このプロトコルは、正常および疾患を有する血管の膜電位を測定し、膜電位、血管調子、および血流を調節する薬理学的薬剤を評価するために使用することができる。血管内の血管平滑筋はシンチウムにあるため、この技術から生じる膜電位は生理学的範囲に近い。手順を開始する前に、デュアルチャンネル差動電気計アンプを、容器室の近く、所望の場所に置きます。BNC-BNCケーブルを使用して、アンプチャンネルAまたはBの出力をデジタイザのチャンネル入力に接続します。マイクロマニピュレーターにプローブを取り付け、マイクロマニピュレーターを顕微鏡に向け、ミオグラフを振動のないテーブルに向けます。マニュアルに記載されているように、アンプの前面にあるノブとスイッチを実験に適した位置に配置します。次に、適切な電極でアンプの回路グランドにバスグランドを接続し、ケージがアンプのシャーシに接地されていることを確認します。ホウケイ酸ガラスマイクロ電極を調製するには、標準的なプーラーを使用して、直径が1マイクロメートル未満の短い、緩やかな8〜10ミリメートルのテーパーを達成し、より高い抵抗と小さな先端のために2回ループします。マイクロファイバーのシリンジを使用してマイクロ電極を塩化3モルカリウムで満たし、注入中にプランジャーをゆっくりと引き込み、液体が満杯になるスペースを可能にし、マイクロ電極内に気泡が形成されるのを防ぎます。完全に装填されたマイクロ電極をマイクロ電極ホルダーに置き、慎重に、しかししっかりと退屈な穴を通してホルダーに電極シャンクを押し込みます。ラボ組織で余分な液体を取り除き、電極ホルダーアセンブリをアンププローブに接続します。電極試験を実施して、電極抵抗を測定します。次に、記録ソフトウェアを開き、ファイルに名前を割り当て、後で分析するためにファイルを保存します。次に、蒸留水でミオグラフチャンバーを数回リンスしてから、正常な生理塩溶液(PSS)の5ミリリットルでチャンバーをロードします。5または10ミリリットルのシリンジを使用して、気泡を導入することなくガラスカニューラと付属のチューブの両方を濾過された通常のPSSで満たし、鈍い鉗子を使用して2つの10-0モノフィラメントナイロン縫合糸で半結び目を作ります。次に、解剖顕微鏡の下で解剖鉗子を使用して、部分的に閉じた縫合糸結び目を先端からわずかに離れた両方のカニューレに置く。中大脳動脈分離の場合は、スプリングハサミと鉗子を使用して、ラット脳の内径100~200マイクロメートルの中大脳動脈の枝切れのない部分を識別して解剖する。細かい鉗子を使用して中大脳動脈をガラスカニューラに取り付け、縫合糸を締めてカニューレに動脈を固定します。遠位カニューレを閉じて、動脈内に流れがないようにし、流入ピペットをPSSの貯留層に接続します。逆顕微鏡とイメージングソフトウェアに取り付けられた電荷結合デバイスカメラを使用して、カニュール中大脳動脈を視覚化します。MCA の軸方向の長さを、剛性もフラクシッドにも表示されないよう、おおよその長さに設定します。37°Cで95%の酸素と5%の二酸化炭素と浴液を平衡化します。アンプのグラウンドをミオグラフのPSSに浸します。容器チャンバーを照らし、顕微鏡を通して浴液中のマイクロ電極の先端を可視化する。マイクロマニピュレーターを使用して、血管の外壁近くにマイクロ電極の先端を移動し、記録を開始します。微小電極の先端を容器に向かってゆっくりと動かし、容器の中心を目指す。先端が容器に近いところに来たら、筋肉の膜を妨げるために1つの急速な動きで電極を前進させる。膜が浸透すると、膜電位の変化が観察される可能性があります。膜が突き刺さったらマイクロマニピュレーターに触れないでください。膜電位変化が記録されたら、マニピュレータを使用して1回の急速な動きで微小電極を取り除き、記録を停止してからデータファイルを保存します。不浸透は、負の値に対する急速なたわみがあり、膜電位が最低30秒間安定し、電極を取り外すと電圧が突然ゼロミリボルトに戻ると成功したと考えられます。正常な不浸透および膜電位安定化の後、目的の薬物は、浴中に浸透することができ、膜電位の変化を記録することができる。この代表的な実験では、塩化カリウムによる灌流は膜を約6ミリボルト分極し、カルシウム活性化カリウムチャネルのカルシウム依存性活性化剤を灌流しながら、ほぼ4ミリボルトで膜を超分極させた。覚えておくべきことは、直径が1マイクロメートル未満の短い段階的なテーパーを持つ耐性の高いマイクロ電極を引っ張るということです。
