この方法論は、BBN発がん性物質を用いて膀胱腫瘍を再現的に生成し、MRIを介して膀胱腫瘍の負担の大きさと拡張を画像化するために使用することができる。この技術は、高い診断品質で短時間の取得時間を持つオペレータに依存しない迅速なイメージングアプローチを使用して、腫瘍発生の中間段階でマウスのハイスループットアッセイを実施します。膀胱癌腫瘍形成を誘導するには、不透明な飲料水容器に0.05%BBNを加え、6週齢のC57黒6匹のマウスに温水を提供する。次に、週に2回、地元のIACUCガイドラインに従って膀胱腫瘍に関連する苦痛の徴候を監視する。暴露後16~24週、100~200マイクロリットルの無菌生理食塩水を備えた25~27本のゲージ針を装着した1ミリリットルの注射器をBBN処理動物に注入する。10分後、マウスを磁気共鳴画像装置の画像ホルダーに移し、生理学的記録コンピュータに接続された直腸温度プローブを挿入して、動物の体温を監視します。次に、マウスの下腹部に4チャンネルの直交レシーバーコイルを置き、統合されたイメージングソフトウェアで自動調整を開始し、マウス本体全体の3軸画像セットを取得します。この参照画像から、対象領域を特定し、軸、冠状、矢状の平面に沿って 3 組の直交スライスされた画像を取得します。定常進行イメージングシーケンスを使用して、TRを900ミリ秒、TEを2ミリ秒に、FAを70、14平均に設定します。次に、イメージングプラットフォームソフトウェア内で適切な幾何学パラメータを選択し、0.5ミリリットルの厚さの膀胱全体の一連のスライスと0.148ミリメートルの面内解像度を取得します。すべての画像が得られたら、膀胱全体を覆うスライスのセットを適切な医療画像分析ソフトウェアプログラムにエクスポートし、生成された画像をスクロールして膀胱の中間点にスライスを見つけ、膀胱壁と内腔の視覚化を可能にし、定量分析のために膀胱の中心にある代表的な軸索図を選択します。次に、膀胱の外側のエッジと内側の内腔の周囲の境界をトレースして、選択した代表的な軸方向のビュー内の対象領域を慎重に描き、外側の端から内腔を引いて膀胱壁の表面積を計算します。BBN曝露後20週間、70%エタノールで切開部を清掃し、鉗子を使用して腹壁の皮膚を持ち上げます。陰部のシンフィシスからxiphoidプロセスへの中線切開を行い、腹膜を把握して腹膜腔の鋭い切開を可能にする。正中線下腹部の膀胱を特定し、鉗子を使用して膀胱のドームを把握する。精嚢、直腸、脂肪を含む周囲の構造から膀胱を解剖する。膀胱のドームを臍と腹壁に接続する臍帯の中央値を切断します。膀胱の近くに尿管を切り、膀胱の頭蓋を持ち上げ、膀胱を取り除くために尿道を切る。次に、切除された膀胱をPBSですすいで、すぐにバランスで組織を計量します。ここで、膀胱壁3D再構成の安定した進行磁気共鳴画像を有する代表的な真の高速画像化および腫瘍を有するマウスおよび腫瘍を有しない対照マウスの病理画像が示されている。磁気共鳴画像に由来する膀胱壁厚は、毎週、元生体が得た膀胱重量測定と相関する。MRI由来膀胱壁厚および元生体取得膀胱体重データの検査は、腫瘍段階と同様に、非筋肉浸潤性膀胱癌および筋肉浸潤性膀胱癌による病理を階層化する際の関連も示している。完全な膀胱を持っているとき、マウスの画像を取得するのが最善であることを覚えておいてください。膀胱腫瘍のMRイメージング後、マウスは、適宜、同様の腫瘍特性を有するコホートに分割することができる。この技術は、研究者が私たちの患者の結果を改善するために新しい免疫療法やその他の治療法を探求することを可能にします。BBNは発がん性物質であり、この化合物を取り扱う際には常に適切な予防措置を講じるべきであることを覚えておくことが重要です。
