purification and analytics of a monoclonal antibody from chinese hamster ovary cells using an automated microbioreactor system

从自动微尺度生物反应器中采集的细胞培养液使用快速蛋白液相色谱法(FPLC)进行纯化,如图1所示,纯化蛋白的关键质量属性(CQAs)具有多种特征下游分析方法。这是自动化微生物反应器系统的主要优点;CQA 的差异可以在各种条件下快速评估。由质谱法处理的CHO-生产mAbs的N-甘油数据应如图2所示的色谱图所示。该图描述了两个色谱图之间的比较,显示一个样本中的 mannose 5 峰值 (M5) 要低得多。如果只观察到噪声基线而不是峰值,则可能意味着色谱设置有故障或过程不成功。使用控件可以简化故障排除。首先,从 dextran 阶梯评估 FLR 峰值;这些峰值表示色谱系统工作正常。接下来,将实验获得的峰值与从经过处理的完整 mAb 标准中获得的峰进行比较。如果标准的峰值可见,但没有识别样本峰值,则 mAb 样本的处理不正确。这可能是由于SDS或核亲子在缓冲液中干扰N-甘油标签和纯化。
SEC-MALS 可用于评估另外两个 CQA:抗体的聚合配置文件和分子量。具有代表性的 SEC-MALS 色谱图与图 3所示的色谱图相当。分子质量分布和绝对分子量是使用具有1.37 mL+(mg_cm)-1和0.185 mL/g的分光系数的所需软件确定的。由于峰值调用和在软件中的基线设置是手动执行的,因此结果可能因用户而异。图 3中单体 IgG1 的绝对分子量为 1.504 x 105 Da = 0.38%(蓝色),高阶复合物为 7.799 x 105 Da = 3.0%(红色)。如峰1的红摩尔质量分布所示,骨料的多分布度远远大于单体。图3。样品数量少,聚合作为CQA的重要性,使得该技术成为自动化微生物反应器系统极具价值的补充分析工具。
mCZE的结果为电象图,如图4所示,它显示了单克隆抗体的电荷变异轮廓。该配置文件是被调查的蛋白质的唯一特征,对操作 pH 高度敏感。充电变型轮廓左侧的游离染料峰值也可见。在建立工作pH值时,操作员有一些自由裁量权来平衡分辨率和信号;此外,操作员必须确保与以 ±30 s 迁移的自由染料峰值保持良好的分离。样品在贴标签后可以脱盐以去除此峰值,但这导致信号显著损失。一旦建立了工作pH值,样品配置文件就可以进行比较。虽然通常一致,但标签效率的变化或赋形剂的差异可能导致样品迁移和电荷变型轮廓的微小差异,使得电图难以直接比较。相反,比较方法通常基于基本、主要和酸性物种的百分比。在这种情况下,可以使用 mCZE 识别小至 1-2% 的相对差异。
可以监测氨基酸的消耗,以确定消耗是否导致CQA的变化。质谱仪的色谱图读出法可用于评估在粗生物反应器介质样品中氨基酸绝对定量的成功创建校准曲线。图 5描绘了两个总电子色谱图 (TIC) 和一个提取的电离度图 (XIC) 作为这一过程的代表性结果。在图5A中,图所示的TIC描述了来自缓冲系统的背景信号,因为只注入了水空白。图 5B描绘了氨基酸标准的代表性 TIC,与水空白相比,可以观察到对应于单个氨基酸物种的小峰(如 7.96 分钟的莱氨酸)。为了集成峰值并便于峰值区域(以及浓度)的量化,XIC 用于仅显示来自定义的"色谱图质量窗口"的信号。根据仪器的灵敏度和色谱分离的质量,最佳质量窗口必须由用户确定。在此示例中(图5C),以液田的 XIC (m/z = 147.1144) 的质量窗口为 10 ppm,其中氨基酸标准中的碱在 8.03 分钟时从柱中洗脱。

该协议涉及对从微生物反应器获得的有限样品进行分析，以提取有关产品关键质量属性的重要信息。这些技术的另一个优点是，它们使用最少的分析时间来确定决定制造产品质量参数的关键属性。演示程序将是大卫·鲍尔斯，塔利亚·费森和菲利普·安加特，我的实验室研究员。首先打开连接到纯化系统的软件，放置 15 毫升锥形管，收集纯化抗体洗液和 50 毫升锥形管，以收集高盐洗涤过程中流经到分数收集器。接下来，将0.22微米孔过滤收获的细胞培养液添加到一个带盖状喷嘴的空12毫升注射器中。取下盖后，将注射器喷嘴插入净化系统的手动喷射口。扭动注射器将其拧紧到位并压紧柱塞，直到注射样品的整个体积，并在附加的 10 毫升大体积样品循环中可见。选择保存的方法，并在仪器软件提示时单击"运行"。当所有抗体都未被清除时，立即用一个摩尔三酯碱基中和纯化蛋白质，使pH值约为5.5。要通过离心浓缩纯化抗体，请将100千达顿过滤器放入滤液收集管中。用 500 微升双蒸馏水清洗过滤器，然后离心。重复过滤器清洗两次。第二次洗涤后，丢弃滤液，将冲洗过的过滤器转移到新的离心管中。接下来，在每个过滤器中添加 500 微升样品进行离心。在旋转结束时，将过滤器反转到新的收集管中，以获得具有最终离心的浓缩样品。对于N-甘油标签和分离，稀释每个浓缩抗体样品的7.5微升。该试剂盒中的15.3微升液相色谱质谱仪或LCMS级水，在90摄氏度下用6微升的5%溶液将抗体变性，在90摄氏度下使用5%的酶友好和质谱友好表面活性剂。在变性结束时，让样品冷却到室温三分钟，然后加入1.2微升的肽N-糖酶F，在50摄氏度下孵育5分钟。将样品冷却三分钟后至室温，在室温下用溶解在无水二甲基甲酰胺中的12微升荧光标记试剂标记样品5分钟。在孵育结束时，用358微升乙酮酯稀释标有N-甘油混合物，并在真空歧管中放置一个亲水性相互作用色谱板，并配有石质和废盘。用200微升水调节井，真空度调整为10至15千帕，以确保液体需要15至30秒才能通过亲水性相互作用色谱树脂。在将每口标有甘油混合物的400微升到每口井上之前，用200微升85%乙酰酸酯平衡油井，在加入每个新液体后应用真空。添加所有样品后，用 600 微升（每次洗涤 1% 甲酸和 90% 乙酰三酯）清洗树脂两次，用 600 微升收集管更换废纸盒。然后用三个 30 微升的光谱洗脱缓冲液来躲避标有 N-甘油的成分，然后用 310 微升二甲基甲酰胺在醋酸酯样品洗脱液中稀释池稀释。要分析超性能液相色谱系统上标有 N-甘油洗脱样品，并结合荧光探测器和飞行质谱仪的四倍时间，在移动相中使用 50 毫摩尔铵和 100%LCMS 级乙酮。将初始流速设置为每分钟 0.4 毫升，LC 梯度在洗脱阶段提供增加的铵。设置荧光探测器以测量 265 纳米的激发和 425 纳米的发射，采样速率为 2 赫兹。将飞行的四倍时间设置为 MS1 正离子灵敏度模式，质量范围为 100 到 2，000 道尔顿，扫描时间为 0.25 秒，并采集连续数据。然后将样本加载到自动采样器中设置为 10 摄氏度，然后运行加载方法。要脱盐样品以准备电荷变型分析，请扣掉 0.5 毫升脱盐柱的底部塞子，松开顶部止损柱，然后将脱盐柱放在 1.7 毫升离心管中。将新柱转移到新的微离心管中，并在柱顶部加入80微升，每毫升3.5毫克抗体溶液。将柱与原始方向对齐并离心机列。然后丢弃脱盐柱，彻底混合浓缩样品。在96井板的一个井中，将样品稀释到每毫升25微升的超纯水中2毫克，并在井中加入5微升标签缓冲液和5微升标签试剂。在室温下孵育10分钟后，将60微升的试剂级水与样品混合，用板密封盖住板，进行离心。要准备电荷变型芯片，请取出存储溶液，用水清洗井一、三、四、七、八和十。用 pH 7.2 运行缓冲液更换水，并将 750 微升运行缓冲液添加到缓冲管中。按仪器用户界面上的卸载板，然后从 96 井板上拆下密封件。将板和缓冲管插入 GX2 样品托盘上的指示点，然后按负载板。将芯片放入芯片室，关闭芯片室的盖子，在提示时选择HT蛋白电荷变种测定，然后单击"运行"。通过快速蛋白质液相色谱从自动微尺度生物反应器中采集的细胞培养样品进行纯化，通过各种下游分析方法对纯化蛋白质的关键质量属性进行表征，如所示。通过质谱法处理的中国仓鼠卵巢产生的单克隆抗体的N-甘油数据应类似于这些代表性的色谱图。大小排除色谱多角度光散射可用于评估抗体的聚集特征和分子量。样品数量少和聚合的重要性是使该技术成为自动化微生物反应器系统极具价值的补充分析工具的关键质量属性。微毛细管区电泳的结果是电泳图，可用于显示单克隆抗体的电荷变异型图，这是所调查的对操作pH值高度敏感的蛋白质的独特特征。也可以监测氨基酸的消耗，以确定耗竭是否导致抗体的关键质量属性发生变化。确保制造产品正确、高效地纯化非常重要，因为分析步骤只能使用正确纯化的蛋白质执行。该产品还可以进行肽映射和稳定性测试。但是，一旦蛋白质被用于一种表征方法，它通常不能用于另一种。这些技术允许分析从小批量、高通量的生物加工筛选平台生产的产品，以了解生物加工参数对产品质量的影响。其中一些技术使用浓缩高氯酸、甲酸、N-二甲基甲酰胺，所有这些都是危险的，在佩戴适当的防护设备时应小心处理。
