Protocollen voor inductie van geclusterde stoma hebben in A. thaliana zaailingen wij gepresenteerd door onderdompeling worden behandeld als een sacharose-bevattende middellange oplossing. Zoals u hier ziet, deze methode is zeer eenvoudig en vereist geen speciale vaardigheid maar efficiënt geclusterde stoma kan veroorzaken. Meer dan 45 gewichtspercenten guard cellen zijn geclusterd met 3% sucrose-bevattende middellange oplossing (gemiddelde waarden van meer dan 20 onafhankelijke waarnemingen)6<...

De plant stoma is een essentieel orgaan voor gasuitwisseling voor fotosynthese en transpiratie en stomatal verkeer wordt bereikt door ingrijpende veranderingen in de guard cellen tot ion-gedreven opname- en release van water. Onder een Microscoop, kunnen we zien een patroon van de verdeelde distributie van stoma op de oppervlakken van bladeren en stengels. Deze verdeelde verdeling van stoma wordt beschouwd als om te helpen stomatal beweging, die wordt geregeld door ion en water uitwisseling tussen guard cellen en naburige epidermale cellen1,2. Experimentele inductie systemen voor geclusterde stoma zijn nuttig voor het onderzoek naar het belang van de verdeelde verdeling van stoma.
Er werd gemeld dat de ruimtelijke clustering van stoma kan worden opgewekt door genetische modificatie van essentiële genen voor guard cel differentiatie3,4 of behandeling met een chemisch samengestelde5. We ook gemeld dat onderdompeling behandeling met een middellange oplossing aangevuld met inbegrip van sacharose, glucose, suikers en fructose veroorzaakt stomatal clusters in de zaadlobben van Arabidopsis thaliana zaailingen6. Verminderde callose in nieuwe celwanden scheiden meristemoids en epidermale cellen in de epidermis van sacharose-behandelde zaadlob, suggereren dat sacharose oplossing onderdompeling behandeling negatief beïnvloedt de celwand, waardoor de lekkage werd waargenomen en ectopische actie van belangrijke genproducten voor guard celdifferentiatie (bijvoorbeeld transcriptiefactoren) naar aangrenzende epidermale cellen6. Een soortgelijk mechanisme werd voorgesteld van studies over gsl8/chor mutanten7,8. Onze experimenteel systeem voor reproduceerbare inductie van geclusterde stoma met behulp van sacharose-bevattende middellange oplossing is vrij eenvoudig en goedkoop. Het kan ook worden gebruikt om te onderzoeken van intracellulaire structuren zoals organellen en het cytoskelet in de geclusterde guard cellen wanneer toegepast op transgene lijnen uiten fluorescente markeringen dat etiket intracellulaire structuren9, 10.

Stomatal verkeer bemiddelt de gasuitwisseling plant, die essentieel is voor de fotosynthese en transpiratie. Stomatal te openen en te sluiten zijn bereikt door een aanzienlijke toename en afname guard cel volume, respectievelijk. In dit artikel beschrijven we een eenvoudige induktie systeem voor geclusterde stoma in Arabidopsis thaliana zaailingen door onderdompeling worden behandeld als een sacharose-bevattende middellange oplossing. Onze methode is makkelijk en is rechtstreeks toepasselijk in transgene of mutant lijnen. Grotere chloroplasten worden gepresenteerd als een cel biologische keurmerk van sacharose-geïnduceerde geclusterde guard cellen. Bovendien, wordt een vertegenwoordiger van de confocal microscopische opname van corticale microtubuli weergegeven als een voorbeeld van intracellulaire observatie van geclusterde guard cellen. An Induction System for Clustered Stomata by Sugar Solution Immersion Treatment in Arabidopsis thaliana Seedlings

Deze methode heeft een belangrijke vraag beantwoord van de plantenontwikkelingsbiologie. Dat wil zeggen, wat is het belang van ruimtelijke verdeling van stomata? Ons inductiesysteem voor geclusterde stomata met behulp van sacharoseoplossing is vrij eenvoudig en direct toepasbaar op transgene of gemuteerde lijnen van Arabidopsis thaliana.Voeg om te beginnen 1,1 gram Murashige-Skoog medium zouten en 15 gram sacharose toe aan een beker. Voeg 490 milliliter gedestilleerd water toe aan het bekerglas en meng met de roerstaaf. Gebruik kaliumhydroxide om de pH aan te passen aan 5,8.Verdun vervolgens de oplossing met 500 milliliter gedestilleerd water en breng de verdunde oplossing over op een middelgrote fles. Verwijder met behulp van een schaar de cotyledon uit een 14 dagen oude zaailing.Zweef de cotyledon met de observatie kant naar boven op de daling van Murashige-Skoog oplossing. Zet het exemplaar op het podium van een confocale lasermicroscoop.Gebruik ten slotte heldere veldverlichting om geclusterde beschermcellen te selecteren voor observatie. In dit protocol werd stomatale clustering geïnduceerd in A.thaliana zaailingen. De geclusterde beschermcellen gekweekt in de sacharose met een medium met grotere chloroplasten dan beschermcellen gekweekt in sacharose-vrije omstandigheden.De vergroting van de chloroplasten werd bevestigd met een chloroplast stroma marker en chlorofyl autofluorescenten, wat suggereert dat sacharosebehandeling resulteerde in ophoping van zetmeelkorrels in de chloroplasten. Bovendien, confocale observatie van GFP tub-6 bleek dat de corticale microtubuli waren radiaal georiënteerd in de sacharose behandelde cellen, net als in de sacharose-vrije wachtcellen. Eigenlijk ontdekten we deze methode per ongeluk.Na de bevinding hebben we de milieurespons van geclusterde stomata onderzocht. Wij geloven dat ons systeem kan helpen om de betekenis van de stomata distributie te onderzoeken.
