この方法は、低強度超音波刺激によって生成された機械的な力に生物学的サンプルがどのように反応するかなど、メカノバイオロジーの重要な質問に答えるのに役立ちます。この技術の主な利点は、実験者が非侵襲的に機械的な力をサンプルに適用し、リアルタイムで様々な生物学的境界を測定できることです。まず、垂直プレスドリルを使用して、標準的な35ミリメートル培養皿の底部に12ミリメートルの穴をゆっくりとドリルします。掘削したら、ブレードを使用して皿の底に取り付けられたプラスチック片を取り除き、外面に滑らかな表面を作り出します。次に、皿の外底面に海洋グレードのエポキシまたは接着剤の薄い層を塗布する。接着剤の上に、ポリエステルの2ポイント5ミクロンの厚いフィルムを置き、しっかりと押して、接着剤がフィルムと厚いプラスチック表面の間に均等に広がることを確認します。フィルムを指で遠心的に静かに引っ張り、平らな表面を作ります。接着剤が乾燥したら、ポリエステル底皿を95%エタノールで簡単に洗い換えます。次いで、UVは強い254ナノメートルのUV励起源の下に置くことによって皿を殺菌する。持続時間と強度を調整して、約330ミリジュール/平方センチメートルのUV線量を提供します。次に、市販の細胞外マトリックスタンパク質混合物を1~100の割合で所望の培養培地で希釈する。氷上でマトリックス重合を防止し、100マイクロリットルの培地混合物をポリエステルフィルムに素早く塗布します。無菌性を維持するために、蓋を皿に戻します。一度覆われた後、マトリックスコーティングされたポリエステル底皿を細胞培養物にインキュベートし、炭酸ガスを緩衝したインキュベーターを摂氏37度で6〜12時間培養します。インキュベーション後、過剰培地を吸引し、所望の密度で細胞を直接表面に播種する。直立した顕微鏡の目的の下に水タンクを置きます。次に、市販のオプトメカニカルコンポーネントを用いて、試料ホルダーを目的とトランスデューサの間に配置する。移動部品と変換段階のアクチュエータが水の損傷を避けるためにタンクの外側または水線の上にされていることを確認してください。次いで、浸漬トランスデューサを利用する前に、脱イオン化し、水を脱気した水をサンプルホルダの水平面まで満たします。非腐食性の、市販の光機械部品を使用して、トランスデューサが光路に対して斜めの位置にあることを確認する。これにより、反射された波がサンプルから遠ざかることを保証します。関数発生器の周波数をトランスデューサの公称ピーク周波数に調整します。次に、関数発生器のバーストモードを用いて、所望の持続時間と繰り返し周波数の正常な電圧パルスを作成する。次に、ピーク電圧を希望の値に調整します。パルス持続時間が 2 つの連続するパルス間の経過時間よりも短いことを確認します。関数ジェネレーターの出力をオシロスコープの入力に接続します。オシロスコープを使用して、波形が目的の信号に対応していることを確認します。次に、関数発生器の出力を適切なサイズのRFアンプの電力の入力に接続します。超音波トランスデューサと互換性のある周波数範囲と音響強度で動作するハイドロフォンプローブを使用して、サンプルの位置で目的の視野内でプローブの先端に焦点を合わせます。プローブとトランスデューサの両方が水浴に浸かっていることを確認し、音響軸をハイドロフォンプローブに向かって視覚的に配置して、トランスデューサの総位置合わせを行います。2 つの間の距離がトランスデューサの焦点距離に対応していることを確認します。ハイドロフォンの先端を水以外の物理的な物体でぶつけないでください。次に、ハイドロフォン出力をオシロスコープの信号入力の1つに接続します。次に、同期トリガを関数生成器から別のオシロスコープ入力に接続します。オシロスコープ上で両方の信号を同時に可視化します。次に、プローブに損傷を与えないように、低いデューティサイクルと低振幅で超音波サイクルを少なくしてトランスデューサを駆動します。ハイドロフォンの先端に損傷を与えないように、ハイドロフォンのメーカーの安全な動作条件を確認してください。トランスデューサの表面からハイドロフォンへの超音波の移動時間に応じてS分割ノブを調整します。同期トリガー後にオシロスコープ上のハイドロフォン信号を探します。次に、モータライズされたまたは手動のXYZステージを使用して、トランスデューサをゆっくりと作動させます。最大ハイドロフォン信号と相関する位置にトランスデューサを配置します。ビームを整列させた後、トランスデューサを駆動するさまざまな電圧について、オシロスコープでのハイドロフォン出力のピークからピークへの振幅を測定します。ハイドロフォンの圧力制限を超えないようにしてください。細胞の培養培地を、フルオ-4 AMなどの細胞透過型カルシウム感受性色素の5マイクロモルを含む所望のイメージングバッファーに置き換えます。その後、培養皿を摂氏37度で1時間培養します。インキュベーション後、同じバッファーに色素を含まない細胞を慎重に洗浄します。その後、皿をサンプルホルダーに入れます。490ナノメートルで青色光の照明を使用して細胞を励起します。過度の漂白やピクセルの彩度を避けるために、励起強度とカメラ露出を調整します。長い作業距離を持つ浸漬目的を使用してタイムラプスイメージングを実行し、画像品質を向上させ、望ましくない反射を低減します。ここで、神経膠芽腫細胞は、標準的な培養培地において細胞外マトリックスコーティングポリエステルフィルム上に示されている。これらの細胞はカルシウム感受性蛍光レポーターと共にインキュベートされている。この画像において、赤い点は、個々の蛍光細胞を表し、画像処理ソフトウェアを用いて同定する。超音波で10秒間刺激すると、多くの関心領域で強いカルシウム上昇が視覚化された。対象となるアクティブ領域の数は、時間の経過とともにここに示されます。超音波による10秒の活性化の後、約70%の領域がユーザー定義の閾値を上回ることがわかった。この手順を試みている間、蛍光測定を行う前に超音波ビームを整列させることを忘れないでください。
