1. 育种

</ol>5. 幼体后鱼的喂养

raising the mexican tetra astyanax mexicanus for analysis of post-larval phenotypes and whole-mount immunohistochemistry

为了衡量苗圃协议的成功, 我们记录了成功产卵事件中孵化的幼虫数量和存活的幼虫后鱼类的数量。表 3显示了在循环储罐繁殖1个月的数据, 并包括转移到苗圃容器中存活到 14 dpf 的幼虫数量。在这个月, 存活率从41-81 不等, 结果每个种群有65-293 鱼可用于实验或成长为成年人。
为了确定全贴体免疫染色是否成功, 我们将初级抗体培养的样品与仅用二级抗体培养的样本的荧光进行了比较。荧光信号只在使用原代抗体孵育的鱼类中可见。我们已经使用这个协议成功地标记神经元10 (图 1) 和胰腺细胞6在阶段到 12.5 dpf 在表面和洞穴。
