抗原または酵素を示す生きた細菌胞子は、粘膜ワクチンまたは生体触媒として使用することができる機能的に活性なナノ粒子である。原則として、あらゆるタンパク質を胞子に効率的に吸着させることができる。この技術は単純で、遺伝子操作を必要としません。そのため、環境中への組換え生物の放出は関係しない。ヒトや動物の粘膜空隙学に加えて、酵素を表示する胞子は再利用可能なバイオカタライザーとして開発することができます。ロッキングシェーカーで摂氏25度で1時間クエン酸50ミリモルを補った結合バッファーの200マイクロリットルのモデルRFPの2-5または10マイクログラム濃度のモデルRFPの9番目のB.subtilis野生型精製胞子に2回10回標識することから始めます。インキュベーションの終了時に、遠心分離により結合混合物をペレット化し、その後の吸着効率分析のために上清を新しい円錐管に移す。次いで、1洗浄につき200マイクロリットルの結合バッファーで胞子ペレットを2回洗浄し、2回目の洗浄後に100マイクロリットルの新鮮な結合バッファーでペレットを再懸濁する。表面タンパク質抽出のために、表面スポアタンパク質を可溶化するために、2Xナトリウムドデシル硫酸ジチオトライトールの50マイクロリットルに各吸着胞胞再懸濁液の50マイクロリットルを追加します。摂氏65度で45分後、遠心分離によって混合物を採取し、抽出されたタンパク質の各体積の10マイクロリットルを西ブロット上で、mRFPのN末端に存在するHisタグを認識するモノクローナル抗体を用いて標識された表面タンパク質を同定する。蛍光顕微鏡法により吸着した分子を局所化して定量化するには、吸着胞胞体再懸濁液の5マイクロリットルを95マイクロリットルのPBSに加え、得られた各懸濁液の5マイクロリットルを個々の顕微鏡スライドに加える。ポリL-リジン処理カバースリップを各スライドに配置し、1つのスライドを蛍光顕微鏡ステージに置きます。次に、各分野について、位相コントラストおよび蛍光顕微鏡画像を保存します。画像解析では、ImageJ で蛍光顕微鏡画像を開き、[イメージ] と [種類] を選択して、すべての画像が 8 ビット形式であることを確認します。[分析]メニューから[測定を設定]を選択し、[面積]、[統合密度]、[平均グレー値]が選択されていることを確認します。目的の胞子の周りに線を描画するには、描画ツールまたは選択ツールを使用し、[分析] メニューから [測定] を選択します。選択した胞素の値のスタックを含むポップアップ ボックスが表示されます。少なくとも50個の胞子が選択された後、胞子を含まない複数の領域を選択し、測定を繰り返してバックグラウンド蛍光の読み取りを得る。複数のバックグラウンド測定値を取得した後、結果ウィンドウ内のすべてのデータをスプレッドシートにコピーし、選択した胞子の領域とバックグラウンド蛍光値の積分密度の平均を計算して、補正されたセルごとの蛍光の合計を得ます。間接吸着効率評価のため、 精製されたmRFPの6倍の連続希釈を0.5ナノグラム/マイクロリットル濃度で、結合バッファー中の希釈あたり250マイクロリットルの最終体積に、および各予約された上澄みサンプルの100マイクロリットルの2倍の連続希釈の適切な実験数を実行する。次に、0.45マイクロメートルのカットオフでニトロセルロース膜を9~10センチメートルの大きさに切断し、ドットブロット装置のガスケットの端を越えて伸びることなく、5サンプルずつ6ドットの希釈領域をカバーする。プリウェット膜をドットブロット装置に入れる。膜の正しいサイズを確認し、膜とガスケットの間に閉じ込められた気泡を取り除きます。メーカーが説明したようにドットブロット装置を組み立て、それらの井戸を通って空気が移動するのを防ぐために装置の未使用部分をカバーする。装置の準備ができたら、2つの最も外部車線に標準をロードし、サンプルを井戸ごとに適切な希釈のそれぞれ100マイクロリットルで中央車線にロードする。すべてのサンプルがロードされたら、真空を停止し、サンプルが重力によって膜を通してフィルタリングできるようにする前に、2分間真空ポンプをオンにします。10分後、PBSを100マイクロリットルずつよく洗います。さらに5分間真空ポンプを実行します。洗浄バッファーが装置から完全に排出されたら、真空がオンになっている間、ねじを緩め、ドットブロット装置を慎重に開けます。次いで、標準的なウェスタンブロットプロトコルに従って膜を処理する。フィルタの密度分析では、ImageJ を開き、各ドットのアウトラインを開き、示されているように[解析、測定]コマンドを使用して統合密度を測定します。画像の背景補正を行う場合は、空の領域に円を描き、示されているようにその統合密度を測定します。標準ドットの積分密度を、ロードされたタンパク質の量と相関させてキャリブレーションラインを得るとともに、キャリブレーション曲線を使用して各サンプルドットのmRFP濃度を推定します。次に、非結合分数に残っている mRFP の濃度を計算することができます。吸着された胞子の抽出物を装填したレーン内のみに予想されるサイズのタンパク質の存在は、吸着反応の成功を示す。吸着効率の直接評価は、吸着反応の分画後のペレット画分の蛍光顕微鏡および細胞蛍光測定法で使用され、かつ行うことができる異種タンパク質に依存する。その後、示されるようにImageJを用いて、胞子に存在する蛍光シグナルの定量化を行うことができる。吸着効率の間接分析は、非結合タンパク質を含む上澄分率のドットブロッティング分析によって行うことができる、と非結合タンパク質のその後の密度分析は、胞子に吸着したタンパク質の量の間接計算を可能にする。原理的には、あらゆるタンパク質を粘膜ワクチンや生体触媒として吸着することができるが、実際には吸着の効率はタンパク質部位および化学的特性に依存する。
