fully autonomous characterization and data collection from crystals of biological macromolecules

MXPressP のワークフローは完全に自動的にマウント、x 線ビームの中心、特性評価、および人間 GCSH の結晶のシリーズから完全回折データ セットを収集する ESRF ビームライン山地 1 に使用されました。サンプルがマウントされ、ループが (図 1、左) をスキャンする領域の分析します。回折解析後データ コレクション (図 1右) の結晶内 4 点が選ばれました。その後氏の解決策が発見されたため高品質データセット (表 1) を得られた氏パイプラインを含む自動データ解析パイプラインによって処理します。後者では、急速に得られたデータセットと使用される検索モデル分子置換によって段階的に適しているかどうかを評価するユーザーことができます。さらに、そのこのさらなる分析のための最も有望なデータセットにのみ集中するユーザーを許可、配位子の有無が判断できます。氏によるマニュアルの構造決定は、単一自動絞り込みサイクル (図 2 a) 後に高品質電子密度マップをもたらした。このデータセットの自動パイプラインは Å の分解能; 1.32 でデータをカットただし、ユーザー最高解像度シェルで異なる品質統計 (CC1/2、< I/σ(I) > Rmeas) に到着、低解像度でデータを削減することができます。人間の GCSH 構造の結晶構造は牛蛋白質 (3KlR) の16のそれに似ています。
連続的な電子密度、N 末端ヒスチジンの札から離れて、全体のアミノ酸鎖に対して表示されます。人間とウシの GCSH を区別する 4 つの置換の 3 が電子密度 (イル ・ Val66、Asp/Glu98、およびルー/Phe149; で容易に識別可能図 2b-d)。これは側鎖の電子密度は、部分的にしか解決する (図 1e) 柔軟性のため Asp/Lys125 切り替えのため不明瞭です。現在得られるモデル、R の動作と 20.4% 23.8% の R無料値がそれぞれ、自動および手動モデル構築と洗練のさらなるサイクルによってさらに最適化することができます。
