この方法は、がん細胞の行動調節における成体幹細胞の役割に関する幹細胞分野の重要な質問に答えるのに役立ちます。この技術の主な利点は、生物を使用せずにインビトロで癌細胞および幹細胞相互作用の評価を可能にすることです。この技術は、骨などの硬い組織に対する癌転移における成体幹細胞の促進役割をモデル化する癌転移の可能性を有する。手順のデモンストレーションは、私の研究室の博士研究員であるフセイン・アプディクによって行われます。まず、17歳から20歳までの若年成人から得られた親知らずの中心からパルプ組織を除去するために、無菌抽出鉗子を使用することから始めます。10センチメートルの組織培養皿の冷たい完全なDMEMにティッシュを置き、2〜3ミリメートルの断片にサンプルをミンチするためにメスを使用する。各皿から6ウェルプレートを処理した組織培養の個々の井戸に切り替え、完全なDMEMの200マイクロリットルで各井戸の組織をカバーします。組織が加湿、摂氏37度および5%CO2細胞培養インキュベーターで少なくとも2時間のインキュベーションでウェルボトムに付着することを許可する。インキュベーションの終わりに、2〜2.5ミリリットルの新鮮な完全な培地で組織を供給し、さらに8〜9日間細胞を培養する。培養物が80%の合流度に達したら、PBSの2ミリリットルで各井戸を洗い、続いて37°Cでトリプシンを2ミリリットルで細胞を剥離する。2分後、ウェルあたり完全なDMEMの2ミリリットルで反応を停止し、遠心分離によって細胞を収集します。次いで、細胞の2つのウェルごとに15ミリリットルの新鮮な完全な培地でペレットを再懸濁し、その後の培養のためにT75フラスコに細胞を通過させる。少なくとも8回の経過の後、光顕微鏡で細胞を可視化する。歯髄幹細胞は培養皿に付着し、紡錘様細胞形態を示すべきである。表面マーカー解析では、トリプシン法が実証された後、1.5ミリリットルチューブの室温で20分間4%パラホルムアルデヒドで細胞を固定し、洗浄ごとに500マイクロリットルのPBSで3回洗浄します。最後の洗浄後、細胞に適切な抗体をチューブあたり100マイクロリットルのPBSに、摂氏4度で1時間ラベルを付けます。インキュベーションの終わりに、示されているようにPBSで細胞を3回洗浄し、1チューブ当たり100マイクロリットルのPBSで適切な二次抗体で細胞に4°Cで30分間ラベルを付けます。次いで、PBSでサンプルを3回洗浄し、標準的なプロトコルに従ってフローサイトメトリーによって細胞を分析します。歯髄幹細胞の分化のために、細胞培養インキュベーター中の24時間インキュベートのために完全なDMEMの500マイクロリットルの24ウェルプレートの個々のウェルに4つの細胞に10回10回播種する。翌日、各ウェルの培地を適切な分化培地に交換し、細胞を培養器に戻し、週2回2週間ずつ培地をリフレッシュする。区別は、標準プロトコルに従ってフォン・コッサおよびアルシアブルー染色によって確認することができる。2〜4回の通路の後、培養された歯科用歯髄幹細胞から、培養された培地上清を採取し、培養物が80%合流に達した。次いで、回収した培地を遠心分離し、あらゆる廃棄物組織材料および細胞デブリを除去し、使用するまで培地をマイナス20°Cで保存する。癌細胞治療のために、摂氏37度と5%CO2で一晩インキュベーションのために12ウェルプレートの個々の井戸に5つの腫瘍細胞に1回10〜1回播種する。翌朝、滅菌200マイクロリットルの先端を使用して各ウェルに傷を負わせ、すぐに各ウェルの上清を様々な濃度のコンディション培地を含む新鮮な培地に置き換えます。そして、直ちに反転した顕微鏡下で傷を観察し、一定の時間間隔で傷ついた培養物の画像を得る。解析の最後に、ImageJ で時間の経過に合ったスクラッチクロージャを測定します。歯髄幹細胞の移動を評価するために、第1シードは、DMEMの250マイクロリットルで0.4ミクロンの細孔を有する個々の24ウェルプレートインサートに4つの歯髄幹細胞に10回10回、37°Cで一晩インキュベートする。次に、摂氏37度で一晩インキュベーションのためにDMEMの500マイクロリットルで24ウェルプレートの個々の井戸に4つの腫瘍細胞を5回10回種付ける。翌朝、デモンストレーションのように腫瘍細胞を傷つける。上清を500マイクロリットルの新鮮なDMEMに置き換え、新鮮な培地で供給される各井戸に1つの歯科パルプ幹細胞シードインサートを入れる。その後、直ちに反転した顕微鏡上の細胞を観察し、一定の間隔で細胞を画像化して、歯髄幹細胞の移動を評価します。歯髄幹細胞と癌細胞の直接相互作用を評価するために、各標識培養液をトリプシン化して単細胞懸濁液を得、遠心分離によって解光細胞を採取する。希釈剤Cバッファーで調製した別個のセルリンカー色素溶液中のペレットを再懸濁し、メーカーの指示に従って供給し、細胞をインキュベートして10分間室温にします。100マイクロリットルの牛胎児血清で染色反応を終了し、遠心分離によって細胞を採取する。次に、完全な成長培地を有する細胞を遠心分離してから、完全な培地の2ミリリットルで1対1の比率で6ウェルプレートで4つの歯髄幹細胞および腫瘍細胞当たり5回10回めっきする。24時間または48時間のインキュベーション後にPBSで洗浄を行い、続いて細胞を回収する。次に、5ミリリットルの丸底流サイトメトリーチューブおよび渦中の蛍光活性化細胞選別バッファーの300マイクロリットルの細胞を再懸濁し、標準的なフローサイトメトリー分析プロトコルに従って細胞を分析する前に細胞凝集体を分散させる。歯髄幹細胞は、造血細胞マーカーではなく、間葉系幹細胞表面抗原をめっきし、発現した後に線維芽細胞様細胞形態を示す。また、適切な分化カクテル用途に続く歯髄幹細胞培養液において、骨、コンドロ、および異分化に関連する形態学的および分子レベルの変化も観察される。10%および20%濃度のコンディション培地を有する傷傷腫瘍細胞培養の治療は、コントロール中処理腫瘍細胞培養物と比較してスクラッチ閉鎖を有意に増加させる。歯髄幹細胞挿入による分泌された分子も、コントロール細胞共同培養と比較して、負傷した腫瘍細胞のスクラッチ閉鎖を増加させた。インビトロ細胞培養における歯髄幹細胞と腫瘍細胞の相互作用の蛍光顕微鏡分析は、腫瘍細胞が48時間後に急速に増殖する組織構造の作成を明らかにする。この手順を試みる間、2つの組織サンプルを冷たい媒体に保管し、すぐにサンプルを実験室に運んで細胞死を最小限に抑えることを忘れないでください。その開発後、この技術は、ヒトにおける成人幹細胞癌相互作用を探求する幹細胞分野の研究者への道を開いた。この手順に従って、さまざまな幹細胞および癌タイプの差動標識細胞に対する免疫細胞トレーシングのような他の方法を実行して、成体幹細胞が癌細胞とどのように相互作用し、癌転移を制御するかについての追加の質問に答えることができる。人間のサンプルを扱うことは危険であり、感染症の患者サンプルを分析し、書面による患者の同意を得るなどの予防措置は、常にこの手順を実行する際に実施されるべきであることを忘れないでください。
