Zuverlässige Identifikation von Protein-Lokalisierung ist oft notwendig, bei der Prüfung, molekulare Signalwege in eukaryotischen Zellen. Methoden, subzelluläre Brüche zu erhalten sind übernommen werden Forscher zelluläre Komponenten von Interesse näher zu untersuchen.
Die Mehrzahl der bestehenden Zelle Fraktionierung Methoden fällt in der Regel in zwei große Kategorien, Waschmittel-basierte1,2 und Ultrazentrifugation basierende3,4,5, die sein kann unterscheiden sich durch Schnelligkeit, Präzision und Kosten. Waschmittel je Protokolle verlassen sich auf die Verwendung von Puffern mit zunehmender Waschmittel Stärke um verschiedene Komponenten der Zelle zu solubilisieren. Dies kann ist eine schnelle und bequeme Methode für die Verarbeitung von Proben und kostengünstig, wenn die Anzahl und Größe der Proben klein sind. Waschmittel-basierte Kits können erworben werden, um cytoplasmatischen Membran/Organellen (Gemischter Bruch) und nukleare Fraktionen aus Zellen zu isolieren. Jedoch begrenzen einige Nachteile verbunden mit diesen Kits ihre Nützlichkeit für Forscher. Sie sind entworfen, um einfach ein oder zwei Komponenten der Zelle zu isolieren, aber nicht in der Lage alle Fraktionen aus einer Probe gleichzeitig zu isolieren. Die Verwendung von Reinigungsmitteln bedeutet, dass die Plasmamembran und Membran umhüllten Organellen ebenso solubilisiert werden werden und somit voneinander getrennt werden. Eine zusätzliche Schwierigkeit ergibt sich aus der proprietären Komponenten in diesen Kits, die verhindert, dass Forscher Bedingungen für spezifische Anwendungen zu verändern. Schließlich, sie sind in der Anzahl der Nutzungen eingeschränkt und möglicherweise unerschwinglich für größere Skala Experimente. Non-Reinigungsmittel basierend Kits gibt es für die Isolation von Mitochondrien, aber sie sollen nicht die Plasmamembran zu isolieren und die Probenausbeute ist deutlich geringer als die von Dichte Zentrifugierung isoliert Protokolle6,7 .
Methoden, die Ultrazentrifugation zu nutzen, um Brüche zu erhalten sind mehr zeitaufwendig, aber oft führen zu reiner Brüche als Waschmittel-basierte Kits. Um die Plasmamembranen von Zellen zu isolieren, ohne erste Gefäss-sie (was zu Kontaminationen mit Membran Organellen) verlangt, dass sie durch eine nicht-Reinigungsmittel-Methode, gefolgt von Trennung der zellulären Komponenten über differentielle lysiert werden Zentrifugation – mit der Plasmamembran Isolation erfordern Geschwindigkeiten von 100.000 × g zu erreichen. In vielen Fällen muss Isopycnic Dichte Gradienten Zentrifugation für weitere Trennung von zellulären Brüche oder Entfernung von Verunreinigungen differentielle Zentrifugation folgt. Während diese Methoden gründlich und veränderbar sind, gehören Nachteile, Kosten, Zeitaufwand und die Notwendigkeit einer Ultrazentrifuge zur Trennung von Fraktionen und weitere Reinigung über Dichte Steigung Zentrifugierung. Die meisten High-Speed-Zentrifugen sind Kosten, die für einzelne Ermittler und sind oft geteilt, Ausrüstung an Hochschulen core unerschwinglich ist. Auf diese Weise wird Ultrazentrifugen Verfügbarkeit in diesen Situationen unerschwinglich.
In diesem Protokoll Fraktionierung demonstrieren wir die Isolation der subzellularen Brüche ohne die Verwendung von Reinigungsmitteln Gefäss- und high-Speed Zentrifugation. Diese Methode erlaubt Forschern, die Plasmamembran, Mitochondrien und cytoplasmatische Komponenten einer eukaryotischen Zelle mit minimalen Kontamination zwischen den Fraktionen zu isolieren.

In diesem Protokoll zeigen wir eine Methode, subzelluläre Bruchteile von U937 Zellen ohne den Einsatz von Ultrazentrifugation oder wahllose Reinigungsmittel zu erhalten. Diese Methode nutzt hypotone Puffer, Digitonin, mechanische lyse und differentielle Zentrifugation, Zytoplasma, Mitochondrien und Plasmamembran zu isolieren. Der Prozess kann skaliert werden, um den Bedürfnissen der Forscher, ist preiswert und unkompliziert. Diese Methode ermöglicht abschätzen, Protein-Lokalisierung in Zellen ohne spezielle Zentrifugen und ohne den Einsatz von kommerziellen Kits, die teuer werden können. Wir haben erfolgreich diese Methode cytosolischen trennen, Plasmamembran und mitochondrialen Proteine in die menschliche Monocyte-Zellinie U937 verwendet.

Erfolgreiche Fraktionierung der undifferenzierten U9378 Zellen in Suspension angebaut wurde durchgeführt unter Verwendung des Protokolls genannten und in Abbildung 1dargestellt.
