°c 的温度下将样品在热环器中培养24小时。</li>
	<li>加入2μl 蛋白酶 k (20mg/ml), 在37°c 时在热环素中消化 dnase i., 以吸收30分钟。 注: 可以通过添加1.5μl 的 dtt (500 mm) 和2μl 的 egta (33 mm) 来实现 dnase i 的化学失活, 而不是酶消化。</li>
	<li>加入3.6μl 的磺酸糊精 (50 mg/ml) (相当于 aop 1, 000) 来分解核心 dna 纳米结构。</li>
	<li>按照步骤3中的说明执行 age (图 3 a,图 3A)。包括新鲜的 dna 折纸作为测量和比较凝胶上的带强度的对照。</li>
	<li>根据供应商提供的协议, 利用挤压冷冻提取柱对年龄源带进行消费, 并提取 dna 折纸。</li>
	<li>按照步骤4中所述 (图 2) 进行 nstem 成像。</li>
</ol>10. 胎儿牛血清 (fbs) 的稳定性
<ol><li>将4μl 的裸 dna 折纸或其相应的聚体 (包括 1 nmol 的磷酸盐) 与17μl 的 tb + 10% fbs 混合在 0.2 ml pcr 管中。使用刚解冻的 fbs。</li>
	<li>将样品在37°c 的热循环中培养24小时。</li>
	<li>将样品浸泡在冰浴中。</li>
	<li>如果测试了聚面, 请加入3.6μl 的硫糊精-40kda (50 mg/ml) 进行分离。此外, 如果使用壳聚糖多路复用, 请添加 naoh (参见步骤 6.3)</li>
	<li>按照步骤3中的说明执行年龄段。包括新鲜的 dna 折纸作为测量和比较凝胶上的带强度的对照。</li>
	<li>利用年龄段上的带, 通过挤压冷冻提取柱提取 dna 折纸。</li>
	<li>在 0.2 ml pcr 管中提取的 dna 折纸溶液 (20μl) 中加入2μl 蛋白酶 k (20mg/ml)。在37°c 时在热环器中培养 30分钟, 以消化与纳米结构结合的血清蛋白质。</li>
	<li>用5x500μl 的 tb 清洗样品, 包括 16 mm mgcl2 , 使用超离心柱 (mwco ~ 100 kda) 将蛋白酶 k (mM ~ 28.9 kda) 洗掉。每次旋转在 14, 000xg 的 rt. 3-5分钟。</li>
	<li>按照步骤4中的说明执行 nstem 成像 (图 3)。 注: 从步骤10.6 中提取的凝胶样品可直接用于 nstem 成像。然而, 由于血清蛋白与 dna 折纸纳米结构的结合, 成像可能具有很大的挑战性。因此, 建议使用蛋白酶 k (步骤 10.7-10.9) 来促进成像过程。</li>
</ol>11. 辣根过氧化物酶 (hrp) 功能化 dna 折纸在多形涂层上的可添加性检测
<ol><li>如步骤2所述, 纯化自组装的生物技术-nr (biotin-nr)。</li>
	<li>用 200μl hrp 共轭链霉素 (540 nm, tb 缓冲液) 培养200μl 纯化的生物素-nM (36 nm, 16 mm 补充结核病缓冲液)。加入4.8μl 的 mgcl2 (500 mm), 最终浓度为 14 mm。在热敏管中以650转/5 分的速度在 r. t. 12小时内孵化</li>
	<li>采用 peg 纯化方法 (如步骤2所述) 纯化 hrp 功能化 nr (hrp-nr), 去除未结合的 hrp 酶。在结核病中对沉淀的 hrp-nr 进行重组, 包括 16 mm mgcl2。</li>
	<li>将纯化的 hrp-nr (36 nm) 与不同浓度的6.5μl 聚离子混合, 以1、2、4、10和20的 nM 比制备多普莱斯 (使用公式 3)。</li>
	<li>对非生物素化 nr 重复步骤 11.2-11.2。由于 hrp 酶可能与 dna 折纸非特异性结合, 非生物素化 dna 折纸 (用 hrp 酶和纯化的 peg) 进行处理 (类似于生物素-nr), 将作为检测的对照。</li>
	<li>将6.5μl 的蒸馏水与不同浓度的不同浓度的多角混合, 与1、2、4、10和20的定义 nsp 比相对应。这些样本作为空白组。</li>
	<li>将步骤 11.4-11.6 (12.5μl) 中的制备溶液添加到96孔板 (包括 72.5μl hepes 缓冲液 (25μl hepes) 中, 其中包括202.5μl 的 hepes (25mm m hepes, 20 mm kcl, 200 mm nacl, 0.05% triton x-100, 1% dmso, ph 7.4), 搅拌良好。</li>

</ol>12. 检测血液结合型凝血动物 (hba) 功能化 dna 折纸在多聚涂层上的可重复性
<ol><li>如步骤2中所述, 纯化 hba 函数化 nr (hba-nr)。在结核病中再利用沉淀的 hba-nr, 辅以 6 mm mgcl 2 (较高的盐浓度导致 hba-nr的聚集)。</li>
	<li>将 bba-nr (40 nm) 10μl 与不同浓度的10μl 多角共混, 制备1、2、10和20的定义 nM 比的多普莱斯。</li>
	<li>在与1、2、10和20的 n/p 比率相对应的不同浓度下, 将 naked-nr (40 nm) 或蒸馏水与10μl 的聚合物混合10μl。</li>
	<li>将步骤12.2 和12.2 中的20μl 制备溶液加入96孔板, 包括60μl 的 hepes 缓冲液 (25 mm hepes、20mm kcl、200 mm nacl、0.05% triton x-100、1% dmso、ph 7.4), 搅拌良好。包括至少一个空白样品, 用水代替20μl 聚面或聚阳溶液 (从步骤12.2 和 12.2)。</li>

gene-therapy inspired polycation coating for protection of dna origami nanostructures

设计了三种不同构型的 dna 折纸纳米结构, 包括纳米体 (nr)、纳米粒子 (nb) 和线框纳米结构 (wn) (纳米结构的三维模型如图 2所示)。nr 和 nb 分别是在正方形晶格和蜂窝晶格的基础上设计的, 分别使用 cadnano17 , wn 是利用代达罗软件18创建的。已经公布了 dna 纳米结构的设计和自组装的综合协议, 已经公布了19、20、21、22.根据 stahl 等人描述的协议, 订购、自组装和进一步纯化了形状特异性主食链。23 (步骤 2)。
采用凝胶阻滞法和 nstem 成像法对多层进行了表征。在将 dna 折纸与多面体混合后, 观察到裸纳米结构带向阴极的转移 (图 1a)。这可归因于磷酸盐基团在与多离子结合时负电荷的平衡和复合物整体尺寸的增加。添加额外数量的多阴离子, 如硫酸糊精, 可以逆转多普莱斯的形成。在这方面, 与低分子量的硫酸右旋糖酯 (mw ~ 4kda) 相比, 分子量较高的糊精硫酸盐 (mw ~ 40kda) 更有效 (图 1b)。
用醋酸铀阴性染色 tem 成像 lpei 多层, 对任何颗粒进行成像都很困难。据推测, lpei 可能会阻碍醋酸铀与磷酸盐主干结合, 由于对 dna 折纸的严密屏蔽。因此, 在 nstem 成像之前, lepi 多层中加入了过量的硫酸糊精。未解的 dna 折纸纳米结构无缺陷或分解的迹象。相反, 壳聚糖多层被成功地成像, 无需聚阴离子处理 (图 2)。
所有的裸 dna 纳米结构 (无论其不同的配置) 在37°c 孵育后完全变性在 Mg-zero 个缓冲区, 证实了 nstem 成像。相反, 在 N/P≥1涂覆 lpei 或壳聚糖的纳米结构保持完好。同样, dnase i 滴定检测显示无保护的纳米结构对酶消化的敏感性。虽然裸纳米结构在2小时后在 1 u/ml dnase i 存在的情况下被完全消化, 但 lepi 或壳聚糖封装的 dna 折纸在 mg-零缓冲液中保持完整, 并以 10 umml dnase i 为补充, 至少一天 (图 2)。通过提高多层的 nsp 比, 实现了对溶核消化的更高稳定性。此外, 与壳聚糖相比, lpei 更有效地保护 dna 纳米结构 (图 3a,图 3c), 这可能是由于 lepi 的电荷密度较高, 因此与 dna 24 的结合亲和力更高.使用不同分子量的 lpei 时无差异(图3b)。
