1.参数的生长和维持

</ol>2. 斑马鱼的细菌剂量测定
<ol><li>

</ol>3. 斑马鱼的食源性感染
<ol><li>用帕拉米西亚培养细菌。
	<ol>

		<li>在 15°c 下通过 300 x 克离心旋转样品 5分钟, 使用血清学移液器取出约10毫升的 e3 上清液, 并在锥形管中加入约10毫升的新鲜 1x e3。重复此步骤两次。</li>
		<li>在15°c 下以 300 x g离心样品 5分钟, 取出约10毫升的 e3 上清液, 注意不要破坏颗粒。</li>
		<li>将颗粒重新注入剩余的10毫升 e3 介质中, 并将悬浮液的500μl 转移到新的 1.5 ml 微离心管中。通过在 300 x g离心5分钟以计算出寄生虫数的速度, 将500μl 的副虫颗粒状。</li>
		<li>从500μl 样品中取出 e3 上清液400μl。将36.5% 甲醛溶液加入 20μl, 然后轻轻再悬浮, 在22°c 孵育5分钟。 请注意:此步骤杀死参数, 以便进行计数。</li>
		<li>使用移液器和记录测量实际总体积。用0.4% 的色氨酸蓝色溶液稀释副虫悬浮液 1: 1 v。</li>

		<li>通过在 100 mm tris ph 8.0 中加入旋塞因, 将10头斑马鱼转移到6井板的每口井中, 将含有适当浓度的鱼的新鲜 e3 总体积为3毫升 (按步骤计算), 从而对斑马鱼进行麻醉3.3.2). 确保以最少的液体转移幼虫, 以确保它们从接受者的麻醉中恢复。</li>
		<li>在日光条件下, 在30°c 下在白天孵化器中孵育 2小时, 以确保最佳的捕食雷电条件。</li>
		<li>将含有3毫升新鲜 e3 的新井中, 每次含有 100 mg ticaine, 将斑马鱼至少清洗5次。 请注意:在清洗过程中, 不要试图省略毛滴虫。在没有麻醉的情况下转移移动幼虫会增加对动物造成伤害和痛苦的风险。</li>
		<li>(可选) 将斑马鱼嵌入3毫升的 1% 低熔体琼脂在黑墙6井板中进行成像: 低熔融琼脂糖在1xe3 中制成, 并在微波中加热。一旦熔融, 将毛滴原添加到 160 mg/ml 的最终浓度中。将鱼放置在立体显微镜下, 使用剪下的凝胶加载尖端, 确保头部在左侧, 尾巴在右侧 (图 3)。等待 5分钟, 使琼脂糖设置, 然后覆盖嵌入的鱼与 1x e3 含有 160 mgmml 旋塞因成像。</li>
