盘状线粒体是研究细胞发育过程、细胞信号转导等重要细胞生物学问题的有趣的模型生物。基因操纵的双翅目细胞技术已经很好地发展。可以使用不同的可选择标记和标记重新循环进行转换, 包括同源重组和插入突变。这是由一个注释良好的基因组支持的。然而, 这些方法针对在液体培养中生长的轴突细胞系进行了优化, 很难应用于只以细菌为食的非无轴野生型细胞。轴系中存在的突变干扰 ras 信号, 导致喂养所需的过度红细胞缺乏, 并损害细胞迁移, 这混淆了这些菌株中信号转导和趋化实验的解释。早期试图对非无源性细胞进行基因操作的努力缺乏效率, 需要复杂的实验程序。我们开发了一个简单的转染协议, 首次克服了这些限制。这些对双细胞分子遗传学的一系列重大改进使野生类型的细胞能够像标准的实验室菌株一样容易纵。除了研究未损坏的信号和运动过程的优势外, 破坏基于农氏核细胞生长的突变体现在可以很容易地被隔离。此外, 整个转染工作流程大大加快, 重组细胞可以在几天而不是几周内生成。另一个优点是分子遗传学可以进一步进行新鲜隔离的野生类型的双星样本从环境。这有助于扩大在这些研究领域使用的方法的范围。

外染色体质粒用于记者研究, 其目的是确定细胞内某些蛋白质的定位或突变细胞细胞结构的变化。对于许多方法, 如监测细胞周期, 同时表达两名记者至关重要。现在可以使用我们的双记者染色体外质粒系统 (表 1)。第1天, 细胞被转染, 然后在5小时后添加可选择的标记 g418 (图 1)。在该示例中, 用质粒 ppi289 转染了 nc4、ddb、ax2 和独立衍生的野生分离株 v12m2 和 ws2162 (补充表 1), 该质粒编码为 gfp-tubulina, gfp-tuclina 是微管和 mcherry-pcna 的标记, 是一种蛋白质用于监测细胞周期 (图 2a)。32小时后, 在显微镜下观察到细胞。大多数细胞表达了两种荧光标记的融合蛋白, 这与此前有报道称, 来自同一质粒的两名记者的表达水平相似, 这在使用两种不同的质粒时几乎是不可能的18,19。图 2b 显示了每个细胞系 (nc4、ddb、ax2、v12m2 和 ws2162) 表示所需双记者的代表性单元格。图2c总结了转染效率。nc4 衍生细胞系显示出最佳的转染效率。然而, 对于 v12m2 和 ws2162 的细胞系, 获得了相当多的转染剂。

将靶向载体集成到指定的基因组位点更具挑战性, 需要对生成的细胞系进行更仔细的分析。在图 3中, 尝试在 nc4 中使用ac5-mcherryki。首先, 质粒必须线性化, 以增加转染后重组事件的频率。为此, 质粒 pdm1514 被切割与ngomiv。在琼脂糖凝胶上运行消化后, 获得两个带。4127 bp 波段包含所需的构造 (图 3)。对于转染, 必须从凝胶中提取被消化的 dna, 并按照制造商的说明使用凝胶提取试剂盒进行纯化。
