このプロトコルは、腫瘍神経球の3D構造を維持する方法によってグリオーマバイオマーカーの存在を研究するために使用することができるので重要である。このプロトコルの主な利点は、細胞がストレス操作を受け、平坦化されるサイトスピン法と比較して、神経球の形態が維持されることである。この方法は、細胞が3Dで培養され、構造の維持が重要である他の組織培養システムに適用することができる。まず、細胞がコンフルエントになるまでT-25フラスコ中の腫瘍神経球またはNSを培養する。15ミリリットル円錐形チューブで腫瘍NSを選ぶ。NSを300 Gsで5分間インペレットします。この後、ペレットを1ミリリットルのホルマリン1ミリリットルで再懸濁し、室温で10分間チューブをインキュベートする。再中断されたNSに5ミリリットルのHBSSを追加します。先に述べたように細胞をインペレットする。ペレットを含むチューブを氷の上に置きます。洗浄したNSペレットを2%の温かい液体アガロースの500マイクロリットルで再懸濁する。そして、穏やかなピペットまたは攪拌によって混ぜます。アガロースが重合するまで、アガロース埋め込みNSを氷の上に置きます。その後、アガロースの破片を取り除き、NSを保持します。アガロースの一部をカセットに入れ、組織処理を行います。水浴を摂氏42度に設定します。そして、慎重にミクロトームにブレードを挿入します。その後、パラフィンブロックをミクロトームに入れる。片手で、機械の左側にある各セクションの厚さを制御するレバーを押し下げます。レバーを2番目のストップに押し込み、マイクロトームを30マイクロメートルの厚さに設定します。粗いハンドホイールを回してブロックのトリミングを開始します。サンプルの表面が露出しがちになったら、厚さを制御するレバーを5マイクロメートルまたは10マイクロメートルに設定します。サンプルの表面に達したら、トリミングを停止します。氷で氷箱を満たし、パラフィンブロックが直接氷に触れるのを防ぐのに十分な水を入れます。古いミクロトームブレードを捨て、断面化用の新しいブレードを挿入します。パラフィンブロックをガーゼで乾燥させ、ミクロトームの上に置きます。粗いハンドホイールを回してパラフィンの切り離しを開始します。その後、湿ったペイントブラシを使用して、パラフィンリボンを摂氏42度の水浴に移します。鉗子の曲線を使用して、2つのパラフィンセクションの間に鉗子を置き、セクションを穏やかに押し離します。湿ったペイントブラシを使用して、分離されたセクションを正に帯電した接着顕微鏡スライドに押し込みます。まず、60°Cの金属ラックにスライドを20分間インキュベートしてパラフィンを溶かします。次に、テキストプロトコルに従って室温で再水和列車でインキュベーションを介してスライドを脱パラフィン化します。非特異的抗体結合を防止するために、スライドを水道水に保管します。その後、125°Cのクエン酸バッファーでスライドを30秒間、摂氏90度で10秒間インキュベートします。スライドを冷ましてから蒸留水で5回すすます。次に、過酸化水素0.3%で20分間、室温でスライドをインキュベートする。その後、スライドを5分間、緩やかな攪拌で洗浄バッファーで3回洗浄する。希釈された一次抗体で摂氏4度に設定された加湿チャンバーでスライドを一晩インキュベートします。この後、スライドを5分間、穏やかな攪拌で3回洗います。次に、スライドを室温で希釈したビオチン化二次抗体で1時間インキュベートする。緩やかな攪拌で洗浄バッファーでスライドを5分間3回洗います。次に、暗闇の中で常温でアビジンビオチン化西洋ワサビペルオキシダーゼ複合体中のスライドを1時間インキュベートする。前述のとおりに洗浄を繰り返します。この後、DAB過酸化水素基材ブランドと共にスライドを室温で2~5分間インキュベートします。スライドを水道水でリンスし、ヘマトキシリン溶液に浸してそれらに対抗します。その後、水が透明になるまで水道水でスライドを十分に洗い流します。テキスト プロトコルに従ってスライドを退避します。最後に、スライドをキシレンベースの取り付け媒体で覆い、室温で平らな表面で乾燥させます。このプロトコルでは、NSは、アガロースおよびパラフィンに固定され、埋め込まれる。そして、それらは特定のグリオーマバイオマーカーのために染色されます。クロマチン構造を媒介するシャペロンタンパク質であるATRXの発現量は低い。Ki67は、細胞増殖のバイオマーカーであり、神経膠腫NSにおいて陽性である。最後に、NSはオリゴデンドロサイト分化を媒介する転写因子であるOLIG2にも陽性である。覚えておくべきことは、氷に移る前に温かいアガロースで細胞を再中断し、神経球の均等な分布を確保することです。
